Determination of related impurities in a new active pharmaceutical ingredient-Sodium 4,4′-(propanediamido)dibenzoate

Summary: The new compound malaben exhibits pronounced activity in treating non-alcoholic fatty liver disease and could potentially be used as a drug. Various impurities in the malaben sample were studied and identified using electrophoretic separation and HPLC-ESI/MS analysis techniques, providing structural elucidation for some of the impurities.

Molecularly Imprinted Polymers for Pharmaceutical Impurities: Design and Synthesis Methods

Summary: The safety of medicinal products is determined by their pharmacological and toxicological profiles, which depend on both the active substance's toxicological properties and the impurities it contains. Many studies have been conducted on the separation and purification of active pharmaceutical ingredients (APIs) and the detection of impurities in APIs and drug products. Molecularly imprinted polymers (MIPs) have been applied in these studies to separate impurities and serve as adsorbents in sample preparation. The surface-imprinting polymerization method has shown advantages over the bulk polymerization method in terms of higher adsorption capacity and faster adsorption kinetics of MIPs. However, MIPs are currently only used for organic compounds in the analysis of APIs and drug products, and there is a need to develop MIPs for other impurities, including multi-template MIPs for simultaneous separation of multiple impurities.

Quantification of Genotoxic Impurities by HPLC in Active Pharmaceutical Ingredient Lorazepam

Summary: A simple and selective reversed-phase high performance liquid chromatography method was developed and validated for the quantification of 4-chloroaniline, 2-chloro-N-(4-chloro-2-(2-chlorobenzoyl)phenyl)acetamide and 2-iodo-N-(4-chloro-2-(2-chlorobenzoyl)phenyl)acetamide in lorazepam. The method provided excellent sensitivity and a typical target analyte level of 150 ppm. It was specific, linear, accurate and precise, with low limits of detection and quantification for all the analytes. This method was validated according to the ICH guidelines.

Enantioselective Organophotocatalytic Telescoped Synthesis of a Chiral Privileged Active Pharmaceutical Ingredient

Summary: This study investigated the continuous flow, enantioselective, organophotoredox catalytic asymmetric alkylation of aldehydes using a homemade photoreactor. Significant productivity increase was achieved compared to batch reactions, and the synthesis of an active pharmaceutical ingredient (API) in high enantioselectivity was successfully demonstrated.

Constant rate filtration allows for collection of separate aliquots during washing stages to optimize the washing strategy.

Analytical approach of elemental impurities in pharmaceutical products: A worldwide review

Summary: Elemental impurities can be present in pharmaceutical products, and they can be either toxic or essential elements. Metal contamination, especially with lead, cadmium, mercury, and arsenic, is commonly observed in natural products, traditional medicines, and herbal medicines. Inorganic contamination in pharmaceuticals can be caused by factors such as soil deposition, water contamination, and manufacturing processes. Analytical methods, especially spectroscopy techniques, have been developed and improved to evaluate and monitor metal contamination in pharmaceutical products. International guidelines, such as the USP and EU, provide comprehensive standards for evaluating impurities in pharmaceutical products.

Enantioselective Synthesis of Ozanimod, the Active Pharmaceutical Ingredient of a New Drug for Multiple Sclerosis

Summary: A short enantioselective synthesis of Ozanimod was reported, with the stereogenic center introduced through imine asymmetric transfer hydrogenation, resulting in high enantioselectivity. Starting from commercially available 4-cyano-indanone, enantiomerically pure Ozanimod was obtained in only 5 steps with a high overall yield of 62% and 99% ee.

Development of a rapid GC-FID method to simultaneously determine triethylamine, diisopropylamine, and 1,1,3,3-tetramethylguanidine residues in an active pharmaceutical ingredient

Summary: A rapid GC-FID method was developed to simultaneously determine residual levels of triethylamine (TEA), 1,1,3,3-tetramethylguanidine (TMG), and diisopropylamine (DIPA) in the synthetic route of an active pharmaceutical ingredient (API). The method involved evaluating GC columns with various stationary phases to achieve optimal peak shape and reproducibility, along with screening various inlet liners to improve sensitivity. The study also identified a new reaction mechanism involving TMG, leading to a more accurate final detection method.

Determination of major radionuclidic impurities in K99TcO4 pharmaceutical raw material

Summary: A rapid analytical method for sequential separation and determination of alpha- and beta-emitting radionuclidic impurities in Tc-99 radiopharmaceutical raw material was presented. The method involves consecutive HTiO co-precipitation steps and sequential chromatographic method to effectively separate and purify the radionuclides. Method evaluation using spiked (KTcO4)-Tc-99 solutions demonstrated its capability to meet the analytical requirements for radionuclidic impurity evaluation of (KTcO4)-Tc-99 raw material.

Mathematical Modeling and Optimization to Inform Impurity Control in an Industrial Active Pharmaceutical Ingredient Manufacturing Process

Summary: This study developed a dynamic model for a stage in the pharmaceutical manufacturing process and validated it using industrial experimental data. The model was used to evaluate the impact of operating parameters on key performance indicators, confirm the adherence to quality attribute limits, and identify process setpoints for potential improvement. Global sensitivity analysis revealed that only reactor-related parameters were significant, while nonlinear optimization showed that impurity limits were respected throughout the operating space and suggested a shifted process setpoint for enhanced yield and reduced impurity content.

Summary: This manuscript details the chemical process development and multi-kilogram synthesis of rovafovir etalafenamide, a phosphonamidate prodrug nucleotide reverse transcriptase inhibitor for HIV-1 treatment. Strategies for impurity control and improving reaction steps were explored, resulting in the development of a chromatography-free approach and discovery of new crystalline intermediate forms. The processes were successfully executed on a large scale to produce API for clinical studies.

The study confirmed the structure of the dimer and validated the polymerization sites and stereochemical configuration of the trimer.
