Panax ginseng extract (PGe) has been shown to possess immunomodulatory effects in healthy dairy cows at drying off and to trigger an adequate immune response to protect from an experimental intramammary infection (IMI) with Staphylococcus aureus in a murine model. S. aureus is one of the major pathogens isolated from bovine IMI; being capable to invade and survive within mammary epithelial cells. However, the precise mechanism by which PGe interacts with bovine mammary epithelial cells (MAC-T) and bovine macrophages in the course of a S. aureus infection remains unclear. We evaluated the effect of PGe on MAC-T cytokine response and on the internalization of S. aureus into MAC-T. In addition, we evaluated the effect of PGe on the phagocytic activity of macrophages isolated from bovine mammary secretions. Results shown that MAC-T cells TLR4 and NF-kappa B mRNA expression was not affected by PGe at all evaluated times. IL-6 mRNA expression and protein level and IL-4 protein level were significantly induced in MAC-T treated with 3 mg/ml of PGe. PGe at 3 mg/ml reduced significantly the internalization of two S. aureus strains in MAC-T. In addition, PGe did not affect the percentage of phagocytosis and the NO and ROS production of macrophages co-cultured with two strains of S. aureus. These results, obtained in in vitro models together with those obtained in in vivo previous studies carried out in bovines and mice can contribute to improve the understanding of the effects of PGe following inoculation in bovine mammary glands.

Involvement of matrix metalloproteinases and their inhibitors in Staphylococcus aureus chronically infected bovine mammary glands during active involution

Summary: The study revealed that during active involution, the expression and activity of MMP-2 and -9 in mammary tissue were significantly higher in S. aureus-infected quarters compared with uninfected quarters, along with increased expression of TIMP-1 and -2. The imbalance of MMPs/TIMPs could lead to more severe damage to mammary tissue in S. aureus-infected quarters.

Characterization of an incipient granulosa cell tumour in a Holstein cow: Steroid hormone receptors and coregulators expression

Summary: This study aimed to describe a case of incipient formation of granulosa cell tumor (GCT) in a 7-year-old Holstein cow and characterize it by its immunohistochemical pattern and hormonal profile. The research found that a highly differentiated pattern could be determined through a panel of immunohistochemical markers in the GCT concomitant with normal follicular development. This study provides new insights into GCTs in cattle before ovarian function is compromised.

Summary: The functional capability of a cross-linked hydrogel composed of sulfated glycosaminoglycans and a cationic cellulose for treating osteoarthritis was demonstrated in experimental animal models. In long-term follow-up after implantation, the hydrogel showed superior safety, permanence, and reduced cartilage damage compared to commercial hyaluronic acid.

Application of an optimized and validated LC-MS/MS method for the quantification of free 3-nitrotyrosine in plasma, urine and liver tissue of lactating dairy cows

Summary: In dairy cows, oxidative stress during the transition period can be influenced by environmental conditions such as heat stress. A study developed and validated a method for accurate determination of 3-NT in blood, urine, and liver samples in dairy cows using LC-MS/MS. Results showed higher 3-NT concentrations in cows that gave birth during spring, along with higher BHBA and lower glucose concentrations. Liver TAG content and aspartate transaminase activity were also higher in cows that gave birth during spring.

A review on inflammation and angiogenesis as key mechanisms involved in the pathogenesis of bovine cystic ovarian disease

Summary: Cystic ovarian disease (COD) is a significant cause of reproductive failure in dairy cattle. This review focuses on the role of inflammation and angiogenesis in the development of follicular cysts, particularly in bovine species. The study suggests that changes in the expression of cytokines and angiogenic factors contribute to the abnormal persistence of follicular cysts. Pro-inflammatory and anti-inflammatory cytokines play a crucial role in regulating angiogenesis, particularly in ovarian cells from follicular cysts and persistent follicles. The imbalance in cytokine expression and pro-angiogenic growth factors may contribute to follicular persistence and the recurrent appearance of COD in dairy cattle.

Summary: This study evaluated the impact of heat stress during gestation on molecular aspects at the ovarian level in adult offspring. The results showed associations between heat stress variables during gestation and the methylation status of CpG sites in the 5'UTR of ESRI and ESR2, as well as the expression of ESRI, ESR2, PGR, and coregulatory proteins in dominant follicles of adult cows.

These findings suggest 1A-116 as a promising treatment option for glioma.

Summary: In this study, self-assembled bovine serum albumin nanoparticles loaded with genistein were PEGylated to avoid possible immunogenic reactions and rapid clearance from the bloodstream. The PEGylation process increased the particle size of the nanoparticles and improved their biological properties, reducing cytotoxicity and genotoxicity.

Summary: This study reported the synthesis and performance of thermoresponsive nanogels based on poly(N-isopropylacrylamide) (pNIPAM). The cytotoxicity, internalization ability, antigen protection and delivery capability of the nanogels were evaluated in vitro and in vivo. The results demonstrated that the pNIPAM nanogels can stimulate a humoral immune response.

Biomarkers of oxidative stress and liver function in early lactation and their relationship with the reproductive efficiency of multiparous grazing dairy cows in Argentina. A retrospective study

Summary: This study aimed to analyze the possible relationship between days to conception and oxidative stress biomarkers and liver functional parameters in dairy cows. A reliable method for the accurate measurement of malondialdehyde (MDA) was developed. The results showed that there were no differences in systemic concentration of certain biomarkers between high and low days to conception groups. However, cholesterol concentration, plasma 3-nitrotyrosine (3-NT) concentration, and superoxide dismutase activity were found to be significantly different between the two groups. Lower levels of 3-NT and MDA in the liver were associated with better reproductive performance in dairy cows.

Effect of Parenteral Supplementation of Minerals and Vitamins on Oxidative Stress Biomarkers and Hepatic Fatty Acid Metabolism in Dairy Cows During the Transition Period

Summary: This study aimed to investigate the effects of parenteral vitamin and mineral supplementation on hepatic fatty acid metabolism and oxidative stress biomarkers in transition cows. The supplemented group received injections of vitamin A palmitate, vitamin E acetate, copper edetate, zinc edetate, manganese edetate, and sodium selenite, while the control group received injections of sodium chloride. Results showed that supplemented animals had higher GSH-Px activity, lower and higher concentration of 3-NT in the liver and plasma, respectively, higher expression of carnitine palmitoyltransferase 1, and lower triacylglycerol content in the liver. No differences were found in other parameters between groups. These findings suggest that vitamin and mineral supplementation has beneficial effects on GSH-Px activity, hepatic 3-NT concentration, and metabolic adaptation in transition cows.
