A Tolerogenic Artificial APC Durably Ameliorates Experimental Autoimmune Encephalomyelitis by Directly and Selectively Modulating Myelin Peptide-Autoreactive CD4+ and CD8+ T Cells

In this study, a tolerogenic artificial APC (TaAPC) was developed to directly and selectively modulate myelin-autoreactive CD4(+) and CD8(+) T cells in the myelin oligodendrocyte glycoprotein (MOG)(35-55) peptide-induced experimental autoimmune encephalomyelitis in C57BL/6J mice. Cell-sized polylactic-coglycolic acid microparticles were generated to cocouple target Ags (MOG(40-54)/H-2D(b)-Ig dimer, MOG(35-55)/I-A(b) multimer), regulatory molecules (anti-Fas and PD-L1-Fc), and self-marker CD47-Fc and encapsulate inhibitory cytokine (TGF-beta 1). Four infusions of the TaAPCs markedly and durably inhibited the experimental autoimmune encephalomyelitis progression and reduced the local inflammation in CNS tissue. They circulated throughout vasculature into peripheral lymphoid tissues and various organs, but not into brain, with retention of 36 h and exerted direct effects on T cells in vivo and in vitro. Two infusions of the TaAPCs depleted 65-79% of MOG(35-55)-specific CD4(+) and 46-62% of MOG(40-54)-specific CD8(+) T cells in peripheral blood, spleen, and CNS tissues in an Ag-specific manner and regulatory molecule-dependent fashion; induced robust T cell apoptosis; inhibited the activation and proliferation of MOG peptide-reactive T cells; reduced MOG peptide-reactive Th1, Th17, and Tc17 cells; and expanded regulatory T cells. They also inhibited IFN-g/IL-17A secretion and elevated IL-10/TGF-beta 1 production in splenocytes but not in CNS tissue. More importantly, the TaAPCs treatment did not obviously suppress the overall immune function of host. To our knowledge, this study provides the first experimental evidence for the capability of TaAPCs to directly modulate autoreactive T cells by surface presentation of multiple ligands and paracrine release of cytokine, thus suggesting a novel Ag-specific immunotherapy for the T cell-mediated autoimmune diseases.

Collective influence of substrate chemistry with physiological fluid shear stress on human umbilical vein endothelial cells

Summary: The treatment of cardiovascular diseases relies heavily on vascular scaffold materials to ensure normal physiological activity of endothelial cells. This study investigated the combined effects of substrate chemistries and fluid shear stress on HUVECs, showing that FSS enhances the production of key substances while the release of ATP, NO, and PGI(2) is influenced by both substrate chemistries and FSS. The study also suggests that F-actin organization and focal adhesions of HUVECs are controlled by substrate chemistries, which in turn affects the responses triggered by FSS.

Identification of HLA-A2 restricted epitopes of glypican-3 and induction of CTL responses in HLA-A2 transgenic mice

Summary: In this study, 25 HLA-A0201 restricted GPC3 epitopes were predicted and 5 were identified as real-world epitopes for specific CD8(+) T cell activation. These epitopes were further confirmed to induce robust CD8(+) T cell responses and cytotoxicity targeting HepG2 cells, providing potential for the development of T cell epitope vaccines and evaluation of GPC3 specific T cell responses.

Summary: The study constructed RBD-Fc proteins that can bind to specific mutant virus strains and demonstrated in mouse immunization experiments that these proteins can induce high levels of IgG antibodies and cross-neutralizing antibodies. Furthermore, RBD-Fc proteins with multiple amino acid substitutions showed higher antibody titers and neutralizing-antibody titers compared to those with single substitutions.

A Systematic Review of T Cell Epitopes Defined from the Proteome of Hepatitis B Virus

Summary: Hepatitis B virus infection is a global health issue with no eradicative therapy available. The lack of validated T cell epitopes hampers the development of vaccines and T cell therapies, with most epitopes restricted to common HLA supertypes. More research is needed to map a comprehensive repertoire of T cell epitopes covering the HBV proteome and global populations.

The Use of Molecular Dynamics Simulation Method to Quantitatively Evaluate the Affinity between HBV Antigen T Cell Epitope Peptides and HLA-A Molecules

Summary: This study proposes a novel prediction strategy based on structure to assess the affinity between HBV antigenic peptides and HLA molecules. Through residue scanning, peptide docking, and molecular dynamics methods, the researchers obtained the molecular docking model of HBV peptide and HLA, and calculated the binding affinity using the MM-GBSA method. The results were consistent with experimental assays, indicating the accuracy of the affinity prediction process and the reliability of the HLA homologous modeling strategy.

A systemic review of T-cell epitopes defined from the proteome of SARS-CoV-2

Summary: This review provides a comprehensive map of T-cell epitopes from SARS-CoV-2 antigens, the HLA supertypes that present these epitopes, and the strategies to screen and identify T-cell epitopes. A total of 1349 CD8+ T-cell epitopes and 790 CD4+ T-cell epitopes have been defined, mostly presented by common HLA supertypes. However, the currently defined T-cell epitope repertoire does not cover the HLA polymorphism of major populations in specific geographic regions.
