Summary: This study investigated the effects of sulforaphane (SFN), the main ingredient in broccoli, on the proliferation of leukemia stem-like cells in acute myeloid leukemia cells. The results showed that SFN inhibited the proliferation of leukemia stem-like cells and regulated the stem characteristic of leukemia cells. Furthermore, SFN was found to negatively modulate proliferation of these cells by affecting the Sonic Hedgehog pathway.

Summary: This study found that low-dose exposure to phthalates (PAE) can promote the stem cell properties of breast cancer stem cells (BCSCs) through the regulation of ANp63 alpha and the Sonic Hedgehog (SHH) signaling pathway. The importance of this study lies in revealing the mechanism of PAE exposure on BCSCs and providing a theoretical basis for further investigation of the lowest observed effect concentrations of PAE and its relevance.

Summary: This study investigates the mechanism of cigarette smoke-induced and galectin-3-mediated epithelial-mesenchymal transition (EMT) in airway epithelial cells (AECs). The results show that cigarette smoke extract (CSE) exposure affects the survival, morphology, migration, invasion, and clonogenicity of AECs, along with an increase in the expression of EMT markers, galectin-3, and RUNX-2. Inhibition or silencing of RUNX-2 downregulates galectin-3 and modulates EMT marker expression, migration, invasion, and clonogenicity in CSE-exposed AECs.

Summary: Gastric cancer is a deadly malignant tumor often characterized by recurrences or metastasis even after eradication therapy. Cancer stem cells (CSCs) play a crucial role in this process, and the sonic hedgehog (SHH) pathway is important in maintaining CSC characteristics. The role of p63 proteins in regulating CSCs is not well understood. Dietary diallyl trisulfide (DATS) has been shown to have preventive effects against gastric cancer, but its effect on targeting gastric CSCs is poorly understood. This study investigates the role of ?Np63/SHH pathway in gastric CSCs and the inhibitory effect of DATS on gastric CSCs.

Summary: Clear cell renal cell carcinoma (ccRCC) is the deadliest neoplasm of the urinary tract. The expression of Sonic Hedgehog (SHH) was found to be higher in grade 1 tumors and decreased with tumor progression. PTCH expression was lower in grades 1 and 2 compared to the control and grade 4, while SMO expression was higher in grade 4 than in other grades and the control. High PTCH and low SMO expression were associated with better survival rates in ccRCC patients.

Summary: B-Raf kinase inhibitors such as vemurafenib induce stemness-related gene expression and promote self-renewal of thyroid cancer stem cells by activating the sonic hedgehog pathway transcription factor Gli1. Inhibitors of the hedgehog pathway significantly reduce the expression of stemness-related genes in thyroid cancer cells, enhancing the anti-tumor activity of B-Raf kinase inhibitors when used in combination. Targeting both B-Raf and the hedgehog pathway may help overcome drug resistance in thyroid cancer.

Summary: Acute kidney injury (AKI) is a common complication in patients with potentially life-threatening diseases, and vitamin D deficiency is found to be related to AKI induced by ischemia/reperfusion (I/R). This study shows that cholecalciferol can attenuate renal cell injury and dysfunction caused by I/R surgery through reducing ROS production, inhibiting NF-KB activation, and inhibiting GSDMD-mediated pyroptosis.
