Summary: Endoscopy and tissue biopsy are the standard methods for diagnosing esophageal adenocarcinoma, but they are invasive and not suitable for population screening. In this study, a surface-enhanced Raman scattering lectin-immunoassay was developed to accurately measure serum levels of specific proteins. The results suggest that this method could be a promising liquid biopsy platform for screening esophageal adenocarcinoma.

Summary: SERS analysis coupled with classification algorithms is a promising candidate for point-of-care medical diagnosis. However, there are still gaps in our understanding of the biochemical information provided by SERS analysis of biofluids. This study provides a systematic analysis of the molecular information obtained from SERS analysis of serum and urine samples from breast cancer patients and controls, and compares the performance of different machine learning algorithms for sample classification.

Summary: There is a strong interest in developing liquid biopsy assays for detecting cancer-specific alterations in circulating tumor DNA obtained from noninvasive blood draws. This paper describes the development and validation of a multiplex digital methylation-specific PCR platform for the detection of non-small cell lung cancer (NSCLC) in liquid biopsy samples. The platform demonstrates high analytical sensitivity and superior clinical performance compared to traditional MSP assays.

Summary: This study demonstrates the potential of surface-enhanced Raman scattering (SERS) profiling of serum as a liquid biopsy strategy for detecting renal cell carcinoma (RCC). The SERS analysis successfully distinguished RCC patients from controls and suggests the usefulness of SERS as a diagnostic and screening tool for RCC.

Summary: This study suggests that the resistance mechanisms underlying tumor progression in EGFR-positive NSCLC patients under targeted therapies could be driven by the exon location of the original EGFR-sensitizing mutation. Specifically, the presence of the p.T790M EGFR resistance mutation is significantly associated with EGFR exon 19 mutations, and KRAS activating mutations are more common in tumors with EGFR mutations in exons 18 and 21.

Summary: Hepatocarcinogenesis involves complex genetic and cellular dysregulations. Next-generation sequencing technologies provide new insight into the heterogeneity of hepatocellular carcinoma (HCC) and guide the development and use of targeted therapy and immunotherapy. Liquid biopsy coupled with high-sensitivity sequencing could be a practical and safer approach to detect tumor heterogeneity in HCC patients.

Summary: Breast cancer is a major cause of cancer-related morbidity and mortality for women worldwide, and an accurate and rapid diagnosis is crucial for patient treatment and prognosis. Surface-enhanced Raman spectroscopy (SERS) has been widely used in biomedicine due to its vibrational fingerprint information and high detection sensitivity. In this study, an optimized bimetallic nanosphere (Au@Ag NS) 3D substrate was fabricated for the diagnosis of breast cancer based on SERS analysis of extracellular metabolites. The results showed that normal breast cells and breast cancer cells could be readily distinguished based on their SERS spectra, indicating the potential clinical application of this technique in breast cancer diagnosis.

Summary: In this study, the researchers adapted an existing DNA instability test for use with small clinical samples and showed its effectiveness in tumour biopsies taken during colonoscopic investigation. They also demonstrated the potential of using the test with preoperative urine samples to detect tumours in Lynch syndrome patients. This new assay could enable earlier detection and benefit individuals with Lynch syndrome.

Summary: This study aimed to evaluate the accuracy and sensitivity of an enrichment-free method (AccuCyte(R) system) combined with immunofluorescence staining and high-resolution imaging (CyteFinder(R) instrument) in detecting circulating tumor cells (CTCs). The results showed that the AccuCyte-CyteFinder system had high accuracy and sensitivity in detecting rare CTCs, and the recovery rate was significantly improved compared to traditional methods. This method has the potential to be used in the clinical setting for precision-guided cancer treatment.

Summary: Raman spectroscopy is a non-invasive, rapid, and reliable method for cancer detection, and it has recently shown great potential in identifying disease-specific markers. In this study, vibrational spectra of salivary exosomes from oral and oropharyngeal squamous cell carcinoma patients and healthy controls were recorded using surface enhancement Raman spectroscopy (SERS). The method's ability to discriminate between malignant and non-malignant samples was assessed using chemometric analysis, with a discrimination sensitivity of up to 79.3%.
