A Clinical Update and Global Economic Burden of Rheumatoid Arthritis

Background and Objective: Rheumatoid arthritis (RA) is a predominant inflammatory autoimmune disorder. The incidence and prevalence of RA is increasing with considerable morbidity and mortality worldwide. The pathophysiology of RA has become clearer due to many significant research outputs during the last two decades. Many inflammatory cytokines involved in RA pathophysiology and the presence of autoantibodies are being used as potential biomarkers via the use of effective diagnostic techniques for the early diagnosis of RA. Currently, several disease-modifying anti-rheumatic drugs are being prescribed targeting RA pathophysiology, which have shown significant contributions in improving the disease outcomes. Discussion: Even though innovations in treatment strategies and monitoring are helping the patients to achieve early and sustained clinical and radiographic remission, the high cost of drugs and limited health care budgets are restricting the easy access of RA treatment. Both direct and indirect high cost of treatment are creating economic burden for the patients and affecting their quality of life. Conclusion: The aim of this review is to describe the updated concept of RA pathophysiology and highlight current diagnostic tools used for the early detection as well as prognosis-targeting several biomarkers of RA. Additionally, we explored the updated treatment options with side effects besides discussing the global economic burden.

Drug repurposing against galectin-3 using simulation-based studies

Summary: This study examines the possibility of cardiovascular medications inhibiting Galectin-3-related biological activities using bioinformatics techniques. Through clustering, molecular docking, and molecular dynamics simulation, potential chemical compounds were screened and repurposed. The results show that certain compounds have the ability to inhibit Galectin-3. Further experimental validation and animal model-based studies are needed.

Recent studies have found that SNPs play a significant role in the development and spread of glioblastoma, with many GB-related SNPs being highly abundant in the promoter region, creating genetic vulnerability to glioma.

This study evaluates the inhibitory potential of lisinopril on MARK4, a protein involved in regulating cardiomyocyte contractility. Molecular docking and molecular dynamics simulations demonstrate the stable binding and therapeutic role of lisinopril in managing cardiovascular diseases.

Summary: Finding a link between hormones and microRNAs (miRNAs) is valuable for adjusting genetic composition or cellular functions without gene-level interventions. The cleavage of precursor miRNAs by dicer is an interface link between miRNA and its regulators, disruption of which can affect neurogenesis. Hormonal regulation of miRNAs occurs directly through binding to miRNA promoter elements and indirectly through regulation of post-transcriptional processing proteins' signaling effects. Estrogenic hormones play a significant role in regulating miRNAs in the brain, and their relationship with miR-29, miR-9 could be an etiological route for various neurodegenerative disorders.

Mitochondrial complex I, composed of 44 subunits encoded by nuclear and mitochondrial genes, is particularly important and frequently mutated, leading to various neurological disorders. This review discusses the genetic origins of neurological diseases related to mitochondrial complex I and highlights recent advances in diagnostic and therapeutic approaches.

Summary: Mitochondria play important roles in neuronal development, physiology, plasticity, and pathology through their regulation of bioenergetic, calcium, redox, and cell survival/death signaling. However, a comprehensive discussion on the relevance of isolated brain mitochondria in neuroscience research has been lacking. This mini-review explores the commonly used analytical assays for assessing mitochondrial physiology and dysfunction in organello, with a focus on neuroscience research. It also provides protocols for isolating mitochondria, assessing their quality, and preserving them for future use.

Dysfunction of mitochondria is associated with neurodegenerative disorders such as Parkinson's disease, Alzheimer's disease, Huntington's disease, and amyotrophic lateral sclerosis. This review focuses on the mitochondrial disturbances in these disorders and discusses the potential use of mitochondrial drugs for their treatment and control, including mitochondrial antioxidants, mitochondrial permeability transition blockade, and mitochondrial gene therapy.

Exploration of acute and chronic anti-inflammatory potential of Quercus leucotrichophora A. Camus extracts in Wistar rats: A mechanistic insight

Summary: This research validated the folkloric use of Quercus leucotrichophora (QL) leaf extracts against inflammation and arthritis, and determined the chemical composition using HPLC. The study found that QL leaf extracts exhibited significant antioxidant, anti-inflammatory, and anti-arthritic potential, possibly due to the presence of quercetin, gallic acid, sinapic acid, and ferulic acid.
