Summary: A universal strategy for silane functionalization of metal-organic frameworks (MOFs) was developed, which involved the coupling of silanes with hydroxyl groups of MOFs to form metal-O-Si bonds. The strategy enabled the functionalization of MOFs with a wide variety of applications. Furthermore, the construction of supramolecular nanovalves on the surface of silanized MOFs demonstrated excellent controlled drug release and inhibitory effect on tumor growth.

Summary: Stimuli-responsive nanoparticle (NP) aggregation is increasingly important in regulating NP assembly into superstructures at different length scales. Various external stimuli, like light, temperature, pH, electric, and magnetic fields, are used to adjust the aggregation of responsive NPs. The review summarizes the strategies for NP aggregation by external stimuli, their recent progress, and applications in nanoscale aggregates, microscale superstructures, and macroscale bulk materials. The future opportunities and challenges for designing functional materials through NP aggregation at different length scales are also discussed.

Summary: Microneedle (MN) patches, made up of miniature needles, have emerged as a promising tool for minimally invasive translocation of biomolecules into the dermis. Stimuli-responsive MN patches are a new drug delivery system that can release cargos in response to internal or external triggers, with various mechanisms of action for different indications. Future opportunities and challenges in clinical translation are also discussed.

Summary: Immunotherapy plays an important role in cancer treatment, but there are challenges in delivering immunotherapeutic agents and activating them effectively at tumor sites. Hydrogels, combined with immunotherapies, offer a chance to enhance the immune response against tumors with reduced side effects. Stimuli-responsiveness is also applied to optimize pharmacokinetics and improve therapeutic outcomes. This review discusses the opportunities of combining immunotherapy with stimuli-responsive hydrogels, such as light, temperature, ultrasound, and magnetically responsive hydrogels, for effective cancer treatment. It also explores the potential of stimuli-responsive hydrogels as vaccine implants against cancer and Covid-19.
