In pig production, inefficient feed digestion causes excessive nutrients such as phosphorus and nitrogen to be released to the environment. To address the issue of environmental emissions, we established transgenic pigs harboring a single-copy quad-cistronic transgene and simultaneously expressing three microbial enzymes, beta-glucanase, xylanase, and phytase in the salivary glands. All the transgenic enzymes were successfully expressed, and the digestion of non-starch polysaccharides (NSPs) and phytate in the feedstuff was enhanced. Fecal nitrogen and phosphorus outputs in the transgenic pigs were reduced by 23.2-45.8%, and growth rate improved by 23.0% (gilts) and 24.4% (boars) compared with that of age-matched wild-type littermates under the same dietary treatment. The transgenic pigs showed an 11.5-14.5% improvement in feed conversion rate compared with the wild-type pigs. These findings indicate that the transgenic pigs are promising resources for improving feed efficiency and reducing environmental impact.

A Cas9-transcription factor fusion protein enhances homology-directed repair efficiency

Guoling Li, Haoqiang Wang, Xianwei Zhang, Zhenfang Wu, Huaqiang Yang

Summary: This study presents a strategy to enhance HDR efficiency in animal cells by spatial and temporal colocalization of the donor and Cas9 through coupling the CRISPR system with the transcription factor THAP11. By appending THAP11-specific DNA binding motifs to the double-stranded donor DNA, the HDR efficiency can be significantly improved. The THAP11-fused CRISPR system achieved a more than twofold increase in HDR-mediated knock-in efficiency, and up to 6-fold increase can be achieved through combinational use with the small-molecule HDR enhancer valnemulin.

Establishment of a pig CRISPR/Cas9 knockout library for functional gene screening in pig cells

Chuanzhao Yu, Haiwen Zhong, Xiaohui Yang, Guoling Li, Zhenfang Wu, Huaqiang Yang

Summary: The study established a pig genome-scale CRISPR/Cas9 knockout library targeting 16,888 genes, which was used to screen pig cells for genes critical for toxin toxicity. The identified candidates were confirmed through gene knockout and toxicity assays, suggesting the library as a promising platform for pig functional genomic analysis.

Non-Coding RNAs Regulate Spontaneous Abortion: A Global Network and System Perspective

Jianyu Gan, Ting Gu, Huaqiang Yang, Zheng Ao, Gengyuan Cai, Linjun Hong, Zhenfang Wu

Summary: This review discusses the function and mechanism of micro RNAs (miRNAs), long non-coding RNAs (lncRNAs), and circular RNAs (circRNAs) in regulating spontaneous abortion (SA). It suggests the existence of a competitive endogenous RNA (ceRNA) regulatory network in the onset and development of SA. Understanding this network is important for the search of potential RNA markers for SA diagnosis and treatment.

A Nectin1 Mutant Mouse Model Is Resistant to Pseudorabies Virus Infection

Xiaohui Yang, Chuanzhao Yu, Qiuyan Zhang, Linjun Hong, Ting Gu, Enqin Zheng, Zheng Xu, Zicong Li, Changxu Song, Gengyuan Cai, Zhenfang Wu, Huaqiang Yang

Summary: This study generated mutant mice with resistance to pseudorabies virus (PRV) by introducing a single amino acid substitution. The mutant mice showed alleviated disease manifestations, decreased death rate, and reduced viral loading in serum and tissue compared to wild-type mice. However, the mutation also caused a defect in eye development.

Summary: Farm animals' salivary glands have the potential to efficiently produce human therapeutic proteins. This study successfully used transgenic pigs to synthesize and purify biologically active human nerve growth factor (hNGF) from their salivary glands. The method employed the replacement of the endogenous gene to avoid contamination during purification. This research is significant for human health and biomedicine.

Pig Coat Color Manipulation by MC1R Gene Editing

Haiwen Zhong, Jian Zhang, Cheng Tan, Junsong Shi, Jie Yang, Gengyuan Cai, Zhenfang Wu, Huaqiang Yang

Summary: In this study, we identified four missense mutations that could affect MC1R protein function for eumelanin synthesis by comparing MC1R gene sequences. Using CRISPR-mediated homologous recombination, we successfully mutated the e allele in Duroc pigs to the dominant E-D1 allele at the MC1R locus, leading to uniform black coat color. The modified pigs carried a heterozygous E-D1/e allele at the MC1R locus and retained the genetic background of Duroc breed. Our work provides a gene editing method for pig coat color manipulation.

Extracellular vesicle-encapsulated miR-21-5p in seminal plasma prevents sperm capacitation via Vinculin inhibition

Yanshe Xie, Zhiqian Xu, Changhua Wu, Chen Zhou, Xianwei Zhang, Ting Gu, Jie Yang, Huaqiang Yang, Enqin Zheng, Zheng Xu, Gengyuan Cai, Zicong Li, Dewu Liu, Zhenfang Wu, Linjun Hong

Summary: The study found that miR-21-5p can be absorbed by sperm through seminal plasma-derived EVs, overexpression of miR-21-5p inhibits sperm capacitation and acrosome reaction, and the expression level of VCL protein is lower in capacitated sperm. These results suggest that miR-21-5p can prevent sperm capacitation through VCL inhibition.
