Biotechnology applied to fish reproduction: tools for conservation

This review discusses the new biotechnological tools that are arising and promising for conservation and enhancement of fish production, mainly regarding the endangered and the most economically important species. Two main techniques, in particular, are available to avoid extinction of endangered fish species and to improve the production of commercial species. Germ cell transplantation technology includes a number of approaches that have been studied, such as the transplantation of embryo-to-embryo blastomere, embryo-to-embryo differentiated PGC, larvae to larvae and embryo differentiated PGC, transplantation of spermatogonia from adult to larvae or between adults, and oogonia transplantation. However, the success of germ cell transplantation relies on the prior sterilization of fish, which can be performed at different stages of fish species development by means of several protocols that have been tested in order to achieve the best approach to produce a sterile fish. Among them, fish hybridization and triploidization, germline gene knockdown, hyperthermia, and chemical treatment deserve attention based on important results achieved thus far. This review currently used technologies and knowledge about surrogate technology and fish sterilization, discussing the stronger and the weaker points of each approach.

Reproductive biology of the Amazonian amphibian fish the splash tetra Copella arnoldi with emphasis to histological characterization

Summary: This study describes important components of the reproductive biology of the Amazonian amphibian fish Copella arnoldi, including sex ratio, length-weight relationship, and gametogenesis. The results show that the species is capable of spawning throughout the year, with only certain individuals capable of spawning during different seasons. Additionally, this species exhibits a rare type of semicystic spermatogenesis, which is described for the first time in the Lebiasinidae family.

Transcriptome and Proteome Analyses Reveal Stage-Specific DNA Damage Response in Embryos of Sturgeon (Acipenser ruthenus)

Summary: This study analyzed the effect of DNA damage on embryo development and found significant changes in the transcriptome and proteome patterns after exposure to genotoxicants. The results suggest a correlation between phenotype formation and changes in transcriptomic and proteomic profiles. Additionally, markers of DNA damage response and adaptive stress response were identified.

Evolutionary conservation of maternal RNA localization in fishes and amphibians revealed by TOMO-Seq

Comparison of RNA localization during oogenesis within Acipenser ruthenus and Xenopus laevis

Summary: The localization of biomolecules within the oocyte drives the spatial differentiation of daughter cells during embryogenesis. While the localization pathways of vegetal maternal transcripts have been well-described, limited information is available about the localization of animal pole transcripts. In this study, TOMO-Seq analysis on two different species was performed, revealing temporal preferences for transcript localization establishment and the presence of both conserved and species-specific transcripts. Additionally, active remodeling of the maternal RNA pool through de novo transcription and degradation was observed.

Optimization of in vitro culture conditions of common carp germ cells for purpose of surrogate production

Summary: This study optimized the cultivation conditions for germ cells in common carp, and the results showed that using hESC media and RTG2 cell line as feeder cells promoted germ cell proliferation and survival. Additionally, opening the carp from the dorsal side minimized the risk of contamination. These findings are important for common carp breeding, genetic resource conservation, and development of specific techniques.

Temperature modulates gonadal development and sex ratio of pirancanjuba Brycon orbignyanus

Summary: The effect of temperature on gonadal development and sex ratio in Brycon orbignyanus was assessed in this study. Fish were kept at three different temperatures (20°C, 23°C, and 26°C) from the beginning of exogenous feeding. Results showed that the sexual pattern was temperature-dependent, with undifferentiated individuals at 20°C and a higher proportion of immature males at 23°C and males in maturation process and regression phase at 26°C. Testosterone levels were quantified in all treatments, while detectable estradiol levels were found only at 23°C. Continuous exposure to 23°C and 26°C favored testicular development, with faster gonadal development observed at 26°C.

Vitrification of the ovarian tissue in sturgeons

Summary: The aim of this study was to test the efficacy of needle-immersed vitrification (NIV) for preserving oogonia (OOG) in sterlet and Russian sturgeon ovarian tissue in order to enhance conservation efforts for these endangered fish species. The study found that vitrification of the ovarian tissue using specific solutions resulted in viable OOGs, which were able to successfully colonize recipient gonads after transplantation. The simplicity and practicality of the vitrification procedure make it a valuable tool for conservation work in the field.

Intracytoplasmic sperm injection in sturgeon species: A promising reproductive technology of selected genitors

Summary: In this study, the researchers applied intracytoplasmic sperm injection (ICSI) technology to endangered sturgeons and found that it successfully produced embryos and larvae. Furthermore, molecular analysis showed genome integration of both egg and sperm donor species, demonstrating the potential of ICSI for assisted reproduction in sturgeons.

Oogenesis and in vitro reproduction of the twospot astyanax Astyanax bimaculatus (Linnaeus, 1758) exposed to conspecific alarm substance

Summary: Stress can either promote or inhibit reproduction in fish, and the release of conspecific alarm substance (CAS) after a predator attack is one stressor that affects fish. This study focused on evaluating the effects of CAS exposure on the oogenesis and reproduction of the twospot astyanax Astyanax bimaculatus. The results showed that CAS exposure did not cause visible changes in the ovaries of female fish, but it led to precocious ovulation without successful offspring, reducing breeding success compared to the control group.

Does dietary exposure to 17?-ethinylestradiol alter biomarkers related with endocrine disruption and oxidative stress in the adult triploid of Danio rerio?

Summary: This study investigated the comprehensive effects of 17 alpha-ethinylestradiol (EE2) in zebrafish, focusing on endocrine disruption, oxidative stress, and detoxification processes. Adult male triploid zebrafish were exposed to two concentrations of EE2 in their feed for six weeks. The high concentration of EE2 caused extensive mortality and significant changes in reproductive disorders, gonad histology, vitellogenin level, and selected gene expression. The environmentally relevant concentration of EE2 showed no significant differences in vitellogenin but did affect antioxidant and detoxifying enzymes. This is the first study to report the effects of subchronic oral exposure to EE2 in adult triploid zebrafish.

Hydrogel encapsulation as a handling and vitrification tool for zebrafish ovarian tissue

Summary: The efficiency of sodium alginate hydrogel encapsulation technique during zebrafish ovarian tissue vitrification was evaluated. The results showed that the technique maintained the membrane integrity but did not have a cryoprotective effect. Halving the cryoprotectant concentration during encapsulation did not increase lipid peroxidation, and it provided support for oocytes after in vitro maturation.

Reproductive cycle of the flying fish Hemiodus unimaculatus (Bloch, 1794) (Characiforms: Hemiodontidae) in the upstream area of the Tucurui hydroelectric plant, Pará, Brazil

Summary: The reproductive cycle of Hemiodus unimaculatus in the upstream area of the Tucurui reservoir was studied. The results showed that the species goes through development phases in fall and winter, and spawning capable phases in spring and summer. Despite fishing restrictions, the species' population is declining in the upstream area of the reservoir, likely due to the incomplete coverage of the closed fishing season and the imbalance in the sex ratio.
