Summary: The unique characteristics of adult-onset atopic dermatitis (AOAD) compared to pediatric-onset AD persisting into adulthood (POAD) have not been well-studied, hindering the development of targeted therapeutics. This study analyzed skin and blood samples from AOAD, POAD, and healthy controls and found that both AOAD and POAD showed immune and barrier dysregulations with shared Th2/Th22 hyperactivation. POAD had greater inflammation and compromised epidermal barrier compared to AOAD. AOAD also had more dysregulated proteins in serum, including pro-inflammatory and cardiovascular-risk markers.

Summary: The study demonstrated that both 200 mg and 100 mg doses of abrocitinib showed significant efficacy in treating moderate-to-severe atopic dermatitis compared to placebo, with the 200 mg dose being superior to dupilumab in improving itch response. Neither dose of abrocitinib showed significant differences from dupilumab in most other key secondary endpoints at week 16.

Summary: This study aimed to investigate the safety and efficacy of abrocitinib in treating moderate-to-severe AD in adolescents and adults. The meta-analysis of 7 studies found that 100 mg or 200 mg of abrocitinib significantly improved IGA and EASI-75 responses, and it was still superior to placebo in both the adolescent and adult groups. Abrocitinib showed higher efficacy and safety in the treatment of moderate-to-severe AD.

Summary: The objective of this study was to assess whether cardiovascular risk factors mediate the association between physical activity and brain integrity markers in older adults. The results showed that higher physical activity was associated with higher gray matter volume and cerebral glucose metabolism. Lower insulin level and BMI were related to higher gray matter volume. Mediation analyses confirmed that insulin level and BMI mediated the association between physical activity and gray matter volume.
