PTEN PDZ-binding domain suppresses mammary carcinogenesis in the MMTV-PyMT breast cancer model

Phosphatase and tension homolog (PTEN) is a potent tumor suppressor that possesses a PDZ-binding domain (PDZ-BD) at the end of its carboxyl terminus, whose functions during tumorigenesis remains unclear. Here, we crossed a mouse strain with germline deletion of PTEN PDZ-BD with MMTV-PyMT breast cancer model, and found that knockout (KO) mice display normal development of mammary glands, but have both increased breast tumorigenicity and lung metastasis. Orthotopic allograft experiments suggest the loss of PTEN PDZ-BD in breast cancer cells rather than in tumor microenvironment plays a prominent role in increasing tumor burden. Through RNA-sequencing, we observed a significant downregulation of myoepithelial marker genes in both KO primary breast cancer and orthotopic allografts. Moreover, these myoepithelial marker genes are significantly down regulated in human breast cancer tissues, and are associated with poorer clinical prognosis. In addition, several homeobox genes were also identified to be downreguated in KO breast cancer, whose expressions showed significant positive correlation with myoepithelial marker genes. Overall, our findings suggest a novel tumor suppressive role of PTEN PDZ-BD in a murine model of breast cancer, and the mechanism involves the dysregulation of homeobox genes which may result in defective myoepithelial differentiation in breast cancer cells.

Androgen Receptor as an Emerging Feasible Biomarker for Breast Cancer

Summary: Biomarkers, such as estrogen receptor alpha(ER alpha), human epidermal growth factor receptor 2 (HER2), and androgen receptor (AR), have been widely used in breast cancer diagnosis, prognosis, and prediction. AR serves different roles in different breast cancer subtypes, acting as a tumor suppressor in ER+ breast cancers but a tumor promoter in ER- breast cancers. AR expression status can also predict the response to hormonal therapy and chemotherapy. Therefore, AR has the potential to be a useful biomarker in breast cancer.

ALK-positive Large B-Cell Lymphoma Presenting as a Circumscribed Breast Mass with Germinal Center Immunophenotype

Summary: We report a rare case of ALK-positive large B cell lymphoma initially presenting as a breast lesion. Immunophenotypic and immunostaining results revealed typical immunoblastic morphology and confirmed the fusion of ALK and CTCL. Additionally, expression of CD10 and BCL6 suggested a germinal center origin for the lesion.

Targeting Ribosome Biogenesis to Combat Tamoxifen Resistance in ER plus ve Breast Cancer

Summary: Breast cancer is a heterogeneous disease, and most cases are estrogen receptor-positive. However, many patients eventually develop resistance to the main treatment drug tamoxifen. Overexpression of c-MYC can drive the development of estrogen receptor-positive breast cancer and confer tamoxifen resistance. Ribosome biogenesis plays a crucial role in this process, and suppressing ribosome biogenesis may help reduce aggressiveness and reverse tamoxifen resistance. Some chemicals have been shown to repress ribosome biogenesis and may potentially reverse tamoxifen resistance in estrogen receptor-positive breast cancer. Identification of predictive markers will be necessary for the future use of these ribosome biogenesis inhibitors in combating tamoxifen resistance.

Aggressive Lymphoplasmacytic Neoplasm With an Unusual In-frame Deletion of MYD88 Associated With TRAF3 and TP53 Mutations and Complex Karyotype

Summary: This study reports an unusual MYD88 in-frame deletion in an aggressive lymphoplasmacytic neoplasm. Only 8 other mature B-cell lymphomas with MYD88 in-frame deletions were identified, but none of them were diagnosed with lymphoplasmacytic lymphoma. Recognizing such in-frame deletions is necessary to understand the mutational spectrum of MYD88 in B-cell lymphomas, and further investigation is needed to determine whether these deletions are overrepresented in lymphoplasmacytic lymphoma compared to other B-cell lymphomas.

Breast MALT Lymphoma: A Clinical, Histomorphologic, and Immunophenotypic Evaluation

Summary: This study analyzed the clinical and pathological features of 5 primary and 6 secondary breast MALT lymphomas diagnosed in our institution over a 20-year period. Most primary and secondary breast MALT lymphomas had similar clinical presentations, but primary lymphomas tended to occur in older patients and thyroid abnormalities were common. There were no distinct histopathologic findings in primary lymphomas, indicating that they do not share the distinctive features of primary cutaneous marginal zone lymphoma.

Summary: Overexpressing BQ can confer tamoxifen resistance in ER +ve breast cancer. BQ is an alternative splice variant of NCOR2 generated through the exclusion of exon 11. SRSF5 is involved in the regulation of NCOR2 splicing. Low expression of SRSF5 increases the production of BQ mRNA by excluding exon 11. Thus, targeting the upstream pathway of BQ may reverse tamoxifen resistance.

Overexpression of BQ323636.1 contributes to anastrozole resistance in AR plus ve/ER plus ve breast cancer

Summary: This study revealed that overexpression of a novel splice variant BQ323636.1 is associated with resistance to the non-steroidal aromatase inhibitor anastrozole in ER+ post-menopausal breast cancer. Mechanistic studies showed that BQ overexpression enhances androgen receptor (AR) activity, leading to hyper-activation of AR signaling and increased cell proliferation. Targeting AR can overcome anastrozole resistance in breast cancer with BQ overexpression. Clinical study also demonstrated that high nuclear expression of both BQ and AR is significantly associated with poor survival in non-steroidal aromatase inhibitor-treated ER+ breast cancer patients.
