Maternal cigarette smoking during pregnancy and offspring DNA methylation in midlife

Maternal smoking in pregnancy (MSP) has been associated with DNA methylation in specific CpG sites (CpGs) in infants and children. We investigated whether MSP, independent of own personal active smoking, was associated with midlife DNA methylation in CpGs that were previously identified in studies of MSP-DNA methylation in children. We used data on MSP collected from pregnant mothers of 89 adult women born in 1959-1964 and measured DNA methylation in blood (granulocytes) collected in 2001-2007 (mean age: 43years). Seventeen CpGs were differentially methylated by MSP, with multiple CpGs mapping to CYP1A1, MYO1G, AHRR, and GFI1. These associations were consistent in direction with prior studies (e.g., MSP associated with more and less methylation in AHRR and CYP1A1, respectively) and, with the exception of AHRR CpGs, were not substantially altered by adjustment for active smoking. These preliminary results confirm prior prospective reports that MSP influences the offspring DNA methylation, and extends the timeframe to midlife, and suggest that these effects may persist into adulthood, independently of active smoking.

The tool included a module covering cancer biology, local cancer burden, and risk reduction strategies, and was administered to over 280 high school and college students in New York City. Students identified various strategies for reducing cancer risk at both individual and community levels, suggesting the tool's potential for adoption in other communities and educational institutions.

Summary: In BRCA1 and BRCA2 mutation carriers, the use of combined-type oral contraceptive preparation (COCP) initially increases the risk of breast, ovarian, and endometrial cancer, but in the long term, it significantly decreases the lifetime cancer risk. Risk-reducing bilateral salpingo-oophorectomy and menopausal hormone therapy counteract the long-term benefits of COCP use.

Summary: This study demonstrates an efficient and reproducible method for measuring multiple polycyclic aromatic hydrocarbon metabolites in urinary samples. By optimizing the experimental steps and sample preparation, the method has improved sensitivity and recovery rates. This method is of great importance for epidemiology studies and biomonitoring programs.

Evidence-Based Interventions for Reducing Breast Cancer Disparities: What Works and Where the Gaps Are?

Summary: Breast cancer is the most common cancer in women worldwide, and there are multiple risk factors that can be intervened to reduce the burden of breast cancer. However, there is a lack of evidence-based cancer control programs for key risk factors such as alcohol, breastfeeding, and environmental chemical exposures. Even for existing programs on physical activity, obesity, and tobacco control, only a small fraction meets the quality criteria and are developed for populations facing breast cancer disparities. Therefore, there is a need for evidence-based interventions to address important risk factors and reduce breast cancer disparities.

Stressful Life Events, Social Support, and Incident Breast Cancer by Estrogen Receptor Status

Summary: Chronic stress is hypothesized to be associated with breast cancer, but previous studies have shown mixed results. This study examined the association between self-reported stressful life events and incident breast cancer in postmenopausal women, taking into account estrogen receptor (ER) status and social support. The results showed that there was no relationship between stressful life events and ER-positive breast cancer, but there was an increased risk of ER-negative breast cancer in women with higher levels of stressful life events. This association was stronger in widowed women. Social support did not modify the relationship between stressful life events and ER-negative breast cancer.

Assessment of personal care product use and perceptions of use in a sample of US adults affiliated with a university in the Northeast

Summary: Evidence shows that certain groups, such as femme-identifying individuals and racial and ethnic minorities, bear unequal burdens of chemical exposures from personal care products (PCPs). This study used an online questionnaire to assess PCP purchasing and usage behaviors and perceptions among a sample of US adults. The findings indicate that females use more PCPs than males, and there are variations in usage patterns among different racial and ethnic groups. Study participants generally expressed moderate concern about exposures and health effects from using PCPs, with minimal differences by gender, race, and ethnicity.
