Cyclosporine A/porous quaternized chitosan microspheres as a novel pulmonary drug delivery system

N-[(2-Hydroxyl)-propyl-3-trimethyl ammonium] chitosan chloride (HTCC), a hydrosoluble chitosan derivative, has been extensively investigated as a class of drug delivery vehicles because of its unique features. However the studies on HTCC for pulmonary delivery systems have been rarely conducted. This study aimed to design porous microspheres (MS) containing cyclosporine A (CsA) using HTCC as the carrier. The physicochemical properties and biocompatibility of the MS were evaluated. The in vivo efficacy of MS was evaluated in an asthmatic rat model after pulmonary administration. The results showed that porous MS suitable for inhalation could be readily produced by spray drying method. Optimized porous MS in this study exhibited to be biocompatible and safe to use in the lung, and they were effective in suppression of inflammation in the asthmatic rat model. Above all, our results suggested that HTCC porous MS are promising drug carriers for pulmonary drug delivery. [GRAPHICS]

Chitosan/PLGA shell nanoparticles as Tylotoin delivery platform for advanced wound healing

Summary: Wound treatment is a common healthcare issue, and Tylotoin, a peptide derived from salamander, has shown skin wound healing properties. However, its degradation and administration limitations hinder its use. In this study, Chitosan-PLGA-Tylotoin nanoparticles were developed to deliver Tylotoin for wound healing. The nanoparticles demonstrated good antibacterial properties, biocompatibility, and sustained release of Tylotoin, leading to improved wound healing in vitro and in vivo.

Summary: This research proposes a TD GLM method based on temporal moment analysis, which improves the recovery of HRF by incorporating TD moment data and auxiliary physiological measurements. The results show that properly covariance-scaled TD moment techniques outperform their CW equivalents in both RMSE and correlation, and the proposed TD GLM outperforms regular TD moment analysis while allowing the incorporation of auxiliary measurements.

A Membrane Curvature Modulated Lipopeptide to Broadly Combat Multidrug- Resistant Bacterial Pneumonia with Low Resistance Risk

Summary: A lipopeptide nanomicelle (LP 20) with the ability to modulate membrane curvature was developed, resulting in accelerated and enhanced antibacterial activity against multidrug-resistant (MDR) Gram-negative pathogens. LP-20 effectively targeted multiple components on the bacterial membrane, causing efficient damage to both inner and outer membrane. In addition, LP-20 showed prolonged lifetime in mice infected with MDR pathogens.

Oroxylin A regulates cGAS DNA hypermethylation induced by methionine metabolism to promote HSC senescence

Summary: Relevant studies have shown that Oroxylin A promotes senescence in hepatic stellate cells (HSC) by activating the cGAS-STING pathway. The activation of this pathway is dependent on the downregulation of DNMT3A, which suppresses cGAS gene DNA methylation. Oroxylin A prevents the production of methionine metabolites, leading to a decrease in SAM levels and consequently a reduction in DNMT activity. In vivo experiments also confirmed the induction of HSC senescence by Oroxylin A, with partially offset by DNMT3A overexpression.

Summary: Developing novel multifunctional wound dressing materials capable of delivering active molecules for resolving bacterial infections and promoting wound healing is highly desirable. In this study, a multifunctional chitosan/alginate hydrogel decorated with beta-cyclodextrin modified polydopamine-bioactive glass nanoparticles was developed. It demonstrated photothermal performance, nitric-oxide release activities, and potential for treating bacterially infected wounds. The results showed that this nanocomposite hydrogel has multiple functions in preventing bacterial infections, accelerating angiogenesis, and wound regeneration, indicating promising application in wound healing.

Preliminary study on the efficacy of rituximab in the treatment of idiopathic membranous nephropathy: A single-centre experience

Supramolecular Nanozyme System Based on Polydopamine and Polyoxometalate for Photothermal-Enhanced Multienzyme Cascade Catalytic Tumor Therapy

Summary: The study successfully developed a supramolecular cascade catalytic nanozyme system for photothermal-enhanced multienzyme cascade catalytic and glutathione (GSH) depletion activities, triggering apoptosis-ferroptosis synergistic tumor therapy. The system was fabricated using β-cyclodextrin-functionalized polydopamine (PDA) as the substrate, entangled with polyoxometalate (POM), and assembled with adamantane-grafted hyaluronic acid and glucose oxidase (GOx) for tumor targeting and GOx loading. The catalytic function of GOx facilitates the conversion of glucose to H2O2 and gluconic acid, generating hydroxyl radical ((OH)-O-& BULL;) through POM-mediated cascade catalysis. Additionally, the PDA-POM-mediated photothermal hyperthermia boosted the cascade catalytic effect and increased ROS production, promising potential for photothermal therapy/nanocatalytic cancer therapy.
