Summary: Research has shown that RSPO2 and RSPO3 can serve as potential therapeutic targets as they modulate Wnt signaling and promote epithelial cell proliferation and survival. High levels of RSPO2 and RSPO3 expression are characteristic of a subset of colorectal and breast cancers, and are associated with an activated Wnt signaling gene expression profile. Knockdown of RSPO2 reduces Wnt signaling and proliferation in breast cancer cells.

Systematic Review and Meta-Analysis: Efficacy of Vancomycin Taper and Pulse Regimens in Clostridioides difficile Infection

Kanika Sehgal, Inuk Zandvakili, Raseen Tariq, Darrell S. Pardi, Sahil Khanna

Summary: The study showed that vancomycin taper-and-pulse regimen is superior to pulse alone or taper alone for resolving CDI. Taper-and-pulse combination had a higher resolution rate compared to taper or pulse alone. Further research is needed to compare vancomycin taper-and-pulse with other treatment options.

Dietary Vitamin E Intake Is Associated With a Reduced Risk of Developing Digestive Diseases and Nonalcoholic Fatty Liver Disease

Eleonora Scorletti, Kate Townsend Creasy, Marijana Vujkovic, Mara Vell, Inuk Zandvakili, Daniel J. Rader, Kai Markus Schneider, Carolin Schneider

Summary: Increased dietary vitamin E intake is associated with reduced rates of several gastrointestinal diseases and overall mortality. Diabetic and overweight individuals with higher vitamin E intake have fewer diagnoses of nonalcoholic fatty liver disease (NAFLD).

Truty, Mitesh Borad, Akhilesh Pandey, Gregory J. Gores, Rory L. Smoot

Summary: A novel LCK inhibitor, NTRC 0652-0, was found to inhibit YAP signaling in CCA cells. It decreased tyrosine phosphorylation, nuclear localization, and co-transcriptional activity of YAP, leading to apoptotic cell death in CCA cell lines. FGFR2 fusion-positive CCA was identified as a susceptible subset, and NTRC 0652-0 showed preclinical efficacy in patient-derived organoid and xenograft models.

Aspirin is associated with a reduced incidence of liver disease in men

Mara Sophie Vell, Arunkumar Krishnan, Kirk Wangensteen, Marina Serper, Katharina Sophie Seeling, Leonida Hehl, Miriam Daphne Rendel, Inuk Zandvakili, Marijana Vujkovic, Eleonora Scorletti, Kate Townsend Creasy, Christian Trautwein, Daniel James Rader, Saleh Alqahtani, Kai Markus Schneider, Carolin Victoria Schneider

Summary: Regular aspirin use is associated with a reduced risk of liver disease in men without an elevated risk of gastrointestinal bleeding or ulcers. The protective effect of aspirin appears to be dependent on the duration of intake, and there may be sex-related differences in its effects.

Summary: This study uses natural language processing (NLP) to analyze data in electronic health records and identifies the underdiagnosis of nonalcoholic fatty liver disease (NAFLD) in clinical practice. The study also finds a method to identify high-risk patients.

A genome-first approach to mortality and metabolic phenotypes in MTARC1 p.Ala165Thr (rs2642438) heterozygotes and homozygotes

Summary: Carriers of MTARC1 p.Ala165Thr exhibit a distinctive lipid phenotype along with reduced risk of non-alcoholic fatty liver disease and liver-related mortality, without an increased risk of cardiovascular death. This highlights MTARC1 as an important modifier of liver disease with potential therapeutic implications.

Physical activity is associated with reduced risk of liver disease in the prospective UK Biobank cohort

Carolin Schneider, Inuk Zandvakili, Christoph A. Thaiss, Kai Markus Schneider

Summary: This study aimed to clarify the association between accelerometer-measured physical activity and chronic liver disease by examining risk of overall and specific liver diseases and their progression in relation to accelerometer-based physical activity in 96,688 participants in the UK Biobank. The results showed a clear, dose-dependent protective association between accelerometer-measured physical activity and liver disease development and progression, suggesting the potential use of wearables for personalized prevention of liver diseases.
