Summary: The development of an anti-HER2/anti-FAP/anti-mPEG tri-specific antibody (TsAb) for converting mPEG-coated liposomal doxorubicin to immunoliposomes shows improved tumor accumulation and therapeutic efficacy, overcoming the challenges posed by TAFs in solid tumor treatment. The TsAb-modified mPEGylated liposomes exhibit specific targeting and enhanced cytotoxicity against both HER2(+) breast cancer cells and FAP(+) TAFs.

Summary: Membrane protein complexes play a crucial role in various biological functions, but their binding affinity is less explored compared to globular proteins. Mutations in these complexes can affect binding affinity and impair critical functions, leading to diseases. To address this, the MPAD database was developed, providing experimental binding affinity data, sequence, structure, and functional information, membrane-specific features, experimental conditions, and literature information for membrane protein-protein complexes and mutants. MPAD can be used to understand the factors influencing binding affinity and the impact of mutations on membrane proteins, with potential applications in structure-based drug design.

Summary: The transmembrane protein GARP binds latent TGF-beta 1 to form complexes on the surface of Tregs, B-cells, and platelets, which release active TGF-beta 1 upon stimulation. Blocking TGF-beta 1 activation by Tregs with anti-GARP:TGF-beta 1 mAbs overcomes resistance to PD1/PD-L1 blockade in murine tumors. Furthermore, antibody-mediated blockade of GARP:TGF-beta 1 or genetic deletion of Garp in Tregs, B cells, or platelets does not alter immune responses in mice, suggesting potential for cancer immunotherapy without impairing other immune defenses.
