The immune system as a chronotoxicity target of the anticancer mTOR inhibitor everolimus

The circadian timing system controls many biological functions in mammals including xenobiotic metabolism, detoxification, cell proliferation, apoptosis and immune functions. Everolimus is a mammalian target of rapamycin inhibitor, whose immunosuppressant properties are both desired in transplant patients and unwanted in cancer patients, where it is indicated for its antiproliferative efficacy. Here we sought whether everolimus circadian timing would predictably modify its immunosuppressive effects so as to optimize this drug through timing. C57BU6J mice were synchronized with light-dark 12h:12h, with L onset at Zeitgeber Time (ZT) 0. Everolimus was administered orally to male (5 mg/kg/day) and female mice (15 mg/kg/day) at ZT1, during early rest span or at ZT13, during early activity span for 4 weeks. Body weight loss, as well as hematological, immunological and biochemical toxicities, were determined. Spleen and thymus were examined histologically. Everolimus toxicity was less severe following dosing at ZT13, as compared to ZT1, as shown with least body weight inhibition in both genders; least reductions in thymus weight both in males (p < 0.01) and females (p < 0.001), least reduction in female spleen weight (p < 0.05), and less severe thymic medullar atrophy both in males (p < 0.001) and females (p < 0.001). The mean circulating counts in total leukocytes, total lymphocytes, T-helper and B lymphocytes displayed minor and non-significant changes following dosing at ZT13, while they were decreased by 56.9% (p < 0.01), 45.5% (p < 0.01), 43.1% (p < 0.05) and 48.7% (p < 0.01) after everolimus at ZT1, respectively, in only male mice. Chronotherapy of everolimus is an effective way to increase the general tolerability and decrease toxicity on the immune system.

Assessment of Minimal Residual Disease in Childhood Acute Lymphoblastic Leukemia: A Multicenter Study From Turkey

Summary: Assessing minimal residual disease (MRD) is crucial for risk classification and personalized therapy in childhood acute lymphoblastic leukemia (ALL). A study in Turkey retrospectively collected data of pediatric ALL patients treated with Berlin-Frankfurt-Munster (BFM) protocols. MRD assessment was done using real-time quantitative polymerase chain reaction (qPCR) and multiparametric flow cytometry (MFC). MRD monitoring led to upgrading 2% of patients from intermediate risk group to high-risk group. Factors such as age ≥10 years, poor response to prednisone, PCR-MRD ≥10(-3) on day 33 and day 78 were identified as poor prognostic factors affecting event-free survival (EFS). Establishing infrastructure and ensuring standardization for MRD methods are crucial for optimal management of children with ALL.

The relationship between serum A proliferation-inducing ligand and B-cell activating factor levels with disease activity and organ involvement in systemic lupus erythematosus

Summary: This study aimed to investigate the association between serum B-cell activating factor (BAFF) and A proliferation-inducing ligand (APRIL) levels with disease activity and clinical findings in SLE patients. The results showed that both BAFF and APRIL levels were higher in patients with SLE compared to the control group, but there was no correlation between BAFF/APRIL levels and disease activity scores. Serum BAFF levels were higher in patients with renal disease activity, while APRIL levels were significantly correlated with proteinuria. The study suggests that BAFF can be a valuable biomarker in SLE, especially in patients with renal activity.

Comparison of mitogen-induced proliferation in child and adult healthy groups by flow cytometry revealed similarities

Summary: This study investigated and compared mitogen-stimulated proliferation responses of lymphocytes in children and adult healthy donors. The findings revealed similarities between the two age groups, with the only difference being a significantly high proliferation of pediatric CD4(+) T cells in response to PHA. The study also found that the CD4(+) T cell responses against PHA diminished with advancing age in pediatric individuals.

Summary: This study evaluated the changes in intracellular cytokine expression patterns in different T-lymphocyte subsets after ozone therapy in patients with refractory idiopathic granulomatous mastitis (IGM). The results showed that ozone therapy stimulated a specific immune response in T cells and downregulated the expression of TGF-β in regulatory T cells. These immune system changes can improve wound healing and restore immune dysfunction in patients with refractory IGM.

Summary: This study aimed to determine the expression of IFN-gamma R1 and IL-12R beta 1 in patients with MSMD, their relatives, and healthy individuals, and to evaluate the importance of flow cytometry as a fast and reliable method in the diagnosis of MSMD. The results showed that the determination of IL-12R beta 1 and IFN-gamma R1 deficiencies by flow cytometry can be used as a rapid and reliable method for the diagnosis of MSMD.

Tail-Approach-Based Design and Synthesis of Coumarin-Monoterpenes as Carbonic Anhydrase Inhibitors and Anticancer Agents

Summary: In this study, sixty novel coumarin-monoterpene compounds were synthesized and evaluated for their inhibitory effects on hCA isoforms and anticancer potentials. All synthesized molecules selectively inhibited CA IX and XII, with compounds 23 and 42 showing the strongest inhibitory activity against hCA IX. Cytotoxicity evaluation revealed that compounds 14 and 63 exhibited the highest cytotoxicity on MCF-7 cells, while compound 23 showed the strongest cytotoxic effect on both PC-3 and HT-29 cell lines. Furthermore, compounds 14, 23, and 66 showed a reduction in CA IX and CA XII protein expression and increased apoptosis in different cell lines. The modeling studies demonstrated that only the open coumarin form of the compounds could interact directly with the active-site Zn2+ ion.

The Evaluation of Multidrug Resistance-Related Protein 1 as a Prognostic Factor in the Pediatric B-cell Acute Lymphoblastic Leukemia: A Pilot Study

Summary: Acute lymphoblastic leukemia (ALL) is the most common type of cancer in children, and multidrug resistance is a major obstacle in its treatment. This study investigates the expression of multidrug resistance-related protein 1 (MRP1) in children with B-ALL and compares it with other prognostic factors. The results show that there is no significant difference in MRP1 expression between different risk groups and other prognostic factors such as flow cytometric minimal residual disease (MRD) and DNA index.

Diurnal Changes in Capecitabine Clock-Controlled Metabolism Enzymes Are Responsible for Its Pharmacokinetics in Male Mice

Summary: The circadian timing system is responsible for regulating the pharmacokinetics of drugs over a 24-hour period. Understanding the temporal changes in pharmacokinetics is crucial for anticancer drugs with narrow therapeutic ranges and dose-limiting side effects. This study evaluated the diurnal rhythm-dependent metabolism of capecitabine and found that the levels of metabolizing enzymes varied throughout the day, leading to differences in pharmacokinetics. This highlights the importance of considering the administration time of capecitabine to maximize efficacy and minimize adverse effects.

Intracellular Levels of IL-10 and STAT3 in Patients with Chronic Lymphocytic Leukemia

Summary: In this study, STAT3 and IL-10 levels were investigated in CLL patients. The results showed increased IL-10+CD19+, IL-10+ and STAT3+CD19+ in lymphocytes of patients compared to healthy subjects. The expression of IL-10 was found to be increased in CD19+ B cells of CLL patients, while the expression of STAT3 was lower in patients compared to healthy controls. These findings suggest that the levels of IL-10 and STAT3 expression in CLL B cells differ from normal B lymphocytes and may play a role in the biology of CLL.

This review summarizes the current knowledge on circadian rhythmicity in adipose tissue and examines the potential links between alterations in adipose tissue and metabolic diseases, as well as discussing therapeutic approaches and clock genes as potential targets for chronotherapy.

Investigation the role of nitric oxide pathway in TNF-& alpha; induced HUVEC cells

Summary: The study investigated the role of nitric oxide (NO) in HUVEC cells under inflammatory conditions induced by tumor necrosis factor alpha (TNF-a). The phosphorylation of ERK, Akt, and eNOS was examined to identify TNF-a induction mechanisms. Sildenafil and L-NAME were used to examine the role of NO. The results showed that TNF-a increased nitrite/nitrate production and decreased cell viability, while sildenafil and L-NAME had no effect on cell viability but decreased nitrite/nitrate levels in stimulated HUVEC cells. TNF-a had no effect on the phosphorylation of ERK, Akt, and eNOS, but increased total eNOS levels. Sildenafil and L-NAME had no effect on protein phosphorylation.
