CYBB/NOX2 in conventional DCs controls T cell encephalitogenicity during neuroinflammation

Summary: This study reveals that the nicotinamide adenine dinucleotide phosphate (NADPH) oxidase 2 in conventional dendritic cells (cDCs) regulates MOG antigen processing through LC3-associated phagocytosis (LAP), enabling encephalitogenic T helper cells to initiate and sustain autoimmune neuroinflammation. Genetic ablation of Cybb in cDCs restrains T-H cell recruitment into the CNS and ameliorates clinical disease development upon the adoptive transfer of MOG-specific CD4(+) T cells.

Summary: This study characterized the metabolic profile of hiPSC-CM during maturation using various methods and found that their metabolic pathways and substrate utilization were progressively restructured. As time progressed, the oxidative capacity and fatty acid utilization of hiPSC-CM increased, while glucose oxidation decreased. There was evidence of increased coupling of glycolysis to mitochondrial respiration and decreased glycolytic branch pathways. These findings are important for studies that involve metabolism in hiPSC-CM.

Inhibition of T cell recruitment facilitates axonal regeneration.

A conditional null allele of Dync1h1 enables targeted analyses of dynein roles in neuronal length sensing

Summary: Size homeostasis is essential for cells, especially large cells like neurons. A motor-dependent length-sensing mechanism involving reduction in microtubule motor levels was proposed and validated in Dync1h1Loa sensory neurons. In this study, a new mouse model with a conditional deletion allele of exons 24 and 25 in Dync1h1 was described. Heterozygous mice with reduced dynein expression exhibited accelerated growth, mild impairments in motor functions, and delayed recovery from peripheral nerve injury.

Summary: Signaling between the endoplasmic reticulum (ER) and mitochondria is essential for regulating various physiological processes. A recent study suggests that the binding of VAPB and PTPIP51, mediated by the FFAT motif in PTPIP51, is crucial for this signaling. However, cellular experiments supporting this finding are lacking. In this study, we show that the coiled-coil domain in PTPIP51, but not the FFAT motif, is essential for its role in ER-mitochondria contacts and Ca2+ delivery.

Summary: Fluorescent InP-based quantum dots, combined with zinc and ZnS or CdS shells, have been synthesized using zinc carboxylates and diethyldithiocarbamate precursors. The addition of zinc enhances the emissive and structural properties of the quantum dots, while reducing interfacial defects. Structural analysis reveals that the core/shell particles are homogeneous extended alloys with full-depth inclusion of zinc.

Characterising the chemical and physical properties of phase-change nanodroplets

Summary: Phase-change nanodroplets, composed of a stabilising shell and a perfluorocarbon core, have recently gained attention as ultrasound theranostic nanoparticles. Nanodroplets can vaporise into echogenic microbubbles forming cavitation nuclei when exposed to ultrasound. However, methods to quantify the perfluorocarbon core in nanodroplets are lacking. In this study, nanodroplets were fabricated using lipids shell and perfluorocarbons, and the amount of perfluorocarbon in the core was assessed using 19F NMR and FTIR. The results showed that the fluorine signal accurately correlated with the perfluorocarbon concentration, allowing for quantification of the core. The study also found that different perfluorocarbons had varying rates of content loss and generated cavitation comparable to commercial contrast agent microbubbles.
