A new genosensor for meningococcal meningitis diagnosis using biological samples

In this work, a new electrochemical biosensor for DNA detection of bacterial meningitis is proposed. The system is based on specific DNA fragments from the Neisseria meningitidis genome as a probe incorporated on graphite electrodes modified with poly(4-aminophenol). Detection of a complementary oligonucleotide sequence, a specific 710-base pair amplicon, and the genomic DNA of bacteria was carried out by differential pulse voltammetry, using ethidium bromide as an electroactive indicator of hybridization. The complementary oligonucleotide and the genomic DNA of Neisseria meningitidis were quantified by the genosensor, showing detection limits of 0.6 ng mu L-1 and about 6 ng mu L-1, respectively. Morphological differences were observed between hybridized and unhybridized surfaces by atomic force microscopy. The biosensor showed high selectivity, discriminating non-specific targets, and high stability retaining over 98% of its original activity after 120 days of storage. The bioelectrode was effective in discriminating the genomic DNA in samples with human serum without significant interference, proving to be an interesting platform for meningococcal meningitis diagnosis.

Summary: This study reports the construction of a novel nanostructured immunosensor for detection of troponin I biomarker. The immunosensor exhibited a wide work range and low detection limit, as well as good selectivity and specificity in real samples. Furthermore, the immunosensor showed the possibility of reuse and featured a simplified and inexpensive platform, demonstrating excellent potential for application in the early diagnosis of acute myocardial infarction.

Phylogenetic Investigation of Communities by Reconstruction of Unobserved States 2 (PICRUSt2) showed different predominant metabolic pathways between the two groups.

Exploring the mycobiota of bromeliads phytotelmata in Brazilian Campos Rupestres

Summary: The phytotelmata is a water-filled tank on a terrestrial plant and plays a crucial role in bromeliad growth and ecosystem functioning. However, the fungal community in this aquatic ecosystem is still poorly understood. This study used deep sequencing to analyze the fungal communities in the phytotelmata of two bromeliads species in Brazil. The results showed that each bromeliad harbors a distinct fungal community, possibly influenced by the physicochemical characteristics of the phytotelmata and plant morphological features.

Vitamin B12 attenuates leukocyte inflammatory signature in COVID-19 via methyl-dependent changes in epigenetic markings

Summary: COVID-19 induces chromatin remodeling in host immune cells, and vitamin B12 downregulates inflammatory genes through methyl-dependent epigenetic mechanisms. This study evaluated the potential of B12 as an adjuvant drug using blood cultures from COVID-19 patients. B12 normalized the expression of inflammatory genes and increased the sulfur amino acid pathway, which regulates methyl bioavailability. The downregulation of CCL3 by B12 was strongly correlated with hypermethylation of CpGs in its regulatory regions. Transcriptome analysis showed that B12 attenuates the effects of COVID-19 on inflammation-related pathways.

The technology shows great potential for vaccine assessment and monitoring seroprevalence.
