Summary: Lung cancer is a deadly disease with a high incidence and mortality rate. The epithelial-mesenchymal transition (EMT) process in cancer cells plays a crucial role in metastasis. Ethacrynic acid (ECA) has been found to inhibit EMT in lung cancer cells. However, the effect of ECA on immune checkpoint molecules in cancer has not been fully studied.

Summary: The progression of diabetic kidney disease (DKD) is associated with increased fibronectin (FN) levels in proximal tubular epithelial cells. Remodelling of cell adhesion in DKD is closely related to epithelial-mesenchymal transition (EMT). Integrin 86, a transmembrane protein that regulates cell adhesion and migration, and its ligand FN play significant roles in DKD. The expression of integrin 86 and EMT are elevated in DKD, and their aggravation can be reduced by knockdown of integrin 86 or Notch1. Furthermore, urinary integrin 86 is significantly increased in DKD patients.

Summary: Neutrophil extracellular traps (NETs), released by neutrophils, have been found in colorectal cancer (CRC) tissues and are associated with disease progression. This study used high-resolution microscopy to detect citrullinated NETs in colon cancer tissues and found that citrullinated NETs were significantly associated with high tumour grades and lymph node metastasis. In vitro experiments also showed that purified NETs induced filopodia formation and cell motility in CRC cell lines, indicating their potential contribution to CRC metastasis.

Summary: Chemoresistance resulting from cancer stem cells (CSCs) and epithelial-mesenchymal transition (EMT) leads to inconsistent chemotherapeutic efficacy. The presence of CSCs and EMT allows cancer cells to switch between differentiated and stem-like states, known as cellular plasticity. Phosphorylated signal transducer and activator of transcription 3 (pSTAT3) is increasingly recognized as a major factor in CSCs and EMT, with preclinical studies showing reversal of chemoresistance through STAT3 pathway inhibition. This review focuses on mechanisms involving STAT3 and its target genes in regulating EMT, and also discusses the status of clinical trials using STAT3 pathway inhibitors.

Summary: The study found PRDX5 to be highly expressed in non-small cell lung cancer tissues, and its demethylation status in the promoter region was related to the TNM stage. The binding of STAT3 to PRDX5 promoter region enhanced PRDX5 protein expression. PRDX5 promoted cell migration and invasion under oxidative stress conditions in NSCLC through activation of the Nrf2 signaling pathway.

Summary: It has been reported that the activities of NAD(+)-dependent deacetylase decrease in the aging mouse liver, while NMN treatment can increase healthspans. However, the changes in the acetylomic landscape and the effects of NMN treatment on protein acetylation in the aging liver have not been studied. In this study, immunoprecipitation coupled with label-free quantitative LC-MS/MS was used to identify the acetylome and investigate the effects of aging and NMN on liver protein acetylation. The results showed that the acetylation levels of proteins involved in oxidation-reduction and metabolic processes increased in aged livers, while the acetylation levels of proteins involved in transport and translation processes decreased. NMN treatment inhibited the age-related increase in acetylation of proteins regulating fatty acid beta oxidation, the TCA cycle and valine degradation.

Summary: This study proposes the concept of single-atom-kernelled nanoclusters and synthesizes a Pd-based trimetal nanocluster with a single-Ag atom-kernel using an alloying strategy. The nanoclusters with single-silver atom kernels exhibit higher activity and selectivity compared to the bimetal precursor nanoclusters. The single atoms in the kernel have significant influence on catalytic performance by affecting the electronic configuration.

Summary: There are unresolved fundamental issues regarding metal nanoclusters, including the transformation from face-centered cubic (fcc) to hexagonal close-packed (hcp) structures and photoexcitation electron transfer. This study successfully achieved the fcc-to-hcp structure transformation and revealed size and shape-dependent photoexcitation electron transfer for metal nanoclusters. A new thermally induced ligand exchange method was developed to synthesize a rod-shaped hcp Au42(SCH2Ph)32 nanocluster, which exhibited specific absorption and dual emission properties. The findings also suggest a possible structure transformation pathway and predict a novel rod-shaped hcp Au58(SR)40 nanocluster with a larger aspect ratio. Additionally, a kernel-based middle-to-both ends sp <- sp photoexcitation electron transfer mechanism was observed in rod-like gold nanoclusters, indicating the potential for tunable and practical applications.

Summary: Eukaryotic organisms adapt to environmental fluctuations by altering their epigenomic landscapes and transcriptional programs. Histone H2Bs are globally degraded in Caenorhabditis elegans during starvation, and the mechanisms underlying this degradation involve ubiquitin and ubiquitin-related enzymes. The retention of aberrant nucleosomal H2Bs leads to abnormal gene expression and impacts animal viability. Furthermore, the ubiquitin-proteasome system regulates chromosomal histone turnover in human cells.

Summary: This study reveals the mechanism of airborne defense (AD) against aphids and viruses. The volatile compound MeSA is perceived by neighboring plants and converted into salicylic acid, which activates a signaling cascade to induce plant anti-aphid immunity and reduce virus transmission. Aphid-transmitted viruses encode proteins that suppress AD, making plants less repellent to aphids and more suitable for aphid survival, infestation, and viral transmission.

Summary: This study found that mitochondrial dysfunction, rotenone treatment, elevated Ca2+ levels, and activation of the JNK-STAT3 pathway all contribute to increased resistance of glioblastoma to TMZ. Inhibiting JNK or STAT3 can increase sensitivity to TMZ. Co-administration of TMZ with a JNK or STAT3 inhibitor holds promise as an effective treatment for glioblastoma.
