Analysis of the reptile CD1 genes: evolutionary implications

CD1, as the third family of antigen-presenting molecules, is previously only found in mammals and chickens, which suggests that the chicken and mammalian CD1 shared a common ancestral gene emerging at least 310 million years ago. Here, we describe CD1 genes in the green anole lizard and Crocodylia, demonstrating that CD1 is ubiquitous in mammals, birds, and reptiles. Although the reptilian CD1 protein structures are predicted to be similar to human CD1d and chicken CD1.1, CD1 isotypes are not found to be orthologous between mammals, birds, and reptiles according to phylogenetic analyses, suggesting an independent diversification of CD1 isotypes during the speciation of mammals, birds, and reptiles. In the green anole lizard, although the single CD1 locus and MHC I gene are located on the same chromosome, there is an approximately 10-Mb-long sequence in between, and interestingly, several genes flanking the CD1 locus belong to the MHC paralogous region on human chromosome 19. The CD1 genes in Crocodylia are located in two loci, respectively linked to the MHC region and MHC paralogous region (corresponding to the MHC paralogous region on chromosome 19). These results provide new insights for studying the origin and evolution of CD1.

Summary: RBM20 cardiomyopathy is associated with titin isoform switching and resting Ca2+ leaking. Rbm20 knockout initially promotes larger titin isoform expression and reduces myocardial stiffness in young rats, but the full DCM phenotype takes time to develop. S107 shows therapeutic promise in restoring intracellular Ca2+ levels and cardiomyocyte contractile properties in younger Rbm20 KO rats, but may not maintain cardiac homeostasis in older rats due to multiple pathogenic mechanisms.

RBM20 phosphorylation and its role in nucleocytoplasmic transport and cardiac pathogenesis

Summary: Arginine-serine (RS) domains play critical roles in protein-protein interaction, splicing control, and nucleocytoplasmic transport. By using mass spectrometry, we identified 16 phosphorylation sites on RS domains, including two sites located in the RSRSP stretch. Mutations on these sites affected splicing, nucleocytoplasmic transport, and protein-RNA condensates. Our study also revealed that RBM20 mutations led to cardiac pathogenesis.

Summary: A study found that the RBM20 S639G mutation is associated with dilated cardiomyopathy and early death. By using a knock-in mouse model, researchers showed that this mutation promotes the formation of RBM20 granules in the sarcoplasm and leads to differential expression and splicing of genes related to arrhythmia, cardiomyopathy, and sudden death. Additionally, the knock-in mice exhibited impaired cardiac function.

CryoEM structures of pseudouridine-free ribosome suggest impacts of chemical modifications on ribosome conformations

Summary: This study characterized the structures and conformations of modified and pseudouridine-free ribosomes using electron cryomicroscopy. It found that pseudouridine stabilizes key functional motifs by establishing water-mediated close contacts with the phosphate backbone, while the absence of pseudouridine leads to abnormal inter-subunit movements.

Regulation of translation by ribosomal RNA pseudouridylation

Summary: Pseudouridine is vital to the central dogma as it is enriched in ribosomal, spliceosomal, transfer, and messenger RNA. However, the impact of pseudouridine on the molecular apparatus, such as the ribosome, remains unclear due to a lack of structures without this modification. This study investigated the translation of a hypopseudouridylated ribosome initiated by the internal ribosome entry site (IRES) elements, revealing the loss of pseudouridine-mediated interactions and the link between functional defects and modification loss.
