Summary: Deciding whether to grow or divert energy to stress responses is a major trade-off for plants in fluctuating environments. Our study reveals that three leucine-rich repeat receptor kinases (LRR-RKs) act as direct receptors for PLANT PEPTIDE CONTAINING SULFATED TYROSINE (PSY)-family peptides, mediating the switching between two opposing pathways. Unlike other known LRR-RKs, PSY receptors (PSYRs) activate the expression of stress response transcription factors when the ligands are depleted. Loss of PSYRs leads to defects in plant tolerance to both biotic and abiotic stresses. This ligand-deprivation-dependent activation system allows plants to regulate stress responses in tissues near damaged sites where ligand production is impaired.

Researchers found that protein tyrosine sulfation can weaken ABA signaling by interacting with and sulfating SnRK2.2/2.3/2.6, leading to their degradation. This discovery reveals a novel mechanism for desensitizing ABA responses through protein sulfation.

Summary: Tyrosine-sulfated peptides play a crucial role in regulating plant growth and development, with the disulfated pentapeptide phytosulfokine (PSK) mediating growth through specific receptors. A transcriptome analysis in mutants lacking sulfated peptides identified genes regulated by PSK and other sulfated peptides, showing effects on root growth and development. Additionally, the study revealed unexpected synergistic effects of ligand and receptor deficiencies on root phenotype.

Summary: In this study, a MS-based workflow for sY-peptide characterization was developed, allowing systematic investigation of the sulfome. The key for distinguishing sulfated peptides from phosphopeptides was low collision energy-induced neutral loss. Applying this workflow identified 21 new sulfotyrosine-containing proteins and revealed new interplay between enzymes relevant to both protein and glycan sulfation.

Summary: In this study, high-definition FAIMS was directly coupled to ultrahigh-resolution MS to explore the isotopic fine structure in FAIMS spectra. Different elemental isotopic substitutions produced orthogonal shifts in FAIMS space, forming multidimensional matrices and 3-D trajectories that delineate all considered isomers. This research takes the structural isotopic shift approach to the next level.

Summary: Tyrosine sulfation is a post-translational modification that enhances protein-protein interaction specificity. Sulfotyrosyl residues (sTyrs) play important roles in various biological processes, including high-affinity interactions, viral entry, thrombin interaction, and collagen binding. The lack of discernible binding site consensus makes sTyr an interesting research topic. Improved methods and advanced techniques will contribute to the discovery and understanding of sTyr occurrence and function.

Summary: Protein modifications, including phosphorylation and acetylation, are critical for synaptic plasticity and memory. Tyrosine sulfation, another protein modification, has been shown to play important roles in various biological processes. In this study, it was demonstrated that sulfation contributes to long-term potentiation (LTP) and memory formation. Inhibition of sulfation impaired long-term memory without affecting acquisition or short-term memory. Additionally, LTP-inducing stimulus enhanced protein tyrosine sulfation. These findings suggest that tyrosine sulfation plays an important role in LTP and memory.
