Establishment of a Molecular Serotyping Scheme and a Multiplexed Luminex-Based Array for Enterobacter aerogenes

Serotyping based on surface polysaccharide antigens is important for the clinical detection and epidemiological surveillance of pathogens. Polysaccharide gene clusters (PSgcs) are typically responsible for the diversity of bacterial surface polysaccharides. Through whole-genome sequencing and analysis, eight putative PSgc types were identified in 23 Enterobacter aerogenes strains from several geographic areas, allowing us to present the first molecular serotyping system for E. aerogenes. A conventional antigenic scheme was also established and correlated well with the molecular serotyping system that was based on PSgc genetic variation, indicating that PSgc-based molecular typing and immunological serology provide equally valid results. Further, a multiplex Luminex-based array was developed, and a double-blind test was conducted with 97 clinical specimens from Shanghai, China, to validate our array. The results of these analyses indicated that strains containing PSgc4 and PSgc7 comprised the predominant groups. We then examined 86 publicly available E. aerogenes strain genomes and identified an additional seven novel PSgc types, with PSgc10 being the most abundant type. In total, our study identified 15 PSgc types in E. aerogenes, providing the basis for a molecular serotyping scheme. From these results, differing epidemic patterns were identified between strains that were predominant in different regions. Our study highlights the feasibility and reliability of a serotyping system based on PSgc diversity, and for the first time, presents a molecular serotyping system, as well as an antigenic scheme for E. aerogenes, providing the basis for molecular diagnostics and epidemiological surveillance of this important emerging pathogen.

Attachment of Enterohemorrhagic Escherichia coli to Host Cells Reduces O Antigen Chain Length at the Infection Site That Promotes Infection

Summary: Enterohemorrhagic Escherichia coli regulates its cell wall structure during infection, balancing survival in the intestinal lumen and infection of intestinal epithelial cells. The bacteria express long-chain OAg in the intestinal lumen for increased resistance to antimicrobial peptides, but reduce OAg chain length upon sensing epithelial cell attachment, leading to enhanced injection of T3SS effectors into epithelial cells for host cell infection. This regulatory mechanism may be widespread in pathogenic bacteria and closely related to bacterial pathogenesis.

Development of a Molecular Serotyping Scheme for Morganella morganii

Summary: A molecular serotyping scheme for Morganella morganii was developed based on genetic variation, along with a traditional serotyping scheme that was shown to be consistent. Additionally, a microsphere-based suspension array with high specificity was introduced. This work provides an important tool for the detection and epidemiological surveillance of M. morganii, with potential wide applicability.

Escherichia coli O157:H7 senses microbiota-produced riboflavin to increase its virulence in the gut

Summary: This study reveals that EHEC O157:H7 acquired a previously uncharacterized regulatory system rbfSR, which senses riboflavin produced by gut microbiota and activates the expression of virulence genes. This acquisition represents an essential step in the evolution of the highly pathogenic O157:H7. The introduction of rbfSR into other EHEC serotypes also increased their pathogenicity, indicating a potential public health issue.

The phosphate-induced small RNA EsrL promotes E. coli virulence, biofilm formation, and intestinal colonization

Summary: Escherichia coli strains including enterohemorrhagic E. coli (EHEC) and enteropathogenic E. coli (EPEC) can cause severe gastroenteritis. An RNA called EsrL promotes the pathogenicity, adhesion, and biofilm formation of these strains. The expression of EsrL is repressed by PhoB under low-phosphate conditions, but relieved in a phosphate-rich environment. EsrL interacts with and stabilizes important transcripts, leading to increased adhesion and colonization in the host intestine.

Summary: This study systematically investigates the influence of MerR family regulators on intestinal colonization of V. cholerae. The researchers found that MlrA significantly promotes the colonization capacity of V. cholerae in the host intestine by directly enhancing the expression of the important virulence factor tcpA. Furthermore, they revealed that mlrA is activated by anaerobic signals in the small intestine of the host through Fnr during infection. These findings provide insights into a MlrA-mediated virulence regulation pathway in V. cholerae, allowing it to sense environmental signals and activate virulence gene expression.

By rewiring the metabolic network of the strain, the researchers achieved the goal of poly-LacNAc glycoprotein biosynthesis and further improved the degree of polymerization. This approach opens up new avenues for efficient and green manufacturing of various glycoprotein structures with diverse biomedical applications, reducing or eliminating the use or generation of hazardous substances.
