Summary: We investigated the roles of Pat1 and Dhh1 in regulating mRNA translation and abundance in yeast cells under nutrient-replete conditions. Our findings suggest that Pat1 and Dhh1 cooperatively repress the expression of numerous non-ESR transcripts by reducing decapping and transcription in wild-type cells. Furthermore, we observed impaired mRNA turnover in mutants lacking Dhh1 or Pat1, leading to derepression of these transcripts. Pat1 and Dhh1 also contribute to translational repression and decreased protein production for multiple pathways involved in cell adhesion, nitrogen metabolism, respiration, and autophagy. These findings highlight the post-transcriptional regulatory functions of Pat1 and Dhh1 during nutrient limitation.

Summary: Conditional degron technologies, such as the auxin-inducible degron (AID) system, are important tools for biological research. In this study, we improved the AID system for the fission yeast Schizosaccharomyces pombe by optimizing all three components: the AID degron, the small-molecule inducer, and the inducer-responsive F-box protein. Our improved AID system showed inducible lethality for 10 essential genes, including ones that could not be effectively inactivated using a previously published AID system.

TERRA is regulated by the telomere-binding proteins POT-1 and POT-2, and its transcripts are heterogeneous in length and form discrete nuclear foci. In germ cells, TERRA is expressed in all stages of meiotic prophase I, and it is upregulated prior to activation of the alternative lengthening mechanism (ALT).

Summary: Cells adjust the abundance of surface nutrient transporters and receptors in response to changing nutrient environments by modulating ubiquitin-dependent endocytosis. The NEDD4 family of E3 ligases plays a role in regulating this process. In this study, the researchers discovered a novel regulation of Pub1, a member of the NEDD4-family of E3 ligases in fission yeast. They found that nitrogen stress inhibits Pub1 function, leading to an increase in the abundance of the amino acid transporter Aat1 at the plasma membrane and increased sensitivity to the toxic arginine analogue canavanine. They also found that TORC2 signalling negatively regulates Pub1, with TORC2 mutants under nutrient stress having decreased Aat1 levels and resistance to canavanine. Furthermore, they found that inhibition of TORC2 signalling increases phosphorylation of Pub1, dependent on Gsk3 activity. Overall, this study demonstrates how the TORC2 nutrient-sensing signalling network regulates Pub1 to modulate the abundance of nutrient transporters.
