Vasomotor dysfunction in human subcutaneous arteries exposed ex vivo to food-grade titanium dioxide

Animal studies have shown that titanium dioxide (TiO2) exposure affects arterial vasomotor function, whereas little is known about the effects in arteries from humans. This study investigated vasomotor responses after direct exposure of human subcutaneous arteries to food-grade TiO2 (E171) (14 or 140 mu g/ml) for 30 min and 18 h. Vasomotor responses to bradykinin, 5-hydroxytryptamine (5-HT), sarafotoxin 6c (S6c) and nitroglycerin were recorded in wire-myographs. Vasoconstrictor responses to 5-HT were increased in arteries exposed to E171 for 18 h (P < 0.05). Furthermore, an increase in S6c responses was seen in low concentration E171 exposed arteries (30 min exposure; P < 0.05). The vasorelaxation response to nitroglycerin was increased in low concentration E171 exposed arteries (30 min exposure; P < 0.05). Vasorelaxation responses to bradykinin were unaffected after treatment with E171. There was no difference in gene expression levels of intercellular cell adhesion molecule 1, vascular cell adhesion molecule 1, 5-hydroxytryptamine receptor 1B, 5-hydroxytryptamine receptor 2A, endothelin receptor A and endothelin receptor B in E171 exposed arteries after exposure to TiO2 for 30 min or 18 h. In conclusion, this study shows that the same type of vasomotor dysfunction is found in artery segments of rats and humans following ex vivo exposure to E171.

Genotoxicity by rapeseed methyl ester and hydrogenated vegetable oil combustion exhaust products in lung epithelial (A549) cells

Summary: Biofuel is an attractive alternative to petrodiesel due to its lower environmental impact. This study compared the genotoxicity of exhaust particles from the combustion of petrodiesel, rapeseed methyl ester (RME), and hydrogenated vegetable oil (HVO) in lung epithelial cells. The results showed that petrodiesel and RME generated the same level of DNA strand breaks based on equal concentrations of total polycyclic aromatic hydrocarbons (PAH), but RME had lower PAH emissions per fuel energy content, resulting in lower genotoxic risk compared to petrodiesel for on-road vehicle emissions.

Summary: Measurement of DNA migration in the comet assay can be done through image analysis or visual scoring, with the latter accounting for a significant portion of the published results. This study examines the variability in visual scoring of comets among different investigators and suggests the use of three reference sets for scoring. The results show both inter-investigator and intra-investigator variation in scoring, indicating a need for further standardization, but also demonstrate that visual scoring remains a reliable method for analyzing DNA migration in comets.

Factors Affecting the Vitamin C Dose-Concentration Relationship: Implications for Global Vitamin C Dietary Recommendations

Summary: In this study, the relationship between vitamin C status and demographic and lifestyle factors was investigated using NHANES 2017-2018 datasets. It was found that gender, smoking, and body weight have significant impacts on vitamin C requirements. Males and smokers require higher vitamin C intakes to achieve adequate serum concentrations. Overall, the general population should consume around 110 mg/day of vitamin C, smokers require approximately 165 mg/day, and heavier individuals (100+ kg) require about 155 mg/day to reach comparable vitamin C concentrations.

Effect of Remote Ischaemic Preconditioning on Perioperative Endothelial Dysfunction in Non-Cardiac Surgery: A Randomised Clinical Trial

Summary: Endothelial dysfunction occurs due to inflammation and excessive reactive oxygen species production during surgical stress. Remote ischemic preconditioning (RIPC) may have anti-oxidative and anti-inflammatory properties, but it did not affect endothelial function or markers of nitric oxide (NO) bioavailability until 24 hours after sub-acute laparoscopic cholecystectomy. Surgical removal of the inflamed gallbladder reduced markers of NO bioavailability and oxidative stress caused by acute cholecystitis.

Summary: The study finds that potassium bromate is a suitable positive control for the Fpg-modified comet assay, which is widely used to measure oxidatively generated damage to DNA. Although all laboratories obtained positive concentration-response relationships, there was variation in the levels of Fpg-sensitive sites between different laboratories. However, the consistency between the measurements of %Tail DNA values and visual scores of comet images done in the central laboratory suggests a potential source of inter-laboratory variation.

Summary: This study investigated the effects of emissions from cooking and candle burning on inflammatory changes in individuals with mild asthma. The results showed that cooking emissions significantly increased levels of oxidatively damaged DNA and lipids in the blood, while candle emissions had a minimal impact on the respiratory system.
