Ovarian cancer risk, ALDH2 polymorphism and alcohol drinking: Asian data from the Ovarian Cancer Association Consortium

The aldehyde dehydrogenase 2 (ALDH2) polymorphism rs671 (Glu504Lys) causes ALDH2 inactivation and adverse acetaldehyde exposure among Asians, but little is known of the association between alcohol consumption and rs671 and ovarian cancer (OvCa) in Asians. We conducted a pooled analysis of Asian ancestry participants in the Ovarian Cancer Association Consortium. We included seven case-control studies and one cohort study comprising 460 invasive OvCa cases, 37 borderline mucinous OvCa and 1274 controls of Asian descent with information on recent alcohol consumption. Pooled odds ratios (OR) with 95% confidence intervals (CI) for OvCa risk associated with alcohol consumption, rs671 and their interaction were estimated using logistic regression models adjusted for potential confounders. No significant association was observed for daily alcohol intake with invasive OvCa (OR comparing any consumption to none=0.83; 95% CI=0.58-1.18) or with individual histotypes. A significant decreased risk was seen for carriers of one or both Lys alleles of rs671 for invasive mucinous OvCa (OR=0.44; 95% CI=0.20-0.97) and for invasive and borderline mucinous tumors combined (OR=0.48; 95% CI=0.26-0.89). No significant interaction was observed between alcohol consumption and rs671 genotypes. In conclusion, self-reported alcohol consumption at the quantities estimated was not associated with OvCa risk among Asians. Because the rs671 Lys allele causes ALDH2 inactivation leading to increased acetaldehyde exposure, the observed inverse genetic association with mucinous ovarian cancer is inferred to mean that alcohol intake may be a risk factor for this histotype. This association will require replication in a larger sample.

Summary: This study investigated the difficulty in removing totally implantable venous access devices (TIVADs) and identified associated risk factors. Interventional radiology (IR) approaches for difficult removal were also explored. The results showed that indwelling duration and subcutaneous leakage of fluid were significant risk factors for difficult removal. IR techniques were found to be minimally invasive and useful in some cases.

Summary: This study analyzed the relationship between cigarette smoking, alcohol consumption, and head and neck cancer risk factors in less developed and more developed countries. The results showed that the duration of smoking and alcohol consumption had different effects on the risk of specific cancer sites depending on the country's level of development. These findings suggest that the degree of industrialization and economic development affects the relationship between smoking, alcohol, and head and neck cancer.

Summary: This study utilized GWAS, genetic correlation analysis, and polygenic risk modeling to investigate the genetic causes of BE/EA. The findings identified new risk loci and candidate genes associated with BE/EA development. The study also revealed differences in the etiology of BE and EA and demonstrated improved risk prediction models when combining PRS with risk factors. These findings provide valuable insights into the mechanisms underlying BE/EA and improve our understanding of the disease.

Summary: Pedigree analysis showed that most LHON family members carrying a mitochondrial risk variant do not develop vision loss. Mitochondrial haplotype plays a major role in the risk of vision loss from LHON. The prevalence of LHON variants in the general population is relatively low, and the penetrance of LHON is very low in carriers.

Common analgesics and ovarian cancer survival: the Ovarian cancer Prognosis And Lifestyle (OPAL) Study

Summary: This study investigated the association between nonsteroidal antiinflammatory drug (NSAID) use and 5-year cancer-specific survival in Australian women with ovarian cancer. The results showed that frequent use of NSAIDs, including aspirin and nonaspirin NSAIDs, was associated with better survival compared to nonusers and infrequent users. These findings suggest that NSAID use might improve ovarian cancer survival.

Summary: Refractive error is a complex eye condition caused by genetic and environmental factors. Genome-wide association studies have identified common genetic risk factors, but a significant portion of the heritability is still unexplained. By conducting gene-based association tests, we identified 129 unique genes associated with refractive error in a large dataset. Some of the novel candidates, such as PDCD6IP, PER3, and P4HTM, show promise for future functional studies and validation.

Summary: The effect of body mass index (BMI) on esophageal and gastric carcinogenesis varies depending on subtype or subsite. In this study, a pooled analysis of 10 population-based cohort studies in Japan, we observed an inverse association between BMI and esophageal squamous cell carcinoma, but a positive association with gastric cardia cancer. Additionally, a positive association was found for overweight or obese individuals with esophageal adenocarcinoma and gastric cardia cancer. The study highlights the subtype- or subsite-specific impact of BMI on upper gastrointestinal cancer in the Japanese population.

Health services costs for ovarian cancer in Australia: Estimates from the 45 and Up Study

Summary: This study aimed to estimate the direct health system costs of women diagnosed with ovarian cancer in Australia during 2006-2013, and found that the costs are substantial. Summary: Cancer cachexia is a metabolic disorder characterized by a decrease in B vitamin-related liver enzymes. The relationship between enzymes and their associated metabolites may depend on protein abundance and cofactor subtype. These metabolic/proteomic changes can be utilized for the diagnosis of cancer cachexia through blood samples.

The Heterogeneous Impact of Prediagnostic Folate Intake for Fluorouracil-Containing Induction Chemotherapy for Head and Neck Cancer

Summary: This study is the first to suggest an association between prediagnostic folate intake and head and neck squamous cell carcinoma (HNSCC) survival that differs based on fluorouracil (FU)-containing chemotherapy. In the FU-containing chemotherapy group, higher folate intake is associated with better overall survival, while no clear association is observed in the non-FU-containing chemotherapy group. This finding indicates that the folate status affects the heterogeneity of one-carbon metabolism in HNSCC.
