Summary: Overexpression of histone reader TRIM24 in mouse mammary epithelia can drive the development of mammary carcinosarcoma tumors with similarities to human MpBC tumors. The direct oncogenic target of TRIM24 is Met, leading to aberrant PI3K/mTOR activation, and targeting these pathways may have therapeutic potential in TRIM24-expressing TNBC.

Summary: This study evaluated the characteristics and outcomes of surgical treatment in older patients with gynecologic cancers. The study included 131 patients aged 65 years and above who underwent surgery for gynecologic cancers. The results showed that older patients had good surgical outcomes and tolerable postoperative complications, indicating that surgical treatment can be safely offered to older patients with gynecologic cancers.

Summary: Metaplastic breast carcinoma (MBC) is a rare and aggressive type of triple-negative breast cancer with poor prognosis. This study aimed to evaluate the demographics and characteristics of MBC, as well as survival outcomes based on presentation and treatment. Factors such as tumor size, grade, stage, and patient age were found to be correlated with worse prognosis, while the best overall survival was seen in patients who underwent combined surgery, chemotherapy, and radiotherapy. Black patients had significantly worse outcomes compared to White patients, with higher rates of aggressive tumor features at presentation. The most common genetic mutations observed were TP53, PIK3CA, and LRP1B.

Summary: This study compared characteristics and outcomes of patients with metaplastic breast cancer (MBC) by treatment sequence, finding no significant survival difference between neoadjuvant chemotherapy (NACT) and adjuvant chemotherapy (ACT) recipients. NACT recipients were typically younger than ACT recipients.

Summary: Metaplastic breast carcinoma (MBC) is a rare and aggressive subtype of breast cancer with poorer survival compared to other triple-negative breast cancers. Among MBC patients, those with heterologous mesenchymal subtype had the best survival, while those with squamous subtype had the worst survival. This calls for further research to identify tailored therapies for MBC's unique biology.

Summary: Metaplastic breast cancer (MpBC) is a rare aggressive malignancy with poor prognosis. The relationships between clinical and pathological features, treatment approach, and outcomes of MpBC remain underexplored. In this review article, existing data on MpBC regarding clinical, pathological and genomic features, management, and outcomes are synthesized, and potential targets for future clinical trials are identified.

Summary: This study retrospectively analyzed the association between the use of metformin and COVID-19 outcomes in patients with type 2 diabetes. The results showed that compared to sulfonylureas, metformin was associated with a lower risk of mechanical ventilation and mortality. However, there was no significant association compared to dipeptidyl-peptidase-4-inhibitors.

Summary: The incidence of postoperative mortality and complications was higher in the reoperation group compared to the primary surgery group. Strong risk factors for operative mortality among patients undergoing reoperation included urgency status, ejection fraction <30%, IE, dialysis, chronic kidney disease, New York Heart Association class 3/4, concomitant coronary artery bypass grafting and aorta procedure, tricuspid valve surgery only, multivalve surgery and age. Matching analyses showed higher relative odds of operative mortality among patients with and without IE in reoperation procedures.

Summary: A rare case of metaplastic matrix-producing breast carcinoma is reported, diagnosed as BI-RADS category 4A via ultrasound, with lymph node metastasis observed in the right axillary region. The patient underwent surgery, chemotherapy, and radiotherapy, but developed distant metastases to the lungs and liver 24 months after surgery and died 3 months later. This case highlights the importance of further examination and biopsy for BI-RADS 4A breast masses before surgery, as well as regular postoperative follow-up.

Summary: This study compares the survival outcomes of metaplastic breast cancer (MPBC) with triple-negative breast cancer (TNBC) and identifies prognostic factors for both types of cancer. The study finds that MPBC is associated with worse overall survival (OS) and disease-free survival (DFS) compared to TNBC. Additionally, within the MPBC group, the subtype with mesenchymal differentiation has a worse DFS compared to other subtypes.

Summary: Most tumor sequencing currently lacks a matched normal control, and germline testing is usually performed independently. The concordance rates between germline and tumor sequencing results are poorly understood. This study compares tumor and germline sequencing results in patients with breast, ovarian, pancreatic, and prostate cancer, and evaluates a computational modeling algorithm's potential to predict germline status based on tumor-only comprehensive genomic profiling (CGP) results.

Summary: This study investigated the impact of tumor phenotype on surgical decision-making and overall survival in early-stage breast cancer. The study found that HER2+ and triple-negative tumors were less likely to undergo unilateral mastectomy compared to hormone receptor-positive tumors. Unilateral and bilateral mastectomy had worse 5-year overall survival compared to breast-conserving surgery. In conclusion, more extensive surgery did not result in better survival outcomes regardless of tumor phenotype.

Summary: Identifying factors associated with persistent pain after breast cancer surgery may facilitate risk stratification and individualised management. In this multicenter study, a predictive model was developed from a sample of Asian and American women. The independent risk factors for persistent pain after surgery were found to be age, diabetes, pre-operative pain score at sites other than the breast, previous mastitis, and perceived stress scale.

The results showed that among women aged 40-50, chemotherapy was only significantly beneficial for overall survival in patients with pN1 disease and an RS of 31-50.

Summary: This study aimed to investigate modifiable contributors to breast cancer treatment from the perspectives of patients and providers and assess the alignment of their perceptions. Both patients and providers identified suboptimal communication, parking and transportation issues, and competing family-caregiving responsibilities as modifiable barriers to care. However, there were also significant discrepancies between the perceptions of patients and providers.

Summary: This study examined the potential impact of neoadjuvant chemotherapy (NACT) on post-mastectomy radiotherapy (PMRT) recommendations for cT3 breast cancer. The results indicated that the use of PMRT was influenced by the response to NACT and the presence of lymph node involvement, rather than in-breast disease alone.

Summary: This study assessed the impact of surgical clerkship curricula on students' likelihood of pursuing a surgical residency. The results indicated that offering more extracurricular surgical clinical opportunities was associated with more students choosing surgical careers. However, the duration of the clerkship, the number of required and subspecialty services, and advanced clinical electives did not have a significant influence on students' career choices.

Summary: This article systematically reviewed prognostic models developed and/or validated for older patients with breast cancer. These models, mostly based on clinico-pathological factors, were used to predict response to treatment, recurrence, and mortality. While external validation was rare, most of the models presented in this review represent promising tools for clinical application.
