Summary: Single-cell transcriptome sequencing provides higher resolution for cellular differences in papillary thyroid carcinoma. Through integrative analysis of two datasets, we identified 31 consistently differentially expressed genes across multiple cell types, which are related to adaptive immune response and autoimmune thyroid diseases. Experimental validation suggests that KRT7 may be a potential biomarker for PTC epithelial cells.

Summary: This study analyzed the dysregulated profile of circular RNAs (circRNAs) in papillary thyroid cancer (PTC) tissues and found that circAGTPBP1 was upregulated and correlated with tumor characteristics. The downstream targets of circAGTPBP1 were identified as miR-34a-5p and NOTCH1. Functional experiments showed that circAGTPBP1 knockdown inhibited PTC cell migration, invasion, and metastasis, and miR-34a-5p inhibitor reversed these effects. CircAGTPBP1 knockdown also inhibited tumor growth in vivo, suggesting its potential as a therapeutic target for PTC.

Validation in a separate dataset showed that intravenous immunoglobulin treatment alleviated the deregulated RNA profiles in KD patients.

Summary: In this study, circRNA high-throughput sequencing was used to identify alterations in circRNA expression levels in PTC tissues. circTIAM1 was found to be upregulated in PTC and associated with apoptosis, proliferation, and migration of PTC cells. Mechanistically, circTIAM1 acted as a sponge of microRNA-646 and functioned in PTC by targeting miR-646 and heterogeneous ribonucleoprotein A1. These findings highlight the oncogenic role of circTIAM1 in PTC and suggest its potential as a therapeutic target for PTC progression.

Summary: After the metabolic syndrome, thyroid disorders are the most common endocrine disorders with increasing prevalence. They can be distinguished by hyperthyroidism, hypothyroidism, or thyroiditis, as well as the presence of benign or malignant thyroid nodules. Current methods for detecting thyroid cancer have limitations, and with the advancement of molecular biology techniques, new biomarkers such as mRNA and non-coding RNAs have the potential to improve diagnosis and prognosis.
