Summary: This study reveals a novel G9a-15PGDH signaling axis that is important in the development and progression of cholangiocarcinoma (CCA). 15-PGDH is epigenetically silenced by G9a, and inhibition of G9a can restore 15-PGDH expression and inhibit CCA cell growth.

Summary: Tumor viruses such as HTLV-1, HCV, MCPyV, HR-HPVs, EBV, KSHV, and HBV play a role in approximately 15% of all human cancers, using similar mechanisms to convey cancer hallmarks to infected cells. Perturbed gene expression and epigenetic processes, including DNA methylation and histone modification, are key mechanisms in virus-induced carcinogenesis. Increasing evidence shows that oncoviruses cause epigenetic modifications which are crucial in cancer development.

Summary: In this study, it was found that SNHG1 is significantly upregulated in breast cancer tissues and cells, and its high levels are closely associated with reduced survival rates in patients. Silencing of SNHG1 inhibited the proliferative, migratory, and invasive activity of breast cancer cells and enhanced their sensitivity to cisplatin. The mechanism involves SNHG1 interacting with EZH2 to epigenetically inhibit miR-381 expression, which in turn suppresses tumor progression and overcomes chemoresistance in breast cancer cells.

Summary: This study identified that in cholangiocarcinoma, the expression of SP1-induced LncRNA AGAP2-AS1 is significantly upregulated, playing a crucial role in tumorigenesis. Knockdown of AGAP2-AS1 can inhibit cell proliferation and induce apoptosis, while AGAP2-AS1 also promotes the proliferation of cholangiocarcinoma.

Summary: Tumorigenesis is a complex process involving the acquisition of various physiological traits by cancer cells. Non-coding RNAs (ncRNAs) play a crucial role in regulating multiple cell cycle processes and are considered important in the management of different types of cancer. ncRNAs can function as oncogenes or tumor suppressors and regulate unrestricted expansion, metastasis, and other biological processes in tumor cells. Dynamic attributes such as increased proliferation, migration, invasion, and epithelial-mesenchymal transition are significant determinants of tumor malignancy and therapeutic resistance. Furthermore, the interaction of ncRNAs with immune cells within the tumor microenvironment promotes tumor formation. Exosome-derived ncRNAs contribute to the maintenance of tumor hallmarks and further enhance the complexity of malignant neoplasia.

Summary: A better understanding of the molecular mechanisms underlying resistance to immunotherapy is needed to improve its efficacy. Activation of oncogenic pathways and inactivation of tumor suppressor genes in tumor cells can inhibit immune cell recognition and affect the composition of the tumor microenvironment, leading to impaired anti-tumoral immune response.

Summary: Colorectal cancer (CRC) is a common type of tumor, with half of all patients carrying missense mutations in the TP53 tumor suppressor gene. In addition to genetic mutations, epigenetic changes also play a role in CRC development. This review focuses on epigenetic modifications related to the entire p53 signaling pathway, which can impact the tumor suppressor function of p53 regardless of genetic mutations. The complexity of p53's role in CRC development and patient prognosis suggests the potential for novel diagnostic methods to evaluate genetic and epigenetic changes in the p53 signal pathway for improved diagnostics and management of CRC.

Summary: In this study, the authors demonstrate that the long noncoding RNA LINC01419 enhances the methylation of the ZIC1 promoter, suppresses ZIC1 expression, and activates the PI3K/Akt signaling pathway, thereby promoting the malignant characteristics of hepatocellular carcinoma cells in vitro and facilitating tumor formation and metastasis.

Summary: This study investigated the function and mechanism of long non-coding RNA NKILA in non-Hodgkin's lymphoma. They found that NKILA was methylated in tumor cell lines, leading to the loss of its tumor suppressor effect and sustained activation of NF-kappa B signaling. The results suggest that NKILA is an important tumor suppressor in DLBCL.

Summary: The expression of ZCCHC10 is downregulated in AML patients, and this is negatively correlated with the expression of lncRNA SNHG1. SNHG1 can bind to the ZCCHC10 promoter and recruit DNA methyltransferases DNMT1 and DNMT3B, resulting in hypermethylation of the ZCCHC10 promoter and consequently inhibiting its expression. Downregulation of ZCCHC10 promotes cell proliferation and survival in AML, contributing to disease progression and acquisition of venetoclax resistance. This study identifies the SNHG1/ZCCHC10/p53 signaling axis as a potential therapeutic target in AML.
