Summary: Cancer is a leading cause of death globally, with invasion and metastasis being directly responsible. Current therapeutic approaches for cancer suffer from toxicity and chemoresistance, highlighting the need for alternative treatment options. Natural products, particularly flavonoids, have shown promise as they possess antitumor properties in various study models. This review discusses the regulation of tissue inhibitors of metalloproteinases (TIMPs) expression and the role of flavonoids as therapeutic molecules that target TIMPs. Flavonoids exhibit chemopreventive and cytotoxic effects against different types of cancer through various mechanisms and also regulate crucial signaling pathways involved in cancer cell migration, invasion, and metastasis.

Understanding the multiple functions of CAFs and developing intervention strategies are important for improving breast cancer therapy. Further research is needed to identify specific subsets of CAFs that can be targeted for therapy.

Summary: This study reveals that in papillary thyroid carcinoma (PTC), tumor-associated macrophages (TAMs) may influence lymphatic invasion of cancer cells by damaging lymphatic vessels together with cancer cells. An increase in TAMs was observed in PTC, and a significant correlation between M2 cells inside and outside the lymphatic vessels was found.

Matrix remodeling in the TME, particularly by matrix metalloproteinases (MMPs), plays a crucial role in modifying ECM barriers and releasing soluble ECM fragments that impact cells both within and outside the TME.

Summary: In this study, it was found that EAA, isolated and purified from coral symbiotic fungi, could inhibit tumor metastasis and invasion. Through cell experiments and protein analysis, it was discovered that EAA reduced the activity of MMPs and VEGF by regulating the phosphorylation of downstream MAPK, PI3K/AKT, and NF-kappa B pathways, thereby inhibiting the migration and invasion of tumor cells.

Metastasis mainly occurs in the bone, and current treatment methods focus on palliative care and slowing down the progression. Recent advances in molecularly targeted agents have made breakthroughs in the treatment of metastatic breast cancer, but immunotherapies have not been fully applied in estrogen receptor-positive tumors. Therapeutic approaches targeting the bone microenvironment of metastatic breast cancer have potential applications in other bone-resident cancers.

Summary: Glioblastoma stem cells (GSCs) in the aggressive brain cancer Glioblastoma multiforme (GBM) evade immune responses by impairing the function of tumor-infiltrating NK cells through a direct cell-to-cell contact mechanism mediated by alpha v integrin and TGF-β activation. Treatment with inhibitors of integrin or TGF-β signaling, or with TGFBR2 gene-edited allogeneic NK cells, prevents GSC-induced NK cell dysfunction and tumor growth, suggesting a potential therapeutic strategy for GBM.
