Current methodology of MTT assay in bacteria - A review

The MIT (3-(4,5-dimethylthiazol-2-y1)-2,5-diphenyltetrazolium bromide) tetrazolium assay is a popular tool in estimating the metabolic activity of living cells. The test is based on enzymatic reduction of the lightly colored tetrazolium salt to its formazan of intense purple-blue color, which can be quantified spectrophotometrically. Under properly optimized conditions the obtained absorbance value is directly proportional to the number of living cells. Originally, the MTT assay was devised for use in eukaryotic cells lines and later applied for bacteria and fungi. As the mechanism of MIT reduction was studied in detail mostly considering eukaryotic cells, the lack of information resulted in generating a vast variety of MTT based protocols for bacterial enzymatic activity evaluation. In the presented article the main aspects of the MIT assay applicability in bacterial research were summarized, with special emphasis on sources of inaccuracies and misinterpretation of the test results.

Covalent Inhibition of Bacterial Urease by Bifunctional Catechol-Based Phosphonates and Phosphinates

Summary: A new class of bifunctional inhibitors of bacterial ureases was developed in this study, showing complex structure-dependent kinetics of inhibition. One compound in particular exhibited remarkably reactive inhibition against a specific urease enzyme. The compounds also demonstrated potential in whole-cell inhibition assays, with some showing slow-binding and reversible kinetics.

Phosphinic acid-based enzyme inhibitors

Summary: The phosphinic acid functionality is a valuable scaffold for constructing biologically active compounds, particularly enzyme inhibitors. This article presents two recent achievements in using phosphinic acids as ligands for hydrolases, including the synthesis of a dipeptide analog inhibitor of metalloaminopeptidases and a catechol-based inhibitor of urease, as well as discussing the activity and mode of binding of phosphinic compounds to enzymes.

Dehydrated at Different Conditions and Powdered Leek as a Concentrate of Biologically Active Substances: Antioxidant Activity and Phenolic Compound Profile

Summary: This study found that the green shaft of leek contains more nutrients and higher antioxidant capacity compared to the white shaft, with the dominant phenolic acids being ferulic acid and protocatechuic acids. Additionally, higher drying temperature led to the degradation of phenolic compounds and reduced antioxidant properties of leek shafts.

Optimized Ebselen-Based Inhibitors of Bacterial Ureases with Nontypical Mode of Action

Summary: Screening of 25 analogs of Ebselen led to the discovery of highly potent inhibitors of Sporosarcina pasteurii urease, with a Ki value in a low picomolar range. The exceptional activity was attributed to the presence of a dihalogenated phenyl ring, which increased pi-pi and pi-cation interactions. Biological studies showed good potency for monosubstituted N-phenyl derivatives in inhibiting ureolysis in Proteus mirabilis cells.

Naringenin and Its Derivatives-Health-Promoting Phytobiotic against Resistant Bacteria and Fungi in Humans

Summary: This review discusses the antimicrobial activity of naringenin and its derivatives as a phytobiotic against antibiotic-resistant bacteria and fungi. With the increase in antibiotic resistance, it has become a global health problem. The potential molecular mechanisms of action of naringenin are also described. The review emphasizes the importance of using plant-derived substances to prevent and treat infections in the era of drug resistance.
