Phytotoxic Activity of Metabolites Isolated from Rutstroemia sp.n., the Causal Agent of Bleach Blonde Syndrome on Cheatgrass (Bromus tectorum)

A fungal pathogen soon to be described as Rutstroemia capillus-albis (Rutstroemiaceae, Helotiales, Leotiomycetes) has been identified as the causal agent of bleach blonde syndrome' on the invasive annual grass weed Bromus tectorum (cheatgrass) in western North America. This apparently common but previously undescribed disease causes premature senescence and sterility, but does not affect seed germination or seedling emergence and growth. This study investigated whether the new species produces phytotoxins that could be implicated in pathogenesis. The compounds 9-O-methylfusarubin, 9-O-methylbostrycoidin, 5-O-methylnectriafurone, trans-methyl-p-coumarate and terpestacin were isolated from the solid culture of this fungus. The undescribed absolute stereochemistry at C-3 of 9-O-methylfusarubin and at C-1' of 5-O-methylnectriafurone were assigned by applying electronic and vibrational circular dichroism (ECD and VCD) combined with computational methods and the advanced Mosher's method, respectively. The first three listed compounds are naphtoquinone pigments, while terpestacin is a sesterterpene, and trans-methyl-p-coumarate could be the product of an unusual fungal phenylpropanoid biosynthesis pathway. In a juvenile plant immersion bioassay, both 9-O-methylfusarubin and terpestacin proved to be highly toxic at 10(-4) M, causing wilting and plant death within 10 days. This finding suggests that these two compounds could play a role in pathogenesis on B. tectorum.

In silico pharmacokinetics, molecular docking and dynamic simulation studies of endolichenic fungi secondary metabolites: An implication in identifying novel kinase inhibitors as potential anticancer agents

Summary: Seventy-three anticancer metabolites from different endolichenic fungi previously reported were analyzed for drug likeliness and pharmacokinetic properties. Three compounds, including talaromycin A, pyridoxatin, and terricollene A, showed strong binding affinity and stability, potentially serving as glycolytic inhibitors. These findings contribute to the development of potential glycolytic inhibitors for treating and controlling various forms of cancer, but further experimental investigations are needed to evaluate the efficacy of these compounds.

Synthesis and Stereodynamic and Emission Properties of Dissymmetric Bis-Aryl Carbazole Boranes and Identification of a CPL-Active B-C Atropisomeric Compound

Summary: Bis-aryl carbazole borane derivatives with emissive properties and axial chirality were synthesized. The resolution of a thermally stable atropisomeric pair (compound 1b), which has a B-C chiral axis, was achieved using chiral stationary-phase high-performance liquid chromatography (CSP-HPLC). The complete photophysical properties of all compounds were measured and simulated using time-dependent density functional theory (TD-DFT) calculations, and circularly polarized luminescence spectra were obtained for the atropisomers of compound 1b, with their absolute configuration determined by a TD-DFT simulation of the electronic circular dichroism (ECD) spectra.

Structure-Activity Relationship (SAR) Study of trans-Cinnamic Acid and Derivatives on the Parasitic Weed Cuscuta campestris

Summary: This study investigates the inhibitory effects of trans-cinnamic acid and its derivatives on the growth of Cuscuta campestris, a parasitic weed causing significant yield losses in crops. The findings reveal that some derivatives of trans-cinnamic acid exhibit enhanced growth inhibition activity against C. campestris seedlings, with the methyl ester derivative being the most active compound. This structure-activity relationship study provides valuable information for the design of herbicidal treatments targeting parasitic weeds.

Strong Chiroptical Effects in the Absorption and Emission of Macrocycles Based on the 2,5-Diaminoterephthalate Minimal Fluorophore

Summary: Chiral fluorescent macrocycles, consisting of dimethyl 2,5-diaminoterephthalate units, can be easily synthesized from inexpensive building blocks. Depending on the concentration, the reaction mainly produces a paracyclophane-like dimer or a triangular trimer. These macrocycles exhibit fluorescence in solution and the solid state, with red-shifted maxima as the size of the macrocyclic ring decreases. Chirality affects their absorption and emission of circularly polarized light.

Summary: This study purified extracts from Ephedra foeminea and identified bioactive compounds with antimicrobial and antibiofilm properties. The compound kaempferol-3-O-alpha-L-(2 '',4 ''-di-E-p-coumaroyl)-rhamnopyranoside exhibited strong antibacterial and antibiofilm activity against S. aureus strains. Molecular docking studies suggested its potential applicability in biomedical and biotechnological fields.

Ethylidene-Tethered Chromene-Pyrone Hybrids as Potential Plant-Growth Regulators from an Endolichenic Phaeosphaeria Species

Summary: Six new dimeric spiciferones (1-6) and four known metabolites (7-10) were produced by the lichen-associated fungus Phaeosphaeria sp. The new structures were determined through spectroscopic analysis, and their absolute configurations were established using electronic circular dichroism calculations. Compounds 1 and 3-6 represent the first instances of ethylidene-bridged dimers from the building blocks 4H-chromene-4,7(8H)-dione and alpha-pyrone, and compound 2 is a unique homodimer of spiciferone. Compounds 1, 2, and 5-9 exhibited significant growth inhibition on the dicot weed-like plant Arabidopsis thaliana at a concentration of 100.0 μM. Notably, compound 8 showed the strongest inhibitory activity against the fresh weight and root elongation of A. thaliana, with IC50 values of 32.04 and 26.78 μM, respectively, while compounds 1, 8, and 9 promoted the growth of A. thaliana at lower concentrations. Additionally, compounds 2 and 6 exhibited weak inhibition on the root elongation of the monocot rice, while compounds 1 and 8 exhibited dose-dependent growth-promoting effects on the shoot and root elongation of rice.

Specialized Metabolites Produced by Phytotopatogen Fungi to Control Weeds and Parasite Plants

Summary: Weeds, including parasite plants, have become a serious problem for farmers, causing significant losses in agricultural and herding production and hindering reforestation practices and important infrastructure. To combat these pests, synthetic herbicides have been widely used, leading to environmental pollution and risks to human and animal health. This review explores the potential of fungal phytotoxins as bioherbicides, as well as the commercial availability and future perspectives of bioherbicides based on microbial toxic metabolites.

Production of (10S,11S)-(-)-epi-Pyriculol and Its HPLC Quantification in Liquid Cultures of Pyricularia grisea, a Potential Mycoherbicide for the Control of Buffelgrass (Cenchrus ciliaris)

Summary: (10S,11S)-(-)-epi-pyriculol is a phytotoxic metabolite produced by Pyricularia grisea, a fungus that infects the invasive weed species buffelgrass. This study developed a high-pressure liquid chromatography (HPLC) method for quantifying (10S,11S)-(-)-epi-pyriculol in complex mixtures. The results showed that the production of (10S,11S)-(-)-epi-pyriculol varied among different P. grisea isolates and was strongly correlated with phytotoxic activity. The HPLC method could be used for selecting the best strain and cultural conditions for mass production of (10S,11S)-(-)-epi-pyriculol, thus providing a new bioherbicide for the control of buffelgrass.

Alternaria alternata Isolated from Infected Pears (Pyrus communis) in Italy Produces Non-Host Toxins and Hydrolytic Enzymes as Infection Mechanisms and Exhibits Competitive Exclusion against Botrytis cinerea in Co-Infected Host Fruits

Summary: Alternaria alternata is a devastating phytopathogenic fungus that causes black spots in many fruits and vegetables. In this study, the pathogenicity mechanisms and competitive exclusion of A. alternata were investigated, with a focus on its interaction with Botrytis cinerea. The fungus was found to produce strong enzymatic activities and four different toxins, which showed varying levels of necrotic activity on different hosts. Interestingly, A. alternata exhibited competitive exclusion of B. cinerea in host and non-host pear fruits, inhibiting its growth by 70% and 65%, respectively. The toxins Alteichin and alternariol 4-methyl ether showed the strongest inhibition activity against B. cinerea.

These instrumental techniques are valuable for mapping and understanding the supramolecular structures in thin films of chiral organic materials.

CPL calculations of [7]helicenes with alleged exceptional emission dissymmetry values

Summary: Any device based on materials exhibiting circularly polarized luminescence (CPL) requires a high degree of circular polarization, quantified through the dissymmetry factor g(lum) and related quantities such as the CPL brightness. However, maximizing |g(lum)| is not an easy task. Computational tools able to predict dissymmetry values in an accurate way are therefore of utmost importance. Our study tests the predictive power of time-dependent density functional theory (TD-DFT) combined with solvation effects in determining large g(abs) and g(lum) values for specific helicene-based materials.

Summary: We have investigated the effect of achiral substituents on the chiral supramolecular architectures of diketopyrrolo[3,4-c]pyrrole-1,2,3-1H-triazole (DPP) dyes. By decorating the same chiral DPP motif with different substituent groups, we observed significant differences in optical, chiroptical, electrochemical and thermal properties among the three dyes. Our findings provide valuable insights for the development of chiral organic opto-electronics and other innovative technological applications.
