Tracking neural crest cell cycle progression in vivo

Analysis of cell cycle entry/exit and progression can provide fundamental insights into stem cell propagation, maintenance, and differentiation. The neural crest is a unique stem cell population in vertebrate embryos that undergoes long-distance collective migration and differentiation into a wide variety of derivatives. Using traditional techniques such as immunohistochemistry to track cell cycle changes in such a dynamic population is challenging, as static time points provide an incomplete spatiotemporal picture. In contrast, the fluorescent, ubiquitination-based cell cycle indicator (Fucci) system provides in vivo readouts of cell cycle progression and has been previously adapted for use in zebrafish. The most commonly used Fucci systems are ubiquitously expressed, making tracking of a specific cell population challenging. Therefore, we generated a transgenic zebrafish line, Tg(-4.9sox10:mAG-gmnn(1/100)-2A-mCherry-cdt1(1/190)), in which the Fucci system is specifically expressed in delaminating and migrating neural crest cells. Here, we demonstrate validation of this new tool and its use in live high-resolution tracking of cell cycle progression in the neural crest and derivative populations.

Summary: Utilizing single-cell transcriptomic analyses, this study delineated NCC-derivatives in the zebrafish, identifying major cell types and over three dozen cellular subtypes. Gene expression signatures of pigment progenitors, mesenchyme cells, and enteric NCCs were dissected, revealing combinatorial hox gene codes demarcating NCC derivatives, with distinct profiles in neuronal cells. The comprehensive cell-type atlas presented in this study can serve as a valuable resource for further investigations into NCC differentiation mechanisms and evolution.

Summary: Neural crest cells (NCCs) are migratory and multipotent stem cells essential to vertebrate embryonic development. This study identifies the gene Hoxb5b as a regulator of NCC development, demonstrating that elevated Hoxb5b levels promote expansion of zebrafish NCCs and specifically expand expression domains of vagal NCC markers. The study also shows that early increase in vagal NCCs leads to supernumerary enteric neural progenitors, but these fail to properly expand into enterically fated neurons in the gut tissue.

Expanding the eukaryotic genetic code with a biosynthesized 21st amino acid

Summary: Genetic code expansion technology allows the creation of semisynthetic organisms using noncanonical amino acids. However, high concentrations of exogenously fed ncAAs are required due to inefficient cellular uptake and incorporation, especially in eukaryotic cells. To overcome this, researchers have engineered a metabolic pathway for the synthesis of O-methyltyrosine (OMeY) that allows cells to autonomously biosynthesize and incorporate OMeY into proteins at higher levels than exogenously fed cells. This technology has been successfully extended to mammalian cells and zebrafish model, providing valuable models for biological and biomedical research.
