Summary: HLA-G plays a central role in immune tolerance at the maternal-fetal interface and its polymorphism is associated with sHLA-G expression and the risk of recurrent implantation failure (RIF). However, the association of HLA-G polymorphisms with RIF in the general population is unclear and may vary among different ethnic backgrounds. Future studies should consider a combined genetic effect to confirm the association of HLA-G polymorphisms and sHLA-G expressions in relation to RIF.

Summary: Male factors play an important role in infertility of couples. HLA-G gene polymorphisms in seminal plasma may influence the secretion of soluble HLA-G protein in semen and affect male fertility. The G-C-ins haplotype may reduce sperm parameters.

Summary: HLA-G plays an important role in tumor prognosis. The 14 nt insert of HLA-G is more frequent in glioma patients, and elevated plasma levels of sHLA-G are negatively associated with their survival.

Summary: This meta-analysis found no association between HLA-G 14-bp I/D and digestive cancers. However, in HBV/HCV positive hepato-cellular cancer patients, a significant association of HLA-G 14-bp I/D with disease initiation was reported. Overall analysis showed significantly increased sHLA-G in patients, and in Asian gastric cancer patients, sHLA-G was significantly increased in higher grade tumors.

Summary: The meta-analysis shows that HLA-G plays a significant role in digestive cancers. The HLA-G 14-bp I/D polymorphism is associated with HCC in patients with concomitant HBV/HCV viral infections. Increased sHLA-G indicates a poor prognosis for digestive cancer patients.

Summary: This study found that HLA-G 3'UTR polymorphisms may influence the peripheral sHLA-G plasma level in patients and play a key role in the susceptibility and prognosis of cervical cancer. sHLA-G may represent a novel plasma biomarker for the prognosis and potential therapeutic target of cervical cancer.

Summary: This study found that complications of pregnancy caused by immune inflammation, such as preeclampsia and preterm labor, can lead to altered expression profiles of nonclassical human leukocyte antigens. The level of human leukocyte antigen G increases in pregnancies complicated by severe early-onset preeclampsia with small-for-gestational-age neonates.

Summary: The study found that in cases of preeclampsia, there were changes in the expression of HLA-G, but no significant differences in genotypes or isoforms related to preeclampsia were found. Increased expression of the HLA-G1 14 bp insertion allele in the placenta was associated with changes in other HLA-G isoforms and reduced levels of placental sHLA-G.

Summary: The study revealed that pregnant women with PE had increased gravidity, higher BMI before delivery, reduced neonatal weight, gestational weeks, and placental weight, as well as a higher proportion of cesarean section. Significant differences in SNPs were found at locus 29794467, 29796376, and 29799440 between the PE group and the control group. In locus 29799440, patients with genotype CC had a lower risk of PE compared to patients with genotype TT, while there was no significant difference in PE risk between patients with genotype CT and TT.

Summary: Polymorphisms in HLA-G and sHLA-G expression are associated with gynecological cancers, particularly HPV-associated cervical cancer. Additionally, soluble HLA-G levels are significantly increased in cancer patients. These findings have important implications for the diagnosis and immunotherapy of gynecological cancers.

Summary: HLA-G, a non-classic MHC class I molecule, is widely expressed in tumor environment where it plays a key role in immune escape, potentially important for cancer diagnosis and prognosis. However, due to its multiple subtypes, evaluating HLA-G expression in tumors is challenging.

Summary: Gastric cancer (GC) is a commonly occurring malignancy with high mortality due to late diagnosis. Overexpression of the HLA-G molecule in cancer is associated with immune evasion, metastasis, poor prognosis, and lower overall survival. Our study evaluated the plasma levels of soluble HLA-G in patients with GC and benign gastric pathologies, finding higher sHLA-G concentration in GC patients compared to benign pathologies, higher levels in women with GC than men, and significant differences between the benign gastric pathologies assessed. Although sHLA-G is not an adequate marker for staging GC, it has diagnostic potential. Additionally, our study revealed the presence of HLA-G molecules in exosomes, highlighting the need for further research to explore their functional implications and potential non-invasive diagnostic applications.

Summary: The study evaluated the levels of plasma soluble HLA-G (sHLA-G) in COVID-19 patients and healthy controls. The results showed increased levels of sHLA-G in COVID-19 patients, which correlated with disease severity. However, the levels decreased in critically ill patients, possibly indicating immune exhaustion. sHLA-G also exhibited positive correlation with other disease mediators IL-6, IL-8, and IL-10.
