There are only a few studies that have assessed the effect of bisphenol A (BPA) on human blood cells and no study has been conducted to analyze the impact of BPA analogs on human leucocytes. In this study, we have investigated the effect of BPA and its analogs like bisphenol F (BPF), bisphenol S (BPS) and bisphenol AF (BPAF) on apoptosis induction in human peripheral blood mononuclear cells (PBMCs). In order to clarify the mechanism of bisphenols-induced programmed cell death, changes in various signaling molecules of this process have been assessed. We observed an increase in cytosolic calcium ions (Ca2+) level and reduction of transmembrane mitochondrial potential (Delta Psi m) in PBMCs incubated with all compounds examined, and particularly BPA and BPAF. All compounds studied changed PBMCs membrane permeability, activated caspase-8, -9, -3 and induced PARP-1 cleavage and chromatin condensation, which confirmed that they were capable of inducing apoptosis both via intrinsic and extrinsic pathway. Moreover, we have found that modus operandi of bisphenols studied was different. We noticed that BPAF and BPS caused mainly necrotic and apoptotic changes, respectively, whereas BPA induced comparable apoptotic and necrotic effects in the incubated cells. (C) 2015 Elsevier Ltd. Summary: Endocrine disruptors are chemical compounds that cause hormonal imbalance in the body even at low concentrations, leading to various harmful health disorders. PFASs, particularly long- and short-chain variants, have gained significant attention due to their long-lasting presence in the human body and environment, potentially causing short- and long-term health effects.

Benzo[a]pyrene-Environmental Occurrence, Human Exposure, and Mechanisms of Toxicity

Bozena Bukowska, Katarzyna Mokra, Jaromir Michalowicz

Summary: Benzo[a]pyrene (B[a]P) is a representative polycyclic aromatic hydrocarbon (PAH) that is commonly found in the environment and human surroundings. It has been shown to have toxic effects on DNA, reproduction, neurology, and development. The metabolism of B[a]P is mediated by cytochrome P450 (CYP P450) and its polymorphism determines the sensitivity to B[a]P. Additionally, B[a]P may facilitate the replication of viruses.

Glyphosate disturbs various epigenetic processes in vitro and in vivo - A mini review

Bozena Bukowska, Ewelina Wozniak, Paulina Sicinska, Katarzyna Mokra, Jaromir Michalowicz

Summary: Glyphosate has been shown to induce epigenetic changes that may be associated with carcinogenesis. These changes include alterations in DNA methylation, gene expression, chromatin structure, and the regulation of non-coding RNA molecules. These epigenetic changes can persist over time and be passed on to future generations, potentially leading to the development of diseases such as prostate disease or obesity. More research is needed to fully understand the metabolic effects of these changes on organisms.
