Summary: The lack of targeted therapies for triple-negative breast cancer (TNBC) leads to higher mortality rates and risk of relapse compared to other breast cancer subtypes. In this study, researchers screened an FDA-approved drug library and identified four drugs that were highly cytotoxic towards TNBC cells with CREB3L1 deficiency. Homoharringtonine showed the most significant cytotoxicity and synergistic effect when combined with paclitaxel, suggesting its potential for TNBC treatment.

Summary: The study showed that the Nrf2 inhibitor brusatol enhanced the anti-tumor effect of lapatinib against various cancer cells through synergistic effects. Additionally, the combination treatment effectively reduced Nrf2 levels, induced ROS generation, and inhibited phosphorylation of several signaling pathways in cancer cells. Importantly, brusatol significantly improved the anti-tumor effects of lapatinib in a xenograft model, suggesting a potential rationale for combination therapy in treating HER2-positive cancers.

Summary: This study identified a new mechanism for overcoming multidrug resistance in anticancer therapy. By combining anthracyclines with thiosemicarbazones, synergistic effects were observed in cell types derived from pediatric solid tumors. The synergism was independent of Pgp expression and resulted from impaired DNA repair control leading to cell death. The concomitant application of these drugs achieved the strongest synergistic effect with lower concentrations.

Summary: Certain bacterial colonists induce a specific T cell response, even in the absence of infection. By engineering a skin bacterium to express tumor antigens, researchers were able to elicit tumor-specific T cells that infiltrate lesions and possess cytotoxic activity. This reveals the potential for commensal bacteria to be harnessed therapeutically for targeted immune responses.

Summary: A protocol for a copper-catalyzed intermolecular cross-coupling cascade between 2-(2-bromoaryl)-1H-benzo[d]imidazole analogues and proline or pipecolic acid has been developed. The developed protocol allows efficient synthesis of a variety of N-fused pyrrolo or pyrido[1,2-a]imidazo[1,2-c]quinazoline scaffolds with good functional group compatibility. Proline or pipecolic acid acts as both ligand and reactant in the reaction, and a mechanistically consecutive approach for the Ullmann coupling, decarboxylation, oxidation, and dehydration reaction process was presented.

Summary: The present study aimed to investigate the anti-melanoma potential of Prosopis Juliflora leaves methanol extract (PJLME) and its mechanisms of action. The results demonstrated that PJLME can inhibit the growth, migration, invasion, colony formation, and induce apoptosis in melanoma cancer cells. PJLME also exhibited anti-angiogenic potential and could suppress the Akt and Erk signaling pathways. Additionally, PJLME downregulated the expression of epithelial-mesenchymal transition (EMT) and stem cell markers. In vivo experiments further supported the inhibitory effect of PJLME on melanoma tumor growth.

Summary: Six squared-planar Pd(II) complexes with thiazine and thiazoline ligands were synthesized and characterized. Cytotoxicity assay showed that complexes with phenyl substitutions had the lowest IC50 values and induced apoptosis. The presence of bulky substitutions on the ligands, such as phenyl groups, may influence the cytotoxicity of the synthesized chemotherapeutic agents.

Summary: Taxanes have been widely used in cancer therapy, but their mitotic-independent mechanisms in vivo are still unclear. Vennin et al. reveal a mode of action where taxanes directly stimulate T cells to release cytotoxic extracellular vesicles to eradicate tumor cells. Taxanes-pre-treated T cells may enhance anti-tumor effects while avoiding systemic toxicity.

Summary: This study demonstrated that carotenoid-enriched nanoemulsions (CEN) can activate a cytostatic form of autophagy and decrease the expression of the senescence marker p16(INK4) in SAOS400 cells. When combined with gamma-irradiation, CEN can induce cytotoxic autophagy and enhance cell death. Beta-carotene, a component of CEN, did not show radiosensitizing effect, suggesting that other bioactive molecules present in CEN may contribute to its biological effects.
