Intercellular communications-redox interactions in radiation toxicity; potential targets for radiation mitigation

Nowadays, using ionizing radiation (IR) is necessary for clinical, agricultural, nuclear energy or industrial applications. Accidental exposure to IR after a radiation terror or disaster poses a threat to human. In contrast to the old dogma of radiation toxicity, several experiments during the last two recent decades have revealed that intercellular signaling and communications play a key role in this procedure. Elevated level of cytokines and other intercellular signals increase oxidative damage and inflammatory responses via reduction/oxidation interactions (redox system). Intercellular signals induce production of free radicals and inflammatory mediators by some intermediate enzymes such as cyclooxygenase-2 (COX-2), nitric oxide synthase (NOS), NADPH oxidase, and also via triggering mitochondrial ROS. Furthermore, these signals facilitate cell to cell contact and increasing cell toxicity via cohort effect. Nitric oxide is a free radical with ability to act as an intercellular signal that induce DNA damage and changes in some signaling pathways in irradiated as well as non-irradiated adjacent cells. Targeting of these mediators by some anti-inflammatory agents or via antioxidants such as mitochondrial ROS scavengers opens a window to mitigate radiation toxicity after an accidental exposure. Experiments which have been done so far suggests that some cytokines such as IL-1 beta, TNF-alpha, TGF-beta, IL-4 and IL-13 are some interesting targets that depend on irradiated organs and may help mitigate radiation toxicity. Moreover, animal experiments in recent years indicated that targeting of toll like receptors (TLRs) may be more useful for radioprotection and mitigation. In this review, we aimed to describe the role of intercellular interactions in oxidative injury, inflammation, cell death and killing effects of IR. Moreover, we described evidence on potential mitigation of radiation injury via targeting of these mediators.

Role of Tumor Microenvironment in Cancer Stem Cells Resistance to Radiotherapy

Summary: Cancer is a complex chronic disorder, and the interaction between the tumor microenvironment and immune cells plays a crucial role in tumor treatment and development. Factors such as cancer-associated fibroblasts, extracellular matrix, and hypoxia-inducible factor can enhance the proliferation capacity of cancer stem cells, leading to resistance to treatments such as radiotherapy. Additionally, radiotherapy can affect the release of cytokines and growth factors in the tumor microenvironment, altering local immune responses.

The impact of various amounts of fabricating components on the response of PASSAG polymer gel dosimeter: An optimization study

Summary: This study evaluates the dosimetric characteristics of PASSAG gel dosimeter for different amounts of components in its formulation. It was found that the optimized PASSAG gel dosimeter showed increased sensitivity and excellent linear dose response, with dose resolution values varying in the 0-10 Gy range. Temporal stability in R-2 values was observed during 15-30 days post-irradiation, with R-2 dose sensitivity varying over the 30-day period.

Cardiac inflammation and fibrosis following chemo/radiation therapy: mechanisms and therapeutic agents

Summary: Cardiovascular disorders are a major concern among individuals undergoing cancer therapy, with heart diseases being a common cause of mortality in these patients. Inflammatory changes and fibrosis are key mechanisms of cardiac diseases following chemotherapy and radiotherapy. Chronic oxidative stress, cell death, and overproduction of pro-inflammatory and pro-fibrosis cytokines are the underlying mechanisms of cardiovascular diseases following cancer therapy.

It has promising anti-tumor effects and may reduce genomic instability and risks of second malignancies in normal tissues, as well as improve the efficacy of anticancer modalities.

Immunovirotherapy: The role of antibody based therapeutics combination with oncolytic viruses

Summary: Despite the approval of new drugs and targeted therapies, most cancer types remain difficult to treat due to tumor heterogeneity, immune system evasion, and the complex interaction between the tumor microenvironment and immune cells. Oncolytic virus (OV) immunotherapy, which uses viruses to selectively replicate within cancer cells and activate the immune system, has shown promising results. However, challenges such as off-target side effects and non-specific uptake limit its effectiveness as monotherapy. Combining OV with other biotherapeutic agents, such as antibodies and CAR-T cells, has been proposed to enhance the anti-tumor activities of OV.

The results showed that simultaneous exposure to Wi-Fi waves and gamma-ray can increase the number of double-strand breaks in DNA in peripheral blood lymphocytes within 72 hours after technetium injection.

Metabolic risk factors of ovarian cancer: a review

Various pathological factors contribute to ovarian cancer risk, development, and survival, making obesity and DM potential intervention targets for preventing ovarian cancer and improving outcomes.

Evaluation of set-up errors and estimation of set-up margin during external beam radiation therapy of prostate cancer using electronic portal imaging device (EPID)

Summary: The study measured systematic and random set-up errors in prostate cancer radiotherapy using EPID and proposed the optimum CTV-to-PTV margin. Results showed population systematic and random errors in different directions, leading to calculated margins based on different formulas, with overall margins averaging at 2.73, 4.98 and 5.86 mm.
