Screening of lactic acid bacteria with potential protective effects against cadmium toxicity

Cadmium (Cd) is a toxic heavy metal that causes adverse health effects in humans and animals. In the present study, 33 strains of lactic acid bacteria (LAB) were screened for their potential probiotic capacity against Cd toxicity. The Cd binding ability was investigated first, and 10 strains with comparatively higher Cd binding properties were selected for further study through the evaluation of Cd tolerance, antioxidative capacity and acid and bile salt tolerance. The results revealed that as a reference strain, Lactobacillus rhamnosus GG had good Cd tolerance, antioxidative capacity and acid and bile tolerance, but the Cd binding ability of this strain was poor. Another strain, Lactobacillus plantarum CCFM8610, showed significantly better binding ability and Cd tolerance than the other tested strains. This strain also exhibited relatively good antioxidative properties and resistance to the simulated gastrointestinal conditions. This study suggests that L. plantarum CCFM8610 could be used as a potential probiotic in food applications against Cd toxicity. (C) 2015 Elsevier Ltd. All rights reserved.

Quorum Sensing of Lactic Acid Bacteria: Progress and Insights

Summary: The presence of lactic acid bacteria (LAB) as probiotics in the harsh intestinal environment is usually temporary. Quorum sensing (QS), a form of bacterial communication based on signaling molecules, has gained attention for its important role in regulating LAB properties that support colonization in the intestines. QS is also involved in autolysis and bacteriocin production by LAB, which are essential processes for the application of these species as probiotics or starter cultures. This review discusses LAB's QS systems, the factors that may affect QS, the impact of QS on LAB's intestinal colonization, and the possible interactions mediated by QS between probiotics and the gut microbiome.

Endogenous n-3 PUFAs Improve Non-Alcoholic Fatty Liver Disease through FFAR4-Mediated Gut-Liver Crosstalk

Summary: The gut-liver axis is crucial in the development of NAFLD and FFAR4 may act as a bridge between the gut and liver. In this study, FFAR4 deficiency in mice with high endogenous n-3 PUFAs blocked the protective effects on intestinal barrier dysfunction and hepatic steatosis. FFAR4 deficiency also affected the gut microbiota and various metabolic pathways associated with NAFLD aggravation. These findings highlight the role of FFAR4 in the treatment of NAFLD through the gut-liver axis.

Stimuli-Responsive Double Single-Atom Catalysts for Parallel Catalytic Therapy

Summary: This study developed PEGylated manganese/iron-based single-atom catalysts (SACs) that possess incredible catalytic activity. These SACs catalyze the conversion of cellular hydrogen peroxide to hydroxyl radical through a Fenton-like reaction and enhance the decomposition of hydrogen peroxide to cytotoxic superoxide ion via oxidase-like activity. The synergistic antitumor efficacy of these SACs has been demonstrated in in vitro and in vivo experiments. This study proposes new promising single-atom nanozymes with highly efficient biocatalytic sites and synergistic therapeutic effects for ROS-related biological applications in broad biomedical fields.

NASH Drug Development: Seeing the Light at the End of the Tunnel?

Summary: Nonalcoholic steatohepatitis (NASH) is a prevalent chronic liver disease with no approved drugs available. This minireview highlights the heterogeneous nature of NASH and the lack of consensus in outcome measures among clinical trials. We summarize potential therapeutic targets and candidate drugs for NASH treatment. Efficacy comparison of 33 published clinical trials evaluating noninvasive biomarkers and liver biopsy shows encouraging results for fibroblast growth factor 21 (FGF21), resmetirom, and pioglitazone in phase II and III trials, respectively.

Functions and substrate selectivity of diacylglycerol acyltransferases from Mortierella alpina

Summary: In this study, the DGAT1 and DGAT2 isoforms from M. alpina were analyzed and heterologously expressed in S. cerevisiae H1246. MaDGAT1A/1B/2A/2B were able to restore TAG synthesis, with different substrate preferences and selectivity. This study demonstrates the potential of MaDGATs in producing TAGs rich in linoleic acid and eicosapentaenoic acid, and provides a reference for improving TAG production in oleaginous fungi.

Dietary docosahexaenoic acid plays an opposed role in ferroptotic and non-ferroptotic acute kidney injury

Summary: Ferroptosis caused by polyunsaturated fatty acid (PUFA) peroxidation plays a significant role in the pathogenesis of acute kidney injury (AKI), suggesting that dietary intake of PUFA may pose a risk for individuals susceptible to AKI. However, in addition to ferroptosis, other mechanisms such as inflammation-associated necroptosis and pyroptosis also contribute to different types of AKI. Therefore, further evaluation is needed to understand the role of PUFA in different AKIs, especially & omega;3 PUFA which is commonly consumed as a food supplement.

Cis-monounsaturated fatty acids inhibit ferroptosis through downregulation of transferrin receptor 1

Summary: This study found that MUFAs with specific structural features, such as cis-conformation and centered double bond, can inhibit ferroptosis. Inhibition of SCD1 and SCD5 genes sensitizes cells to ferroptosis, suggesting the protective role of endogenous MUFA. These findings provide insights into the potential therapeutic applications of MUFAs in ferroptosis-related diseases.

Akkermansia muciniphila supplementation improves glucose tolerance in intestinal Ffar4 knockout mice during the daily light to dark transition

Summary: Disruption of blood glucose utilization can lead to diabetes mellitus, and free fatty acid receptors (FFARs) play an important role in the genetic and dietary aspects. This study found that FFAR4 has a significant effect on glucose homeostasis, and knockout of the FFAR4 gene impairs glucose tolerance in mice under high-fat feeding and chow diet conditions. Furthermore, deficiency of intestinal FFAR4 affects the composition of gut microbiota, especially at ZT12 under chow diet conditions.

Identification of PRDX5 as A Target for The Treatment of Castration-Resistant Prostate Cancer

Summary: The treatment of castration-resistant prostate cancer (CRPC) has always been a clinical challenge. The traditional approach is to reduce dihydrotestosterone biosynthesis or block androgen receptor (AR) signaling. However, this study suggests that the use of AR inhibitors can actually lead to drug tolerance and resistance. The up-regulation of peroxiredoxin 5 (PRDX5) is shown to promote resistance to AR inhibitors and the development of CRPC. Inhibition of PRDX5 suppresses drug-tolerant persister (DTP) cell proliferation, dampens CRPC development, and stabilizes disease progression in patients.

T-2 Toxin-Mediated β-Arrestin-1 O-GlcNAcylation Exacerbates Glomerular Podocyte Injury via Regulating Histone Acetylation

Summary: This study investigates the role of metabolic disorder/reprogramming-mediated epigenetic modification in the progression of T-2 toxin-stimulated podocyte injury. The researchers found that O-GlcNAcylation plays a vital regulatory role in the development of T-2 toxin-stimulated podocyte injury by activating the mTOR pathway to suppress autophagy. Targeting beta-arrestin-1 or OGT could be a potential therapy for T-2 toxin infection-associated glomerular injury, especially podocyte injury.

Prevalence and quantitative assessment of foodborne viruses on the imported mussels in Polish market

Summary: The aim of this study was to assess the prevalence of human enteric viruses in mussels sold on the Polish market and study the fecal contamination of shellfish. The study found that human enteric viruses were commonly present in shellfish, with norovirus being the most frequently detected. The study also observed a seasonal distribution pattern of viruses in shellfish samples.

Laser-induced fluorescence spectroscopy enhancing pistachio nut quality screening

Summary: Food safety and quality are increasingly important in our society, driving the development of novel optical food sensing technologies. However, current optical sensing technologies are often limited in their sensitivity and product variability. In this study, we developed a novel pistachio nut screening methodology using fluorescence spectroscopy and advanced chemometrics to simultaneously detect shells, tree parts, and aflatoxins. By optimizing hardware and software parameters, we achieved a high classification accuracy for healthy pistachio kernels.

Photon-phonon coupling cold sterilization technology for milk preservation

Systematic review on biosensors for the early detection of mycotoxins as endocrine disruptors

Summary: This study used bibliometric visualisation map analysis to explore the publication outputs, trends, and hotspots for endocrine disruptors (EDMs) biosensor development in relation to mycotoxins. The analysis showed that the current literature primarily focuses on developing biosensors for detecting highly toxic and widely distributed mycotoxins in food, but there needs to be more focus on biosensors for detecting mycotoxins known as endocrine disruptors.

Discrimination amongst olive oil categories by means of high performance-ion mobility spectrometry: A step forward on food authentication

Summary: High-performance-ion mobility spectrometry (HPIMS) was used for the first time in the analysis of olive oil samples, following a simple liquid-liquid extraction procedure. The developed methodology is user-friendly, fast, and inexpensive, complying with the principles of green chemistry. The proposed approach, combined with multivariate analysis, showed potential in classifying olive oil samples based on their organoleptic quality, with high accuracy rates.

Summary: Polycyclic aromatic hydrocarbons (PAHs) are a major cause of human cancer. This study developed a quantitative analysis method for PAH4 using the back propagation neural network (BPNN) algorithm and constant wavelength synchronous fluorescence (CWSF) spectra as the data sets. The method can predict the concentrations of PAH4 in edible oil samples without preprocessing or pre-separation. It has been proven to be a powerful tool for the rapid detection of PAH4.

Formulation with zinc acetate enhances curcumin's inherent and photodynamic antimicrobial effects for food preservation

Summary: Food spoilage is primarily caused by microbial contamination. Curcumin has shown moderate antimicrobial activity as a food preservative. This study formulates curcumin with zinc acetate to enhance its efficacy, resulting in Cur-Zn with increased antimicrobial activity against multiple food-contaminating bacteria and fungi. With light illumination, the antimicrobial activity of Cur-Zn is further enhanced due to the photodynamic inactivation (PDI) effect.

Potential of antimicrobial peptide-fused endolysin LysC02 as therapeutics for infections and disinfectants for food contact surfaces to control Cronobacter sakazakii

Summary: This study demonstrates the enhanced bactericidal activity of engineered lysins fused with amphipathic antimicrobial peptides against Gram-negative bacterium Cronobacter sakazakii. The fusion lysins are capable of disrupting bacterial cells and significantly reducing the risk of infection within a certain range of temperature and pH. Additionally, the study also shows the bactericidal effect of these fusion lysins against other Gram-negative pathogens.

Multi-block approach for the characterization and discrimination of Italian chickpeas landraces
