Summary: Xanthomonas spp. are important plant pathogens that threaten crop yields and food security. RpfF, a key enzyme involved in quorum sensing (QS) in Xanthomonas, is being explored as a potential target for the development of new pesticides. However, there is a lack of comprehensive reviews summarizing the progress in this field. This article provides a comprehensive review of the recent advances in DSF-mediated QS and reported inhibitors targeting the RpfF enzyme for controlling plant bacterial diseases.

Summary: This study investigated the role of ALDH2 in the progression of colorectal cancer (CRC) by targeting its enzymatic activity. The inhibition of ALDH2 reduced migration and stemness properties of CRC cells, and downregulated the expression of oncogenic genes in advanced CRC. The plant-derived isoflavone daidzein may be a potential chemotherapeutic drug targeting ALDH2 in CRC.

Summary: IMB5476, a novel nitrobenzoate microtubule inhibitor, exhibits increased aqueous solubility. It disrupts microtubule networks in cells, causing cell cycle arrest and inducing cell death through mitotic catastrophe and apoptosis. IMB5476 also inhibits angiogenesis and overcomes multidrug resistance.

Summary: In this study, novel phenylahistin derivatives were designed and synthesized, and it was found that appropriate hydrocarbon and unsaturated alkenyl substituents are important for enhancing their activity. Compound 15p showed potent cytotoxic activity against human lung cancer cells at the nanomolar level, inhibited microtubule polymerization, and induced high expression of caspase-3. In vivo experiments demonstrated that compound 15p effectively inhibited the growth of H22 transplanted tumors with lower toxicity compared to standard drugs. These findings suggest that the novel phenylahistin derivatives could be safe and effective potential agents for cancer treatment.

Summary: The upregulation of ALDH2 is closely associated with resistance to PTX in NSCLC. ALDH2-overexpressing NSCLC cells showed reduced PTX sensitivity and increased malignancy. Mechanistically, ALDH2 overexpression activated the RAS/RAF oncogenic pathway. ALDH2 inhibition may be a promising strategy to overcome PTX resistance.

Summary: Elmas et al. developed the OPPTI algorithm to identify overexpressed kinase proteins in multiple cancer types using global mass spectrometry proteomics data. They found that protein-level abnormalities, which are sometimes not observed by genomics, represent vulnerabilities in cancer that could be targeted for precision oncology. The study showed that proteomic analysis can reveal new treatment opportunities and validate genomic alterations in cancer proteins.

Summary: Excessive use of antibiotics poses a threat to public health and the environment. Understanding bacterial response to antibiotics and resistance regulation is crucial in addressing this issue. In addition to traditional mechanisms, bacterial signaling networks have been found to play a role in antibiotic response and resistance regulation. This review provides an overview of how bacterial intraspecific and interspecific signaling networks influence the response to environmental antibiotics, offering theoretical support in combating bacterial antibiotic resistance and mitigating health and ecological issues caused by antibiotic contamination.

Summary: Disulfiram (DSF) is a potential anti-tumor drug, but its anti-cancer mechanism is still unknown. This study reveals that DSF effectively reduces NDRG1 expression, a regulator of tumor metastasis, and inhibits the invasion of cancer cells. In vitro and in vivo experiments confirm that DSF regulates tumor growth, EMT, migration, and invasion of cervical cancer. Our results also demonstrate that DSF binds to HSP90A and affects the expression of NDRG1, providing novel insights into the function of DSF in cancer cells.
