Characterisation of protein-truncating and missense variants in PALB2 in 15 768 women from Malaysia and Singapore

Pei Sze Ng, Rick A. C. M. Boonen, Eldarina Wijaya, Chan Eng Chong, Milan Sharma, Sabine Knaup, Shivaani Mariapun, Weang Kee Ho, Joanna Lim, Sook-Yee Yoon, Nur Aishah Mohd Taib, Mee Hoong See, Jingmei Li, Swee Ho Lim, Ern Yu Tan, Benita Kiat-Tee Tan, Su-Ming Tan, Veronique Kiat-Mien Tan, Rob Martinus van Dam, Kartini Rahmat, Cheng Har Yip, Sara Carvalho, Craig Luccarini, Caroline Baynes, Alison M. Dunning, Antonis Antoniou, Haico van Attikum, Douglas F. Easton, Mikael Hartman, Soo Hwang Teo

Summary: Rare protein-truncating variants (PTVs) in PALB2 confer increased risk of breast cancer in South-East Asian populations, with no association found between rare missense variants and breast cancer risk. Two novel rare missense variants were found to produce unstable proteins, leading to decreased DNA repair function. Despite genetic and lifestyle differences, the prevalence and risk of PALB2 PTVs in this population are similar to those reported in European populations.

Evaluation of Combinations of Nudging, Pricing, and Labeling Strategies to Improve Diet Quality: A Virtual Grocery Store Experiment Employing a Multiphase Optimization Strategy

Soye Shin, Bibhas Chakraborty, Xiaoxi Yan, Rob M. van Dam, Eric A. Finkelstein

Summary: This study tested the effects of different combinations of four intervention strategies on diet quality using a fully functional web-based grocery store. The results indicated that combining food labels, real-time feedback, and ordering strategies was more effective in improving diet quality compared to using only one of these interventions.

Daily park use, physical activity, and psychological stress: A study using smartphone-based ecological momentary assessment amongst a multi-ethnic Asian cohort

Su Hyun Park, Nicholas A. Petrunoff, Nan Xin Wang, Rob M. van Dam, Angelia Sia, Chuen Seng Tan, Falk Mueller-Riemenschneider

Summary: This study found that both daily park use and leisure-time physical activity were associated with reduced stress in the evening, and these relationships were independent of each other. This suggests that each activity on its own is beneficial for reducing stress among adults in urban settings.

Acceptance of healthy lifestyle nudges in the general population of Singapore

Summary: This study examines the public acceptance of lifestyle nudges and identifies that individuals prefer less intrusive approaches for promoting healthy lifestyle. The study also finds that socio-demographic characteristics and lifestyle behaviors are associated with acceptance of nudges, highlighting the importance of taking these factors into consideration when formulating and implementing behaviorally informed health policies.

Dietary patterns and predicted 10-year cardiovascular disease risk in a multiethnic Asian population

Yu Qi Lee, Clare Whitton, Nithya Neelakantan, Rob M. van Dam, Mary Foong-Fong Chong

Summary: This study identified dietary patterns and their associations with cardiovascular disease risk in the Asian population. The 'ethnic breads, legumes and nuts' and 'whole grains, fruit and dairy' patterns were associated with lower risk, while the 'processed food and sugar-sweetened beverages' pattern was associated with higher risk.

Summary: This study evaluates the implementation and effectiveness of a scaled-up physical activity intervention program in Singapore. The results show that the intervention program successfully attracted a large number of participants and effectively increased physical activity levels, despite a decrease in program engagement over time.

Effects of dietary quality, physical activity and weight loss on glucose homeostasis in persons with and without prediabetes in the PREMIER trial

Vlad Gradinariu, Jamy Ard, Rob M. van Dam

Summary: This long-term behavioural trial examined the impact of changes in diet quality, physical activity, and weight loss on improving insulin resistance and fasting glucose concentrations. The study found that behavioural lifestyle interventions can significantly improve glucose metabolism, regardless of whether individuals have prediabetes or not.

Evaluating the Effects of Rewards and Schedule Length on Response Rates to Ecological Momentary Assessment Surveys: Randomized Controlled Trials

Sarah Edney, Claire Marie Goh, Xin Hui Chua, Alicia Low, Janelle Chia, Daphne S. Koek, Karen Cheong, Rob van Dam, Chuen Seng Tan, Falk Mueller-Riemenschneider

Summary: This study aimed to evaluate the effects of study design features on Ecological Momentary Assessments (EMAs) response rates. The results showed that using SMS text messages instead of emails for notifications increased the response rate over time. Changing the available rewards did not lead to a significant difference in the response rate, while increasing the schedule length significantly improved response rates.

Coffee Consumption and Incidence of Cardiovascular and Microvascular Diseases in Never-Smoking Adults with Type 2 Diabetes Mellitus

Yu-Jie Liu, Meng-Yuan Miao, Jia-Min Wang, Quan Tang, Wen-Wen Han, Yi-Ping Jia, Hao-Wei Tao, Yan Zheng, Rob M. van Dam, Li-Qiang Qin, Guo-Chong Chen

Summary: The relationship between coffee consumption and diabetes-related vascular complications is examined in this study. Moderate coffee consumption (2-4 cups/day) is associated with a lower risk of various cardiovascular diseases and chronic kidney disease in individuals with type 2 diabetes who have never smoked.

Acculturation as a Determinant of Obesity and Related Lifestyle Behaviors in a Multi-Ethnic Asian Population

Su Hyun Park, Yu Qi Lee, Falk Mueller-Riemenschneider, Borame Sue Lee Dickens, Rob M. van Dam

Summary: Limited attention has been given to the role of cultural orientation towards different ethnic groups in multi-ethnic settings without a dominant host culture. In this study, we found that acculturation levels were associated with obesity and related lifestyle behaviors. Higher acculturation levels were associated with higher obesity prevalence among Chinese, inverse association among ethnic Indians, and no significant association among Malays. In Malays, greater acculturation was associated with higher dietary quality and less sedentary time. High acculturation levels were also associated with higher sugar-sweetened beverage consumption and more leisure-time physical activity across all ethnic groups.

Differences in type 2 diabetes risk between East, South, and Southeast Asians living in Singapore: the multi-ethnic cohort

Jowy Yi Hoong Seah, Xueling Sim, Chin Meng Khoo, E. Shyong Tai, Rob M. van Dam

Summary: This study compared the risk of type 2 diabetes (T2D) among Chinese, Indian, and Malay ethnic groups and examined the biological factors that contribute to ethnic differences. The results showed that Malays and Indians had approximately twice the risk of T2D compared to Chinese. Higher BMI explained the higher risk for Malays, and factors such as BMI, waist circumference, inflammation, insulin resistance, and high-density lipoprotein cholesterol contributed to the higher risk for Indians. However, part of the higher T2D risk associated with Indian ethnicity remained unexplained. Chinese participants had the lowest adiponectin levels despite their lower diabetes risk.
