Impairment of Novel Object Recognition Memory and Brain Insulin Signaling in Fructose- but Not Glucose-Drinking Female Rats

Excessive sugar intake has been related to cognitive alterations, but it remains unclear whether these effects are related exclusively to increased energy intake, and the molecular mechanisms involved are not fully understood. We supplemented Sprague-Dawley female rats with 10% w/v fructose in drinking water or with isocaloric glucose solution for 7 months. Cognitive function was assessed through the Morris water maze (MWM) and the novel object recognition (NOR) tests. Plasma parameters and protein/mRNA expression in the frontal cortex and hippocampus were determined. Results showed that only fructose-supplemented rats displayed postprandial and fasting hypertriglyceridemia (1.4 and 1.9-fold, p < 0.05) and a significant reduction in the discrimination index in the NOR test, whereas the results of the MWM test showed no differences between groups. Fructose-drinking rats displayed an abnormal glucose tolerance test and impaired insulin signaling in the frontal cortex, as revealed by significant reductions in insulin receptor substrate-2 protein levels (0.77-fold, p < 0.05) and Akt phosphorylation (0.72-fold, p < 0.05), and increased insulin-degrading enzyme levels (1.86-fold, p < 0.001). Fructose supplementation reduced the expression of antioxidant enzymes and altered the amount of proteins involved in mitochondrial fusion/fission in the frontal cortex. In conclusion, cognitive deficits induced by chronic liquid fructose consumption are not exclusively related to increased caloric intake and are correlated with hypertriglyceridemia, impaired insulin signaling, increased oxidative stress and altered mitochondrial dynamics, especially in the frontal cortex.

Summary: The study investigated the anti-steatotic effect of Bempedoic acid (BemA) in a rat model of fatty liver induced by a high-fat diet supplemented with liquid fructose. The results showed that BemA-treated rats exhibited lower body weight, higher liver/body weight, and reduced hepatic steatosis compared to the control group. In addition to inhibiting ATP-citrate lyase (ACLY), BemA was found to have three novel mechanisms, including reducing fructose intake and de novo lipogenesis, increasing liver triglyceride export, and promoting hepatic fatty acid beta-oxidation. These findings suggest that BemA may be a potential approach for treating hepatic steatosis and preventing the progression of non-alcoholic fatty liver disease.

Integration of ligand and structure-based pharmacophore screening for the identification of novel natural leads against Euchromatic histone lysine methyltransferase 2 (EHMT2/G9a)

Summary: In this study, a new natural lead against Protein Lysine Methyltransferase 2 (EHMT2/G9a) was identified using a blended ligand and structure-based pharmacophore screening approach. EHMT2/G9a is associated with cancer, Alzheimer's, and aging, and is considered an emerging drug target. Two pharmacophores, based on the common features of known inhibitors and the interaction profile of crystal structures, were developed and validated. These pharmacophores were used in combination to screen over 741,543 compounds. The screening process also included tests for drug-likeness and toxicity, and the potential inhibitors were further analyzed through docking, simulation, and comparative analysis.

Repurposing of Raltitrexed as an Effective G9a/EHMT2 Inhibitor and Promising Anti-Alzheimer's Agent

Summary: In this study, we report for the first time the inhibition of G9a/EHMT2 and the anti-Alzheimer's activities of the drug raltitrexed. Raltitrexed inhibits G9a directly and reduces the levels of H3K9me2/ H3K9 in an AD transgenic worm model. Furthermore, raltitrexed exhibits a safe toxicity/function range and enhances locomotive function in worms, while reducing amyloid beta aggregates.
