Loss of EGFR confers acquired resistance to AZD9291 in an EGFR-mutant non-small cell lung cancer cell line with an epithelial-mesenchymal transition phenotype

AZD9291 is an irreversible, small-molecule inhibitor which has potency against mutant EGFR- and T790M-resistant mutation. Despite the encouraging efficacy in clinical, the acquired resistance will finally occur. Further study will need to be done to identify the acquired resistance mechanisms and determine the next treatment. We established an AZD9291-resistant cell line (HCC827/AZDR) from parental HCC827 cell line through stepwise pulsed selection of AZD9291. The expression of EGFR and its downstream pathways were determined by western blot analysis or immunofluorescence assay. The sensitivity to indicated agents were evaluated by MTS. Compared with parental HCC827 cells, the HCC827/AZDR cells showed high resistance to AZD9291 and other EGFR-TKIs, and exhibited a mesenchymal-like phenotype. Almost complete loss of EGFR expression was observed in HCC827/AZDR cells. But the activation of downstream pathway, MAPK signaling, was found in HCC827/AZDR cells even in the presence of AZD9291. Inhibition of MAPK signaling had no effect on cell viability of HCC827/AZDR and could not reverse AZD9291 resistance because of the subsequent activation of AKT signaling. When treated with the combination of AKT and MAPK inhibitor, HCC827/AZDR showed remarkable growth inhibition. Loss of EGFR could be proposed as a potential acquired resistance mechanism of AZD9291 in EGFR-mutant NSCLC cells with an EMT phenotype. Despite the loss of EGFR, the activation of MAPK pathway which had crosstalk with AKT pathway could maintain the proliferation and survival of resistant cells. Blocking MAPK and AKT signaling may be a potential therapeutic strategy following AZD9291 resistance.

Summary: This study aimed to establish a scoring model based on early brain injury indicators to predict the functional outcomes of patients with aneurysmal subarachnoid hemorrhage (aSAH) at 90 days. The model, named TAPS, showed good discrimination and calibration in both the derivation and validation cohorts, and proved to be an easy handle tool for clinicians to predict the 90-day outcomes and identify patients at risk for poor prognosis.

The Added Value of Second-Look Ultrasound in Patients With Primary Hyperparathyroidism With Discordant or Negative Previous Imaging Findings

Summary: The study aimed to evaluate the value of second-look ultrasound (US) for preoperative localization in patients with primary hyperparathyroidism with negative or discordant imaging results. The sensitivity, positive predictive value, and accuracy of second-look US were found to be 92.1%, 95.1%, and 88.1%, respectively. Overall, second-look US is a promising method for preoperative localization, especially for patients with negative or discordant imaging findings.

Summary: This study assessed the performances of droplet digital polymerase chain reaction (ddPCR) and next-generation sequencing (NGS) in detecting EGFR T790M mutation in the plasma samples of NSCLC patients. The results showed that ddPCR had a high sensitivity and specificity in detecting EGFR T790M mutation. Furthermore, a higher EGFR T790M abundance was associated with a better response to third-generation EGFR-TKIs.

Summary: Determination of drug binding kinetics in plasma is challenging, and this study introduces a new parameter called dynamic free fraction to describe drug-protein binding kinetics. The dynamic free fraction can be determined by coupling the drug binding assay with a reporter enzyme and high-resolution mass spectrometry. This methodology bypasses the challenges in existing methods and allows assessment of the impact of protein binding kinetics on drug properties.

Bispecific anti-CD3xanti-CD155 antibody mediates T-cell immunotherapy in human haematologic malignancies

Summary: T cells play a crucial role in the cell-mediated antitumour response. Bispecific antibodies (Bi-Abs) have shown promise in recruiting T cells for killing tumours. This study demonstrates the expression of CD155 in a wide range of human haematologic tumours and highlights the effectiveness of the bispecific antibody anti-CD3 x anti-CD155 in activating T cells to target malignant haematologic cells. The results show that CD155Bi-Ab-armed T cells exhibit enhanced cytotoxicity and increased secretion of cell-killing mediators and T-cell-derived cytokines, indicating that CD155 could be a novel target for immunotherapy against haematologic malignancies.

Single-cell transcriptomics in ovarian cancer identify a metastasis-associated cell cluster overexpressed RAB13

Summary: By integrating single-cell sequencing data, researchers discovered a cell subcluster closely associated with metastasis in ovarian cancer and identified RAB13 as its key marker. Experimental results showed that downregulation of RAB13 expression can reduce migration and invasion of ovarian cancer cells, and several potential drugs that may inhibit RAB13 were predicted. This study provides new insights and potential therapeutic targets for the research on ovarian cancer metastasis.

Summary: This study compared the long-term outcome of hemorrhagic stroke or death in patients with conservative management or stand-alone embolization for AVM. The results showed that embolization did not prevent long-term hemorrhagic stroke or death compared with conservative management in the overall cohort, as well as in the unruptured and ruptured AVM subgroups. Stratified analysis suggested that target embolization might be beneficial for unruptured AVMs, while curative embolization improved the outcomes of ruptured AVMs. The long-term neurological status was similar between the two strategies.

Optic Fiber Microsensor Reveals Specific Spatiotemporal Oxygen Uptake Profiles at the Mammalian Ocular Surface

Summary: The uptake of oxygen by cells and tissues is an important indicator of metabolic demand, microenvironment changes, and pathophysiology. Using a non-invasive optical fiber oxygen sensor, researchers have revealed a spatiotemporal profile of oxygen uptake at the ocular surface of rodents and non-human primates. The results showed a distinct oxygen uptake gradient in mice, with a higher influx at the limbus and conjunctiva regions compared to the center of the cornea. The findings provide valuable insights for comparative studies and understanding the response of the cornea and other tissues to various insults or environmental changes.

Rupture-related quantitative hemodynamics of the supratentorial arteriovenous malformation nidus

The results showed that a higher stasis index and slower transnidal relative velocity (TRV) were significantly correlated with AVM rupture and could be used as effective indicators to predict future hemorrhage risk.
