Postharvest responses of sweet cherry fruit and stem tissues revealed by metabolomic profiling

Sweet cherry, a non-climacteric and highly perishable fruit, is usually cold-stored during post-harvest period to prevent senescence; therefore, metabolic profiling in response to cold storage in sweet cherry is of economic and scientific interest. In the present work, metabolic analysis was performed in fruit and stem tissues to determine the metabolic dynamics associated with cold storage in response to 1-methylcyclopropene (1-MCP), an ethylene-action inhibitor, and modified atmosphere packaging (MAP). Fruit (cv. Regina) following harvest were treated with 1-MCP and then cold-stored (0 degrees C, relative humidity 95%) for 1 month in the presence or in the absence of MAP and subsequently maintained at 20 degrees C for up to 2 days. Physiological analysis suggested that cold storage stimulated anthocyanin production, respiration rate and stem browning. Cherry stem exposed to 1-MCP displayed senescence symptoms as demonstrated by the higher stem browning and the lower stem traction force while MAP treatment considerably altered these features. The metabolic profile of fruits and stems just following cold storage was distinctly different from those analyzed at harvest. Marked tissue-specific differences were also detected among sweet cherries exposed to individual and to combined 1-MCP and MAP treatments, notably for amino acid biosynthesis. The significance of some of these metabolites as cold storage hallmarks is discussed in the context of the limited knowledge on the 1-MCP and MAP response mechanisms at the level of cherry fruit and stem tissues. Overall, this study provides the first steps toward understanding tissue-specific postharvest behavior in sweet cherry under various conditions.

Summary: The term terroir is widely used to link geographical characteristics with sensory characteristics of agricultural food products. Geographical indications labeling has been developed to ensure the quality of plant-derived food that is linked to a specific terroir and is considered to have superior sensory properties. However, understanding the molecular components and the contribution of microbiome to the terroir signature remains a challenge. In this study, we provide a comprehensive view of the multi-omic landscape related to the Naxos Protected Geographical Indication potatoes, including transcriptome, proteome, epigenome, and metagenome.

In vitro and in vivo glycemic responses and antioxidant potency of acorn and chickpea fortified gluten-free breads

Summary: Fortifying gluten-free breads with a mixture of acorn flour and chickpea flour significantly improves their nutritional quality and antioxidant potential, while reducing glycemic responses. The inclusion of these flours enhances the concentrations of bioactive compounds and increases antioxidant activities in the breads. The addition of acorn flour also reduces glucose release and improves the glycemic index and load of the breads.

Boron stimulates fruit formation and reprograms developmental metabolism in sweet cherry

Summary: This study reveals the physiological, anatomical, metabolic, and transcriptomic impact of boron on early fruit growth and development in sweet cherry. Boron application during pre-flowering stage increased endogenous boron content, promoted fruit set and mesocarp cell enlargement, and led to accumulation of various sugars, alcohols, organic acids, and amino acids at different developmental stages. Transcriptomic analysis identified boron-responsive genes mainly involved in secondary metabolism, amino acid metabolism, calcium-binding, ribosome biogenesis, sugar homeostasis, and photosynthesis.

Whole genome scanning of a Mediterranean basin hotspot collection provides new insights into olive tree biodiversity and biology

Summary: In this study, the most complete genomic variation map and comprehensive catalog of molecular variation to date for 89 olive tree genotypes originating from the entire Mediterranean basin were developed, revealing the genetic diversity of this commercially significant crop tree and explaining the divergence/similarity among different variants. Additionally, the potential origin or routes of olive tree domestication were characterized through the study of the monumental ancient tree 'Throuba Naxos'.

Carbon sufficiency boosts phenylpropanoid biosynthesis early in peach fruit development priming superior fruit quality

Summary: The manipulation of crop load in peach trees affects carbon supply and the balance between fruit yield and quality. This study assessed the impact of carbon supply on peach fruit quality at different development stages and found that carbon-sufficient fruit had higher quality attributes. The analysis of the fruit's secondary metabolite profile revealed that early metabolic shifts primed fruit quality at harvest, and the synthesis of flavonoids was facilitated by enhanced carbon availability.

Summary: Greek landraces of dill have distinct morphological, genetic, and chemical traits compared to modern commercial cultivars. Landraces have desirable characteristics such as taller height, larger umbels, and denser foliage. Genetic divergence exists between landraces and cultivars, but some gene flow may have occurred. Greek landraces of dill have potential for breeding programs to develop improved cultivars.

Foliar Calcium Effects on Quality and Primary and Secondary Metabolites of White-Fleshed 'Lemonato' Peaches

Summary: This study investigated the effect of pre-harvest foliar calcium application on fruit quality, primary metabolite profile, antioxidant activity, and phenolic compounds of the 'Lemonato' peach. The results showed that the different calcium treatments had no significant effect on the fruit quality characteristics of this peach variety. The response of fruit quality, metabolites, and phenolic compounds to organic calcium and calcium-silicate treatments varied among different orchards, suggesting the involvement of genetic or environmental factors.

Summary: The preparation and properties of novel edible bigel systems based on olive oil oleogels structured with monoglycerides and hydrogels formed with gelatin or gelatin plus kappa-carrageenan were studied. The effect of hydrogel composition and oleogel/hydrogel ratio on physical and thermal properties of the bigels was examined. The results provide comprehensive information regarding the structure and functional properties of bigels with potential applications in foods.

Summary: The influence of incorporating post-distillation solid wastes of aromatic plants into wheat breads was investigated. The SWAP breads had darker color, higher levels of terpenoids and phenolic compounds, and enhanced antioxidant capacity. However, the strong aroma from the fortified breads at 2% levels was not acceptable by consumers.

Physiological and Metabolic Traits Linked to Kiwifruit Quality

Summary: An experiment was conducted to evaluate the relationship between dry matter (DM) and soluble solids concentration (SSC) with acidity content and taste in fully ripened 'Hayward' kiwifruits. Nutrient content and metabolomic analysis were also performed to compare high taste scores (HTS) and low taste scores (LTS) kiwifruits. The results show that non-destructive measurement of DM can serve as an indicator for kiwifruit taste, and that taste is influenced by SSC and acidity. Furthermore, there are clear differences in nutrient content and primary metabolites between kiwifruits with HTS and LTS.

Evaluation of the biocontrol potential of PGPB strains isolated from drought-tolerant tomatoes against fungal pathogens

Summary: Agrochemicals pose a major threat to human health and the environment, while Plant Growth Promoting Bacteria (PGPB) offer a potential alternative to increase crop yields and alleviate plant stress. This study evaluated rhizospheric and endophytic PGPB strains from a drought-tolerant tomato landrace as potential biocontrol agents. Results showed that 10 bacterial strains exhibited antifungal activity, while 8 strains produced ammonia and 5 strains produced hydrogen cyanide. The most effective strain, SAESo-E4, showed resistance to fungal infection in lettuce plants and induced defense-related and signaling pathways.

Emulsion gel enriched with a barley β-glucan concentrate for reducing saturated fat in biscuits

Summary: In biscuit production, substituting solid fats with barley flour emulsion gel can result in healthier and more acceptable products, depending on the level of fat replacement. CmCML11 interacts with CmCAMTA5 to enhance γ-aminobutyric acid (GABA) accumulation by regulating GABA shunt in fresh-cut cantaloupe

Summary: This study investigated the effect of calcium chloride (CaCl2) treatment on GABA accumulation in fresh-cut cantaloupe and the underlying mechanisms. The results showed that CaCl2 treatment increased GABA content and the activities of GAD and SSADH enzymes, while reducing glutamate content and GABA-T activity. Additionally, CaCl2 treatment upregulated the expressions of CmCML11 and CmCAMTA5, as well as several GABA shunt genes, through the transcriptional activation by CmCAMTA5. Furthermore, the interaction between CmCML11 and CmCAMTA5 enhanced the transcriptional activation of GABA shunt genes. Overall, this study reveals that CaCl2 treatment promotes GABA accumulation in fresh-cut cantaloupe through the combined effect of CmCML11 and CmCAMTA5 in regulating the expressions of GABA shunt genes.

Integrated physiological, biochemical, and transcriptomics analyses reveal the underlying mechanisms of high nitrogen use efficiency of black sesame

Summary: This study identified the high nitrogen use efficiency (NUE) black sesame variety 17-156 and analyzed its underlying physiological and molecular mechanisms. The results showed that 17-156 possesses a sophisticated nitrogen metabolizing machinery to uptake and assimilate higher quantities of inorganic nitrogen, simultaneously improving carbon metabolism and growth. Many important genes were up-regulated in 17-156 under high nitrogen condition. Additionally, 38 potential candidate genes were identified for future studies to improve sesame's NUE. These findings provide valuable resources for understanding the regulatory network of nitrogen metabolism and developing sesame cultivars with improved NUE.

Phosphate deficiency responsive TaSPX3 is involved in the regulation of shoot phosphorus in Arabidopsis plants

Summary: This study cloned the full-length cDNA sequence of TaSPX3 gene in wheat and found that TaSPX3 responds to low phosphorus stress in multiple wheat genotypes. Overexpressing TaSPX3 can alleviate phosphorus deficiency symptoms and promote plant growth in Arabidopsis. The study also revealed the interaction of TaSPX3 with other genes related to the phosphorus starvation signaling pathway.

Summary: Selenium (Se) can reduce uptake and translocation of cadmium (Cd) in plants by regulating root morphology. This study investigated the effects of Se(IV) on root exudates, root morphology, root endogenous hormones, and Cd uptake efficiency in rice under Cd stress. The results showed that Se(IV) significantly reduced Cd concentrations in shoots and roots, and decreased Cd uptake efficiency via root hairs. Se(IV) also affected root morphology, root exudates, and the synthesis of hormones like IAA and JA. However, transcriptome analysis revealed no upregulated differentially expressed genes (DEGs) in IAA synthesis.

Cell number regulator 8 from Salix linearistipularis enhances cadmium tolerance in poplar by reducing cadmium uptake and accumulation

Summary: This study identified a cell number regulator gene called SlCNR8 in willow, which enhances resistance to trace metals in transgenic poplar seedlings. SlCNR8 reduces Cd uptake and accumulation, and can be used as a candidate gene for genetic improvement of phytostabilisation of trace metals.
