CRTH2 promotes endoplasmic reticulum stress-induced cardiomyocyte apoptosis through m-calpain

Apoptotic death of cardiac myocytes is associated with ischemic heart disease and chemotherapy-induced cardiomyopathy. Chemoattractant receptor-homologous molecule expressed on T helper type 2 cells (CRTH2) is highly expressed in the heart. However, its specific role in ischemic cardiomyopathy is not fully understood. Here, we demonstrated that CRTH2 disruption markedly improved cardiac recovery in mice postmyocardial infarction and doxorubicin challenge by suppressing cardiomyocyte apoptosis. Mechanistically, CRTH2 activation specifically facilitated endoplasmic reticulum (ER) stress-induced cardiomyocyte apoptosis via caspase-12-dependent pathway. Blockage of m-calpain prevented CRTH2-mediated cardiomyocyte apoptosis under ER stress by suppressing caspase-12 activity. CRTH2 was coupled with G(q) to elicit intracellular Ca2+ flux and activated m-calpain/caspase-12 cascade in cardiomyocytes. Knockdown of caspase-4, an alternative to caspase-12 in humans, markedly alleviated CRHT2 activation-induced apoptosis in human cardiomyocyte response to anoxia. Our findings revealed an unexpected role of CRTH2 in promoting ER stress-induced cardiomyocyte apoptosis, suggesting that CRTH2 inhibition has therapeutic potential for ischemic cardiomyopathy.

Summary: Type I interferons (IFN-I) play a crucial role in protecting against viral infections, with STAT2 being a key component in driving gene expression in response to IFN-I. The phosphorylation of STAT2 at T404 acts as a critical conformational switch, disrupting an inactive STAT1-STAT2 dimer and enhancing the antiviral defense response to IFN-I. Additionally, virus infection-induced activation of IKK-epsilon can directly phosphorylate T404, further facilitating the antiviral defense mechanism.

Summary: The study found that C188-9, a small-molecule inhibitor of STAT3, has anti-fibrotic effects both in vitro and in vivo. Summary: This study reveals the important role of hepatic interferon regulatory factor 3 (IRF3) in regulating glucose homeostasis through the transcription factor Ppp2r1b. Elimination or reduction of IRF3 protects against steatosis and dysglycemia in obese mice. The study also suggests that obesity-induced insulin resistance and the restoration of glucose homeostasis can be reversed by inhibiting IRF3 expression. In addition, the activation of liver IRF3 in obese humans is enhanced and can be improved after bariatric surgery.

STAT2 hinders STING intracellular trafficking and reshapes its activation in response to DNA damage

Summary: In cancer cells, the abnormal presence of DNA in the cytoplasm activates cGAS and STING. STAT2 inhibits the expression of IRF3-dependent genes induced by cGAMP by binding to STING and blocking its intracellular trafficking. STAT2 reshapes STING signaling by inhibiting IRF3-dependent gene induction, but not NF-xB-dependent gene induction. This noncanonical activity of STAT2 depends on the phosphorylation of threonine 404, which promotes the formation of a STAT2:STING complex that keeps STING bound to the endoplasmic reticulum (ER) and increases resistance to DNA damage.
