Development of a functional thyroid model based on an organoid culture system

The low turnover rate of thyroid follicular cells and the lack of a long-term thyroid cell culture system have hampered studies of thyroid carcinogenesis. We have now established a thyroid organoid culture system that supports thyroid cell proliferation in vitro. The established mouse thyroid organoids performed thyroid functions including thyroglobulin synthesis, iodide uptake, and the production and release of thyroid hormone. Furthermore, transplantation of the organoids into recipient mice resulted in the formation of normal thyroid-like tissue capable of iodide uptake and thyroglobulin production in vivo. Finally, forced expression of oncogenic NRAS (NRAS(Q61R)) in thyroid organoids established from p53 knockout mice and transplantation of the manipulated organoids into mouse recipients generated a model of poorly differentiated thyroid cancer. Our findings suggest that this newly developed thyroid organoid culture system is a potential research tool for the study of thyroid physiology and pathology including thyroid cancer. Comparative analysis of opioid use between robotic and open pancreatoduodenectomy

Summary: Compared to traditional open PD, patients undergoing robotic PD required less opioid use during hospitalization while maintaining similar pain scores. Despite a shorter length of stay, a higher percentage of robotic PD patients tapered off opioids prior to discharge. These results provide a rationale for choosing robotic PD when feasible to minimize opioid use.

Borrmann Type Predicts Response to Preoperative Therapy in Advanced Gastric Cancer

Summary: This study aimed to investigate the relationship between Borrmann type and response to preoperative therapy in advanced gastric cancer. Summary: This study aimed to evaluate the impact of biologics on the risk of advanced-stage inflammatory bowel disease (IBD)-associated intestinal cancer. The results showed that in patients with Crohn's disease (CD), no drugs were significantly associated with cancer stage. However, in patients with ulcerative colitis (UC), less use of biologics, 5-aminosalicylic acid, and immunomodulators was significantly associated with advanced cancer stage. Multivariate analysis demonstrated that biologic use was significantly associated with a lower risk of advanced-stage disease.

Clinical Impact of Positive Surgical Margins in Gastric Adenocarcinoma in the Era of Preoperative Therapy

Summary: Microscopically positive (R1) surgical margins after gastrectomy increase gastric cancer recurrence risk, and this study aimed to explore the impact of R1 margins on recurrence patterns and survival. The results showed that patients with R1 margins had aggressive clinicopathologic features and a significantly lower 5-year overall survival rate. Most patients with R1 margins experienced peritoneal recurrence, and the time to peritoneal dissemination recurrence was relatively short.

The results show that the SFV mostly returns to the inferior mesenteric vein, followed by the superior mesenteric vein and splenic vein. The AMCA is present in many cases and is mainly connected to the superior mesenteric artery or its branches.

Summary: This study aimed to compare the benefits of open gastrectomy (OG) and robotic gastrectomy (RG) in gastric cancer patients. The results showed that RG had longer surgery time, smaller blood loss volume, shorter recovery time compared to OG, but there was no significant difference in postoperative complication rates between the two groups. Overall, RG was found to be a safe option for gastric cancer patients.

This suggests that surgical management was conducted adequately even with limited medical resources and staff during the pandemic.

Summary: Glycolysis is increased in pancreatic ductal adenocarcinoma (PDAC) cells, resulting in glucose restrictions on nontumor cells in the tumor microenvironment. Cancer-associated fibroblasts (CAFs) with restricted glucose availability utilize lactate as a fuel and exhibit immunosuppressive activity in the PDAC microenvironment. Coculture experiments revealed that glucose is mainly consumed by tumor cells, while CAFs consume lactate through monocarboxylate transporter 1 to enhance proliferation. Lactate-stimulated CAFs upregulate IL-6 expression and synergistically suppress cytotoxic immune cell activity with lactate. Inhibiting lactate dehydrogenase A (LDHA) reduces tumor growth and improves antitumor immunity in CAF-rich PDAC tumors.

A nano-liposomal carrier containing p-coumaric acid for induction of targeted apoptosis on melanoma cells and kinetic modeling

YRNA and tRNA fragments can differentiate benign from malignant canine mammary gland tumors

They identified two types of oscillations, sarcomeric oscillations and sarcosynced oscillations, and visualized their behavior during propagating waves. The researchers discovered the presence of sarcomeric defect holes and sarcomeric collision holes, which are important indicators for understanding the oscillation properties of sarcomeres. This finding has important implications for improving our understanding of muscle function and its regulatory mechanisms.

TP53INP2 modulates the malignant progression of colorectal cancer by reducing the inactive form of β-catenin

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

The four subunits of rabbit skeletal muscle lactate dehydrogenase do not exert their catalytic action additively

Summary: Oligomeric enzymes are known for their higher catalytic rates compared to monomeric enzymes, but the extent of additivity in their activity is still not well understood. This study used tetrameric rabbit lactate dehydrogenase as a model to examine the kinetics of its catalytic action. Surprisingly, when the concentration of the limiting reactant exceeded that of a single subunit, there was a significant slowdown in the enzyme's conformational rearrangements.

The results showed that indium curcumin and vanadyl curcumin exhibited higher binding affinities and stronger inhibitory effects on amyloid fibrillation compared to gallium curcumin.
