Asymmetrical flow field-flow fractionation to probe the dynamic association equilibria of β-D-galactosidase

Summary: Protein dynamics are crucial in enzyme activity, and AF4 was employed in this study to investigate the dynamic association equilibria of the enzyme beta-D-galactosidase. The research found that specific conditions and magnesium ions can significantly influence protein conformation and oligomerization states, shedding light on the role of divalent metal ions in protein oligomerization and the most active form of the enzyme.

Probing Polyester Branching by Hybrid Trapped Ion-Mobility Spectrometry-Tandem Mass Spectrometry

Summary: The TIMS-QTOFMS technique, combined with ESI, is capable of distinguishing between linear and branched isomeric polyester oligomers. By providing both mobility and m/z data, TIMS-MS allows for the construction of EIMs showing conformational differences among isomers, with branching information obtained in less than 1 minute of analysis time.

Microfluidic ion stripper for removal of trifluoroacetic acid from mobile phases used in HILIC-MS of intact proteins

Summary: A membrane-based microfluidic chip was developed for selective post-column removal of TFA anions from HILIC, improving ionization and reducing TFA adduct formation. Summary: This study identified lactosylation sites of BLG using CE-MS and found that lactosylated BLG significantly reduced antibody binding and degranulation of mast cells. These findings suggest that lactosylation may be a promising approach to decrease the allergenicity of cow's milk allergens.

Summary: Progress has been made in Huntington's disease research, but effective treatments before disabling symptoms are still lacking. The European Huntington Association surveyed individuals at risk and with premanifest HD to determine factors affecting their willingness to participate in research. Motivation to participate in research is high, influenced by subjective and objective factors, HD status, and the role of patient organizations in fostering motivation through education and support.

Studying protein structure and function by native separation-mass spectrometry

Summary: Alterations in protein structure can greatly impact biological function. Analytical techniques that maintain the conformation and function of proteins are crucial for studying changes in their higher order structure and establishing structure-function relationships. The coupling of native protein separations with mass spectrometry is a powerful approach for studying these aspects in a reliable, fast, and straightforward manner. Affinity-based separations are highlighted for obtaining both structural and functional information of proteins simultaneously.

While RPLC-MS and HILIC-MS complement each other in resolving the complexity of proteome and metabolites, they also have technical limitations and potential solutions that need to be addressed.

Characterizing Non-covalent Protein Complexes Using Asymmetrical Flow Field-Flow Fractionation On-Line Coupled to Native Mass Spectrometry

Summary: We developed an online analytical platform combining asymmetrical flow field-flow fractionation (AF4) with native mass spectrometry (nMS) for studying labile higher-order structures (HOS) of protein biotherapeutics. The platform also utilizes UV absorbance, multi-angle light scattering (MALS), and differential refractive-index (UV-MALS-dRI) detectors. We studied the stability and dissociation pathways of the tetrameric biotherapeutic enzyme L-asparaginase (ASNase) and found the presence of intact octamers and degradation products using AF4-MALS/nMS. Exposure of ASNase to high pH disrupted the equilibrium between non-covalent species and led to HOS dissociation. The developed platform provides valuable insights for aggregation and stability studies of protein biopharmaceuticals.

Micro-flow size-exclusion chromatography for enhanced native mass spectrometry of proteins and protein complexes

Summary: Size-exclusion chromatography (SEC) with volatile salts and native mass spectrometry (nMS) is valuable for characterizing proteins and aggregates. However, high salt concentrations in SEC-nMS hinder the analysis of labile protein complexes in the gas phase. To overcome this, narrow SEC columns operated at reduced flow rates were used, resulting in improved protein ionization efficiency and the detection of low-abundant impurities and higher-order structures. Lower gas temperatures ensured minimal structural alterations during transfer into the gas phase, and decreased ionization suppression allowed the use of volatile solvents.
