Neuropharmacological Potential of Gastrodia elata Blume and Its Components

Research has been conducted in various fields in an attempt to develop new therapeutic agents for incurable neurodegenerative diseases. Gastrodia elata Blume (GE), a traditional herbal medicine, has been used in neurological disorders as an anticonvulsant, analgesic, and sedative medication. Several neurodegenerative models are characterized by oxidative stress and inflammation in the brain, which lead to cell death via multiple extracellular and intracellular signaling pathways. The blockade of certain signaling cascades may represent a compensatory therapy for injured brain tissue. Antioxidative and anti-inflammatory compounds isolated from natural resources have been investigated, as have various synthetic chemicals. Specifically, GE rhizome extract and its components have been shown to protect neuronal cells and recover brain function in various preclinical brain injury models by inhibiting oxidative stress and inflammatory responses. The present review discusses the neuroprotective potential of GE and its components and the related mechanisms; we also provide possible preventive and therapeutic strategies for neurodegenerative disorders using herbal resources.

Hepatoprotective effects of norgalanthamine on carbon tetrachloride induced-hepatotoxicity in mice

Summary: This study found that Norgalanthamine has anti-inflammatory and anti-oxidative properties, which can mitigate the liver injury induced by carbon tetrachloride in mice. Exposure to China dust exacerbates testicular toxicity induced by cyclophosphamide in mice

Unique insertion/deletion polymorphisms within histidine-rich region of histidine-rich glycoprotein in Thoroughbred horses

Summary: In this study, DNA samples were extracted from 1700 Thoroughbred horses and analyzed for gene polymorphisms in the HRG gene. Two types of insertion/deletion polymorphisms (D1 and D2) were identified in the histidine-rich region of the HRG gene, with deletion lengths of 45 bp and 90 bp. These genetic polymorphisms in the HRG gene may be associated with athletic ability, productivity, and susceptibility to infectious diseases in Thoroughbred horses.

Peroxiredoxin 5 regulates osteogenic differentiation through interaction with hnRNPK during bone regeneration

Summary: Peroxiredoxin 5 (Prdx5) plays a significant role in regulating bone resorption and bone formation, and its deficiency leads to osteoporosis. Through immunoprecipitation and LC-MS/MS analysis, hnRNPK was identified as an interacting partner of Prdx5, and Prdx5 negatively regulates the expression of osteocalcin mediated by hnRNPK. Additionally, the loss of Prdx5 enhances the expression of osteoclast-related genes during the differentiation of bone marrow-derived macrophages into multinucleated osteoclasts. These findings suggest that Prdx5 plays an important role in maintaining bone homeostasis by regulating bone cell differentiation.

Effect of abdominal irradiation in mice model of inflammatory bowel disease

Summary: Inflammatory bowel diseases can be diagnosed through diagnostic imaging, but radiation exposure in the intestine can worsen the progression of these diseases. To study the impact of radiation on bowel diseases, mice were induced with colitis and exposed to abdominal irradiation. The results showed that abdominal irradiation aggravated the effects of colitis, including decreased survival rate, body weight, and colon length, as well as increased inflammation levels and expression of inflammatory genes. This study provides valuable insights for patients with inflammatory bowel diseases.

Toxicity of a 90-day repeated oral dose of a collagen peptide derived from skate (Raja kenojei) skin: a rat model study

Summary: There were no treatment-related adverse effects observed in rats that received repeated oral doses of CPSS over 90 days. The CPSS showed no toxicity in terms of clinical signs, body weight, food consumption, detailed clinical observation, sensory reactivity, functional assessment, urinalysis, ophthalmic examination, gross pathology, hematology, serum biochemistry, hormone analysis, organ weight, and histopathology.

Retinal transcriptome profiling identifies novel candidate genes associated with visual impairment in a mouse model of multiple sclerosis

Summary: This study used RNA-sequencing to analyze the transcriptomes of retinas in mice with EAE and identified differentially expressed genes (DEGs) related to EAE-induced visual impairment. Gene expression analysis showed that the upregulated genes in the retinas of mice with EAE were primarily associated with immune responses and immune-mediated inflammation. In addition, gene set enrichment analysis revealed a negative enrichment of gene sets related to the neuronal system and phototransduction cascade in EAE-affected retinas. This study provides novel molecular evidence for visual impairments in EAE and indicates directions for further research to elucidate the mechanisms of these visual impairments in MS.

Particulate matter exposure and neurodegenerative diseases: A comprehensive update on toxicity and mechanisms

Summary: This article reviews the neurotoxicity of particulate matter (PM) and its impact on neurodegenerative diseases. Epidemiological and animal model studies have provided some mechanistic insights. The review highlights the effects of PM exposure on the central nervous system and identifies research gaps and future directions. The findings presented in this review are important for informing public health policies and the development of new treatments for neurodegenerative diseases.

Castanopsis sieboldii Extract Alleviates Acute Liver Injury by Antagonizing Inflammasome-Mediated Pyroptosis

Summary: This study investigated the anti-inflammatory effects of the 70% ethanol extract of Castanopsis sieboldii (CSL3) leaf. The results showed that CSL3 inhibited NO release and iNOS expression in LPS-stimulated cells, and antagonized NF-κB and AP-1 activation through MAPK inhibition. CSL3 also decreased NLRP3 inflammasome activation and increased IL-1β expression. In addition, CSL3 protected against acute liver injury by inhibiting inflammasome formation and pyroptosis.
