Depression, a multifactorial brain disorder, is one of the most prevalent diseases worldwide. Several strategies have been developed to counteract the main symptoms and disorders. However, the treatments are usually associated with different side effects or poor effect. For that reason, new necessary approaches are emerging; among them, natural products are good alternatives since no interactions have been described up to now. Flavonoids have been related to antidepressant effects in cell lines and animal models by their action on the amine mechanisms protecting the neuroendocrine and immune systems. The current review includes an approach of some of the main results related to the action of flavonoids on depression found in the literature and a short view of the possible mechanisms involved. Thus, it highlights the potential emerging candidates with strong antidepressant effects which could be effective new compounds.

Highly potent and selective 5-lipoxygenase inhibition by new, simple heteroaryl-substituted catechols for treatment of inflammation

Verena Krauth, Ferdinando Bruno, Simona Pace, Paul M. Jordan, Veronika Temml, Maria Preziosa Romano, Haroon Khan, Daniela Schuster, Antonietta Rossi, Rosanna Filosa, Oliver Werz

Summary: 5-Lipoxygenase (LO) is a key enzyme in the formation of pro-inflammatory leukotrienes (LT) that are important mediators in allergic reactions and inflammatory disorders. In this study, modifications were made to catechol derivatives to increase their effectiveness as selective 5-LO inhibitors. Two compounds, 3,4-dihydroxy-3 '-phenoxybiphenyl (6b) and 2-(3,4-dihydroxyphenyl)benzo[b]thiophene (6d), showed potent inhibition of human 5-LO in both cell-free and cell-based assays. Furthermore, these compounds were effective in inhibiting 5-LO in human blood and in a murine peritonitis model. These novel catechol derivatives have high selectivity and effectiveness as inhibitors of 5-LO under pathophysiological conditions.

Sajad Fakhri, Ebrahim Darvish, Fatemeh Narimani, Seyed Zachariah Moradi, Fatemeh Abbaszadeh, Haroon Khan

Summary: Neurodegenerative diseases are increasing globally, making it crucial to uncover the precise pathophysiological mechanisms and develop novel therapeutic strategies. Non-coding RNAs play a pivotal role in these diseases through various mechanisms, such as gene expression modulation, protein translation interference, and alterations in signaling pathways. The study highlights the role of different ncRNAs and investigates how phytochemicals can modulate ncRNAs in neurodegenerative diseases like Alzheimer's disease, Parkinson's disease, epilepsy, depression, and amyotrophic lateral sclerosis.

Oxidative stress and behavioral responses of moorish geckos (Tarentola mauritanica) submitted to the presence of an introduced potential predator (Hemorrhois hippocrepis)

Maria Febrer-Serra, Nil Lassnig, Victor Colomar, Gabriela Pico, Silvia Tejada, Antoni Sureda, Samuel Pinya

Summary: Stressful situations lead to an increase in reactive oxygen species, while the introduction of new potential predators induces physiological stress in native fauna. This study found that the moorish gecko exhibited behavioral responses and changes in oxidative stress biomarkers when exposed to the invasive predator horseshoe whip snake, indicating its ability to recognize and adapt to the snake.

Effect of alkalinity and light intensity on the growth of the freshwater sponge Ephydatia fluviatilis (Porifera: Spongillidae)

Marc Gost, Samuel Pinya, Antoni Sureda, Silvia Tejada, Pere Ferriol

Summary: The adaptation of sponges to freshwater environments was a major event in their evolutionary history. This study revealed that the growth capacity of the freshwater sponge Ephydatia fluviatilis is positively associated with high alkalinity values and affected by light exposure. These findings explain the greater distribution of this species in alkaline environments and emphasize the importance of symbiosis in E. fluviatilis.

Summary: Cancer is a leading cause of death worldwide, and its development is linked to oxidative stress. The transcription factor Nrf2 plays a vital role in protecting cells against oxidative stress in cancer growth, progression, and therapy resistance. Therefore, innovative inhibitors targeting Nrf2 are being investigated. This comprehensive review explores the potential of phenolic compounds, found in traditional plant-based remedies, as effective anticancer agents by inhibiting Nrf2. The clinical applications of naturally-occurring bioactive compounds as Nrf2 inhibitors, specifically polyphenols, are also discussed.

Polyunsaturated and Saturated Oxylipin Plasma Levels Allow Monitoring the Non-Alcoholic Fatty Liver Disease Progression to Severe Stages

Miguel D. Ferrer, Clara Reynes, Margalida Monserrat-Mesquida, Magdalena Quetglas-Llabres, Cristina Bouzas, Silvia Garcia, David Mateos, Miguel Casares, Cristina Gomez, Lucia Ugarriza, Josep A. Tur, Antoni Sureda, Antoni Pons

Summary: Non-alcoholic fatty liver disease (NAFLD) is characterized by the accumulation of hepatic fat. This study investigated the plasma levels of oxylipins, free polyunsaturated fatty acids (PUFA), and markers of lipid peroxidation in NAFLD patients. The results showed that severe NAFLD patients had higher levels of oxylipins, saturated oxylipins, and free PUFA, suggesting an association between inflammation, fatty acid oxidation dysregulation, and NAFLD progression.

Mediterranean Diet Improves Plasma Biomarkers Related to Oxidative Stress and Inflammatory Process in Patients with Non-Alcoholic Fatty Liver Disease

Maria Magdalena Quetglas-Llabres, Margalida Monserrat-Mesquida, Cristina Bouzas, Isabel Llompart, David Mateos, Miguel Casares, Lucia Ugarriza, J. Alfredo Martinez, Josep A. Tur, Antoni Sureda

Summary: Non-alcoholic fatty liver disease (NAFLD) is characterized by liver fat deposition without alcohol consumption. This study aimed to assess the antioxidant and pro-inflammatory state in NAFLD patients following a 12-month lifestyle intervention based on adherence to a Mediterranean diet (AMD). The intervention resulted in improvements in anthropometric and biochemical parameters, with higher reductions in alanine aminotransferase (ALT) and C reactive protein (CRP) observed in participants with high AMD. Additionally, there were decreases in prooxidant and proinflammatory markers, as well as improvements in physical fitness and intrahepatic fat contents. These improvements were more significant in individuals with higher adherence to the Mediterranean diet.
