Summary: The relationship between plasma EBV-DNA load (PEDL) and Epstein-Barr virus (EBV)-encoded small RNA (EBER) during the early treatment of lymphoma is not well-established. This study found that although EBER affects the levels of PEDL, there is poor concordance between EBER and qualitative analysis of PEDL (qPEDL).

The effects of EBV on children and adolescents with cHL are conflicting.

Summary: The article reports successful cases of using PD-1 inhibitor Sintilimab to treat CAEBV and EBV-HLH. Among the three patients, two had CAEBV and one had EBV-HLH. After treatment, the clinical symptoms of the patients improved significantly, and the EBV-DNA load decreased significantly. The study supports the efficacy of PD-1 targeted therapy in certain patients with CAEBV and EBV-HLH and suggests that Sintilimab may provide a cure without HSCT.

Summary: This study investigated the characteristics of primary EBV infection and reactivation in children in Suzhou, China, finding that primary EBV infection was more common in children aged 36 to <72 months and often led to infectious mononucleosis. Reactivation of EBV mainly occurred in children aged 8 to <36 months and was identified as a risk factor for hemophagocytic lymphohistiocytosis (HLH). Among reactivated patients, HLH had a higher viral load compared to EBV infection and respiratory infection.

Summary: This study reassessed familial aggregation of cancer in high-risk Taiwanese multiplex families with NPC cases and found evidence of NPC coaggregation within families, but not a broader familial syndrome involving NPC and other tumors. The study also observed elevated rates of secondary cancers among NPC cases, mostly head and neck and hematopoietic cancers, suggesting radiation treatment effects on subsequent cancer risk.

Summary: This review provides a comprehensive overview of Epstein-Barr virus-associated mesenchymal tumors, including their clinical features, pathological characteristics, pathophysiology, prognostic factors, and current treatment approaches. Through an extensive literature search, three distinct EBV-associated mesenchymal tumors were identified: EBV-associated smooth muscle tumors, inflammatory pseudotumor-like follicular dendritic cell sarcomas, and EBV-associated osteosarcomas. Although there is still incomplete understanding of the underlying pathophysiology, this review aims to serve as a catalyst for further research to bridge the knowledge gap and contribute to future therapeutic approaches.

Summary: Epstein-Barr virus (EBV) is closely associated with several malignant and lymphoproliferative diseases. Aberrant methylation of viral DNA and the host genome plays a crucial role in EBV-associated diseases. EBV controls gene methylation by hijacking host epigenetic machinery, and its proteins interact with host epigenetic regulatory factors to mediate the methylation of important tumour suppressor gene promoters. Epigenetic modifications, particularly DNA methylation, can be targeted for epigenetic therapy against EBV-associated tumours. The methylation patterns in the host and EBV genomes may also have diagnostic and prognostic value.
