Cell Cycle Regulation of Stem Cells by MicroRNAs

MicroRNAs (miRNAs) are a class of small non-coding RNA molecules involved in the regulation of gene expression. They are involved in the fine-tuning of fundamental biological processes such as proliferation, differentiation, survival and apoptosis in many cell types. Emerging evidence suggests that miRNAs regulate critical pathways involved in stem cell function. Several miRNAs have been suggested to target transcripts that directly or indirectly coordinate the cell cycle progression of stem cells. Moreover, previous studies have shown that altered expression levels of miRNAs can contribute to pathological conditions, such as cancer, due to the loss of cell cycle regulation. However, the precise mechanism underlying miRNA-mediated regulation of cell cycle in stem cells is still incompletely understood. In this review, we discuss current knowledge of miRNAs regulatory role in cell cycle progression of stem cells. We describe how specific miRNAs may control cell cycle associated molecules and checkpoints in embryonic, somatic and cancer stem cells. We further outline how these miRNAs could be regulated to influence cell cycle progression in stem cells as a potential clinical application.

Summary: We conducted a study to identify circulating miRNAs in plasma associated with cognition and incident dementia. Four miRNAs were found to be commonly associated with both cognition and dementia, and their target genes were expressed in the brain, suggesting their potential involvement in dementia pathways.

Summary: Rapidly mutating Y-chromosomal short tandem repeats (RM Y-STRs) have been found to be important in differentiating patrilineally related men and have applications in forensic genetics, anthropological genetics, and genetic genealogy. A large-scale pedigree analysis demonstrated the value of using RM Y-STRs for differentiating male relatives, often achieving individual identification.

Summary: Risk stratification is crucial for early identification and prevention of high-risk individuals and diseases. This study explored the potential of nuclear magnetic resonance (NMR) spectroscopy-derived metabolomic profiles in predicting the risk of 24 common diseases. The results showed that metabolomic profiles were associated with incident rates in all investigated diseases except breast cancer, and the combination of age, sex, and metabolomic profiles outperformed established predictors for 10-year outcome prediction in 15 endpoints. In addition, metabolomic profiles provided predictive information for common diseases even when comprehensive clinical variables were considered. Decision curve analysis demonstrated the clinical utility of predictive improvements at various decision thresholds.

Summary: The spectrum of information related to precision medicine in diabetes includes clinical data, genetics, and omics-based biomarkers that can guide personalized decisions on diabetes care. Metabolomics, an emerging molecular approach, helps understand disease etiology and identify novel biomarkers for complex diseases. Metabolomics can investigate targeted or untargeted metabolites extracted from cells, biofluids, or tissues using high-throughput platforms like NMR and MS techniques. It is proposed as a valuable tool in precision diabetes medicine for discovering biomarkers for diagnosis, prognosis, and management of diabetes through personalized phenotyping and individualized drug-response monitoring.

Plasma circulating microRNAs associated with blood-based immune markers: a population-based study

Summary: This study investigates the association between circulating miRNAs and blood cell compositions and immune markers. The results show significant associations between miRNAs and red blood cell, granulocyte, and lymphocyte counts, as well as with neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), and systemic immune-inflammation index. The study also identifies target genes of several miRNAs that are previously linked to these immune markers. Overall, this study provides insight into the role of circulating miRNAs in immune-related diseases.
