Auraptene regulates Th1/Th2/TReg balances, NF-κB nuclear localization and nitric oxide production in normal and Th2 provoked situations in human isolated lymphocytes

Background: Auraptene as member of dietary coumarins, is found in citrus fruits. Former studies have demonstrated its anti-inflammatory and anti-cancer activity. Purpose: The mechanism of action and immune-modulatory property of this compound on human lymphocytes are greatly unknown. Study Design/Methods: The effect of three concentrations (10, 30 and 90 mu M) of auraptene or dexamethasone (0.1 mM) were evaluated on percentage of cell proliferation and nitric oxide (NO) production as well as secretion and gene expression of cytokines, and NF-kappa B level in PHA-stimulated and non-stimulated lymphocytes. Results: In non-stimulated cells, all three concentrations of auraptene significantly increased the gene expression index of IL-10 (P < 0.05-0.001). The IFN gamma gene expression index, IFN gamma/IL 4 and IL-10/IL-4 gene expression ratio were significantly increased due to the high concentration (90 mu M) of auraptene treatment compared to control group (P < 0.05 0.001). In PHA stimulation, all three concentrations of the extract significantly decreased proliferation, cytokines (IL-4, IL-10 and IFN-gamma) and NF-kappa B level as well as NO production, but IFN-gamma/IL-4 and IL-10/IL-4 ratio were significantly increased compared control group (P < 0.05 0.001). Gene expression of IL-10 and IL-4 was decreased but that of IFN-gamma as well as FN-gamma/IL-4 and IL-10/IL-4 ratio were significantly increased due to all three concentrations of auraptene. Conclusion: The results showed promoting effects of auraptene on T cell subsets toward Th-1 (IFN-gamma) and T-reg (IL10), which suggest its therapeutic value for treatment of Th-2 cells predominant diseases including allergic disease such as asthma and atopic dermatitis as well as cancers.

Summary: Zn- and Cu-doped 13-93B3 borate mesoporous bioactive glasses (MBGs) were synthesized using nitrate precursors in the presence of Pluronic P123. The mineralization process of hydroxyapatite (HAp) on the glass surface was investigated by studying the changes in dynamic surface tension (SFT) of simulated body fluid (SBF). The doped MBGs showed changes in various properties and the SFT of SBF was found to be an important factor for predicting and confirming the HAp mineralization process.

Comparative study on the impacts of visnagin and its methoxy derivative khellin on human lymphocyte proliferation and Th1/Th2 balance

Summary: This study aimed to investigate and compare the anti-inflammatory properties of visnagin and its methoxy derivative khellin on human lymphocytes. The results showed that khellin and visnagin significantly reduced cell proliferation, malondialdehyde (MDA), and nitric oxide (NO) levels, and increased glutathione (GSH) and interleukin (IL)-10 levels. Khellin had a more prominent anti-inflammatory effect compared to visnagin.

The results demonstrated that G. glabra could concentration-dependently mitigate adhesion formation through its anti-inflammatory, anti-fibrosis, and anti-oxidant properties. However, further clinical investigations are required to confirm G. glabra as a promising candidate against post-surgical adhesive complications.

Promising influences of zingerone against natural and chemical toxins: A comprehensive and mechanistic review

Summary: In this study, the therapeutic and protective properties of zingerone were assessed against various chemical and natural toxins. Zingerone showed anti-inflammatory, antioxidant, and anti-apoptotic properties, and provided protection against radiation, chemicals, biological toxins, and oxidative stress. It also exhibited strong radioprotective properties. The authors hope that this review will inspire researchers to conduct further studies on the pharmaceutical applications and efficiency of zingerone in cancer treatment and drug adverse effects.

Low Doses of beta-Caryophyllene Reduced Clinical and Paraclinical Parameters of an Autoimmune Animal Model of Multiple Sclerosis: Investigating the Role of CB2 Receptors in Inflammation by Lymphocytes and Microglial

Summary: This study investigated the protective effect of low doses of beta-Caryophyllene (BCP) on experimental autoimmune encephalomyelitis (EAE) mice, a chronic model of multiple sclerosis (MS). The results showed that BCP can reduce inflammation and improve pathological and clinical defects in EAE mice by modulating the immune system. BCP also increased the levels of anti-inflammatory cytokine IL-10 and decreased blood inflammatory cytokines. Therefore, low doses of BCP may be suggested as a therapeutic approach for MS with potent anti-inflammatory effects and possibly lower toxicity.
